        A dynamic human reliability assessment approach for manned submersibles using PMV-CREAM

        Safety is always acritical focus of exploration of ocean resources, and it is well recognized that human factor is one of the major causes of accidents and breakdowns. Our research developed a dynamic human reliability assessment approach, Predicted Mean Vote-Cognitive Reliability and Error Analysis Method (PMV-CREAM), that is applicable to monitoring the cognitive reliability of oceanauts during deep-sea missions. Taking into account the difficult and variable operating environment of manned submersibles, this paper analyzed the cognitive actions of oceanauts during the various procedures required by deep-sea missions, and calculated the PMV index using human factors and dynamic environmental data. The Cognitive Failure Probabilities (CFP) were calculated using the extended CREAM approach. Finally, the CFP were corrected using the PMV index. This PMV-CREAM hybrid model can be utilized to avoid human error in deep-sea research, thereby preventing injury and loss of life during undersea work. This paper verified the method with "Jiaolong" manned submersible 7,000 m dive test. The"Jiaolong" oceanauts CR(Corrected CFP) is dynamic from 3.0615E-3 to 4.2948E-3, the CR caused by the environment is 1.2333E-3. The result shown the PMV-CREAM method could describe the dynamic human reliability of manned submersible caused by thermal environment.

        Characteristic analysis on train-induced vibration responses of rigid-frame RC viaducts

        A three-dimensional (3D) numerical analysis for the train-bridge interaction (TBI) system is actively developed in this study in order to investigate the vibration characteristics of rigid-frame reinforced concrete (RC) viaducts in both vertical and lateral directions respectively induced by running high-speed trains. An analytical model of the TBI system is established, in which the high-speed train is described by multi-DOFs vibration system and the rigid-frame RC viaduct is modeled with 3D beam elements. The simulated track irregularities are taken as system excitations. The numerical analytical algorithm is established based on the coupled vibration equations of the TBI system and verified through the detailed comparative study between the computation and testing. The vibration responses of the viaducts such as accelerations, displacements, reaction forces of pier bottoms as well as their amplitudes with train speeds are calculated in detail for both vertical and lateral directions, respectively. The frequency characteristics are further clarified through Fourier spectral analysis and 1/3 octave band spectral analysis. This study is intended to provide not only a simulation approach and evaluation tool for the train-induced vibrations upon the rigid-frame RC viaducts, but also instructive information on the vibration mitigation of the high-speed railway.

        Modified exosomal SIRPα variants alleviate white matter injury after intracerebral hemorrhage via microglia/macrophages

        Background: Despite limited efficiency, modulation of microglia/macrophages has shown to attenuate neuroinflammation after intracerebral hemorrhage (ICH). In this context, we evaluated the efficacy of modified exosomal signal regulatory protein α (SIRPα) variants (SIRPα-v Exos) in microglia/macrophages and neuroinflammation-associated white matter injury after ICH. Methods: SIRPα-v Exos were engineered to block CD47-SIRPα interactions. After obtaining SIRPα-v Exos from lentivirus-infected mesenchymal stem cells, C57BL/6 mice suffering from ICH underwent consecutive intravenous injections of SIRPα-v Exos (6 mg/kg) for 14 days. Afterwards, the volume of hematoma and neurological dysfunctions were assessed in mice continuously until 35 days after ICH. In addition, demyelination, electrophysiology and neuroinflammation were evaluated. Furthermore, the mechanisms of microglial regulation by SIRPα-v Exos were investigated in vitro under coculture conditions. Results: The results demonstrated that the clearance of hematoma in mice suffering from ICH was accelerated after SIRPα-v Exo treatment. SIRPα-v Exos improved long-term neurological dysfunction by ameliorating white matter injury. In addition, SIRPα-v Exos recruited regulatory T cells (Tregs) to promote M2 polarization of microglia/macrophages in the peri-hematoma tissue. In vitro experiments further showed that SIRPα-v Exos regulated primary microglia in a direct and indirect manner in synergy with Tregs. Conclusion: Our studies revealed that SIRPα-v Exos could accelerate the clearance of hematoma and ameliorate secondary white matter injury after ICH through regulation of microglia/macrophages. SIRPα-v Exos may become a promising treatment for ICH in clinical practice.

        Influential Parameter Analysis on Vibration Responses of Rigid-frame Viaducts Induced by Running High-speed Trains

        This study is intended to clarify the parametric influence for the vibration responses of rigid-frame viaducts in both vertical and lateral directions caused by running high-speed trains. A developed 3D numerical analysis approach considering trainbridge interaction is applied to investigate the vibration characteristics of rigid-frame viaducts. The analytical model is composed of the high-speed train model with multi-DOFs vibration system for each car and the rigid-frame viaduct model with three bridge blocks. They are linked by an assumed wheel-rail relation through the rail model considering the simulated track irregularities. By using the analytical model, the parametric study is carried out. The impact factors including train speed, train type, track irregularity, rail type and damping are investigated and identified as significant variables. The analytical results are significant and give some instructive information regarding the impact factors of the train-bridge system which could be much helpful for the mitigation of vibration and the maintenance in the high-speed railway.

        Chinese Society of Allergy Guidelines for Diagnosis and Treatment of Allergic Rhinitis

        Allergic rhinitis (AR) is a global health problem that causes major illnesses and disabilities worldwide. Epidemiologic studies have demonstrated that the prevalence of AR has increased progressively over the last few decades in more developed countries and currently affects up to 40% of the population worldwide. Likewise, a rising trend of AR has also been observed over the last 2-3 decades in developing countries including China, with the prevalence of AR varying widely in these countries. A survey of self-reported AR over a 6-year period in the general Chinese adult population reported that the standardized prevalence of adult AR increased from 11.1% in 2005 to 17.6% in 2011. An increasing number of original articles and imporclinical trials on the epidemiology, pathophysiologic mechanisms, diagnosis, management and comorbidities of AR in Chinese subjects have been published in international peer-reviewed journals over the past 2 decades, and substantially added to our understanding of this disease as a global problem. Although guidelines for the diagnosis and treatment of AR in Chinese subjects have also been published, they have not been translated into English and therefore not generally accessible for reference to non-Chinese speaking international medical communities. Moreover, methods for the diagnosis and treatment of AR in China have not been standardized entirely and some patients are still treated according to regional preferences. Thus, the present guidelines have been developed by the Chinese Society of Allergy to be accessible to both national and international medical communities involved in the management of AR patients. These guidelines have been prepared in line with existing international guidelines to provide evidence-based recommendations for the diagnosis and management of AR in China.

        Developmental Hypothyroidism Influences the Development of the Entorhinal-Dentate Gyrus Pathway of Rat Offspring

        Background: Developmental hypothyroidism impairs learning and memory in offspring, which depend on extensive neuronal circuits in the entorhinal cortex, together with the hippocampus and neocortex. The entorhinal-dentate gyrus pathway is the main entrance of memory circuits. We investigated whether developmental hypothyroidism impaired the morphological development of theentorhinal-dentate gyrus pathway. Methods: We examined the structure and function of the entorhinal-dentate gyrus pathway in response to developmental hypothyroidism induced using 2-mercapto-1-methylimidazole. Results: 1,1´-Dioctadecyl-3,3,3´,3´-tetramethylindocarbocyanine perchlorate tract tracing indicated that entorhinal axons showeddelayed growth in reaching the outer molecular layer of the dentate gyrus at postnatal days 2 and 4 in hypothyroid conditions. Theproportion of fibers in the outer molecular layer was significantly smaller in the hypothyroid group than in the euthyroid group atpostnatal day 4. At postnatal day 10, the pathway showed a layer-specific distribution in the outer molecular layer, similar to the euthyroid group. However, the projected area of entorhinal axons was smaller in the hypothyroid group than in the euthyroid group. Anelectrophysiological examination showed that hypothyroidism impaired the long-term potentiation of the perforant and the cornuammonis 3–cornu ammonis 1 pathways. Many repulsive axon guidance molecules were involved in the formation of the entorhinaldentate gyrus pathway. The hypothyroid group had higher levels of erythropoietin-producing hepatocyte ligand A3 and semaphorin3A than the euthyroid group. Conclusion: We demonstrated that developmental hypothyroidism might influence the development of the entorhinal-dentate gyruspathway, contributing to impaired long-term potentiation. These findings improve our understanding of neural mechanisms formemory function.

        The Correlation between Thyrotropin and Dyslipidemia in a Population-based Study

        This study investigated the relationship between serum thyrotrophin levels and dyslipidemia in subclinical hypothyroid and euthyroid subjects. A total of 110 subjects with subclinical hypothyroidism and 1,240 euthyroid subjects enrolled in this study. Patients with subclinical hypothyroidism had significantly lower high density lipoprotein cholesterol (HDL-C) levels than those who were euthyroid. The lipid profiles were each categorized and mean thyrotrophin levels were higher in subjects in the dyslipidemia subclasses than subjects in the normal subclasses. Thyrotrophin was positively associated with serum triglyceride and negatively associated with serum HDL-C in women. Thyrotrophin was also positively associated with total cholesterol (TC) in the overweight population along with TC and LDL-C in overweight women. In the euthyroid population, thyrotrophin was positively associated with TC in the overweight population. In conclusion, serum thyrotrophin was correlated with dyslipidemia in subclinical hypothyroid and euthyroid subjects; the correlation was independent of insulin sensitivity.
