Macrophage and Adipocyte Mitochondrial Dysfunction in Obesity-Induced Metabolic Diseases

        Wang Liwen,Hu Jie,Zhou Haiyan 대한남성과학회 2021 The World Journal of Men's Health Vol.39 No.4

Obesity is one of major health burdens of modern society as it contributes to the growing prevalence of its related comorbidities, such as diabetes, cardiovascular diseases, and some cancers. A series of innate immune cells, especially macrophages, and adipocytes have been implicated in the pathogenesis of obesity. Mitochondrial dysfunction, which is induced by obesity, are critical mediators in initiating inflammation in macrophages and adipocytes, and subsequent systemic insulin resistance. In this review, we discuss new findings on how obesity impairs mitochondrial function in macrophages and adipocytes and how this dysfunction contributes to obesity and its comorbidities. We also summarize drugs that treat metabolic diseases by targeting mitochondrial dysfunction.

Predictors of Difficult Laryngeal Exposure in Suspension Laryngoscopy: A Systematic Review and Meta-Analysis

        Mengshu Wang,Guo Li,Yong Liu,Yuanzheng Qiu,Huihong Chen,Wang Liwen,Donghai Huang,Xin Zhang 대한이비인후과학회 2024 Clinical and Experimental Otorhinolaryngology Vol.17 No.2

Objectives. Considerable research has been focused on independent predictors of difficult laryngeal exposure (DLE) duringsuspension laryngoscopy. However, previous studies have yielded inconsistent results and conclusions. Consequently,we performed a meta-analysis of the existing literature with the aim of identifying significant parameters for a stan-dardized preoperative DLE prediction system. Methods. We systematically retrieved articles from the PubMed, Embase, Web of Science, China National Knowledge Infra-structure, and Wanfang databases up to October 2022. Data from eligible studies were extracted and analyzed usingthe R programming language. The effect measures included odds ratios (ORs) with 95% confidence intervals (CIs) fordichotomous variables and mean differences (MDs) with 95% CIs for continuous variables. Results. The search yielded 1,574 studies, of which 18 (involving a total of 2,263 patients) were included. Pooled analysisdemonstrated that patients with DLE during microsurgery tended to be male (OR, 1.73; 95% CI, 1.16–2.57); wereolder (MD, 5.47 years, 95% CI, 2.44–8.51 years); had a higher body mass index (BMI; MD, 1.19 kg/m2; 95% CI,0.33–2.05 kg/m2); had a greater neck circumference (MD, 2.50 cm; 95% CI, 1.56–3.44 cm); exhibited limited mouthopening (MD, −0.52 cm; 95% CI, −0.88 to −0.15 cm); had limited neck flexibility (MD, −10.05 cm; 95% CI, −14.10to −6.00 cm); displayed various other anatomical characteristics; and had a high modified Mallampati index (MMI)or test score (OR, 3.37; 95% CI, 2.07–5.48). Conclusion. We conducted a comprehensive and systematic analysis of the factors relevant to DLE. Ultimately, we identi-fied sex, age, BMI, neck circumference, MMI, inter-incisor gap, hyomental distance, thyromental distance, sternomen-tal distance, and flexion-extension angle as factors highly correlated with DLE.

Prognostic Threshold of Neuroendocrine Differentiation in Gastric Carcinoma: a Clinicopathological Study of 945 Cases

        Zou, Yi,Chen, Linying,Wang, Xingfu,Chen, Yupeng,Hu, Liwen,Zeng, Saifan,Wang, Pengcheng,Li, Guoping,Huang, Ming,Wang, Liting,He, Shi,Li, Sanyan,Jian, Lihui,Zhang, Sheng The Korean Gastric Cancer Association 2019 Journal of gastric cancer Vol.19 No.1

Purpose: The significance of neuroendocrine differentiation (NED) in gastric carcinoma (GC) is controversial, leading to ambiguous concepts in traditional classifications. This study aimed to determine the prognostic threshold of meaningful NED in GC and clarify its unclear features in existing classifications. Materials and Methods: Immunohistochemical staining for synaptophysin, chromogranin A, and neural cell adhesion molecule was performed for 945 GC specimens. Survival analysis was performed using the log-rank test and univariate/multivariate models with percentages of NED ($P_{NED}$) and demographic and clinicopathological parameters. Results: In total, 275 (29.1%) cases were immunoreactive to at least 1 neuroendocrine (NE) marker. GC-NED was more common in the upper third of the stomach. $P_{NED}$, and Borrmann's classification and tumor, lymph node, metastasis stages were independent prognostic factors. The cutoff $P_{NED}$ was 10%, beyond which patients had significantly worse outcomes, although the risk did not increase with higher $P_{NED}$. Tumors with ${\geq}10%$ NED tended to manifest as Borrmann type III lesion with mixed/diffuse morphology and poorer histological differentiation; the NE components in this population mainly grew in insulae/nests, which differed from the predominant growth pattern (glandular/acinar) in GC with <10% NED. Conclusions: GC with ${\geq}10%$ NED should be classified as a distinct subtype because of its worse prognosis, and more attention should be paid to the necessity of additional therapeutics for NE components.

Identification and Functional Analysis of RelA/SpoT Homolog (RSH) Genes in Deinococcus radioduranss

( Jinhui Wang ),( Ye Tian ),( Zhengfu Zhou ),( Liwen Zhang ),( Wei Zhang ),( Min Lin ),( Ming Chen ) 한국미생물 · 생명공학회 2016 Journal of microbiology and biotechnology Vol.26 No.12

To identify the global effects of (p)ppGpp in the gram-positive bacterium Deinococcus radiodurans, which exhibits remarkable resistance to radiation and other stresses, RelA/SpoT homolog (RSHs) mutants were constructed by direct deletion mutagenesis. The results showed that RelA has both synthesis and hydrolysis domains of (p)ppGpp, whereas RelQ only synthesizes (p)ppGpp in D. radiodurans. The growth assay for mutants and complementation analysis revealed that deletion of relA and relQ sensitized the cells to H2O2, heat shock, and amino acid limitation. Comparative proteomic analysis revealed that the bifunctional RelA is involved in DNA repair, molecular chaperone functions, transcription, the tricarboxylic acid cycle, and metabolism, suggesting that relA maintains the cellular (p)ppGpp levels and plays a crucial role in oxidative resistance in D. radiodurans. The D. radiodurans relA and relQ genes are responsible for (p)ppGpp synthesis/hydrolysis and (p)ppGpp hydrolysis, respectively. (p)ppGpp integrates a general stress response with a targeted re-programming of gene regulation to allow bacteria to respond appropriately towards heat shock, oxidative stress, and starvation. This is the first identification of RelA and RelQ involvement in response to oxidative, heat shock, and starvation stresses in D. radiodurans, which further elucidates the remarkable resistance of this bacterium to stresses.

      • Structural characterization of the pulmonary innate immune protein SPLUNC1 and identification of lipid ligands

        Ning, Fangkun,Wang, Chao,Berry, Karin Zemski,Kandasamy, Pitchaimani,Liu, Haolin,Murphy, Robert C.,Voelker, Dennis R.,Nho, Chu Won,Pan, Choel-Ho,Dai, Shaodong,Niu, Liwen,Chu, Hong-Wei,Zhang, Gongyi The Federation of American Societies for Experimen 2014 The FASEB Journal Vol.28 No.12

<P>The short palate, lung and nasal epithelial clone 1 (SPLUNC1) protein is a member of the palate, lung, and nasal epithelium clone (PLUNC) family, also known as bactericidal/permeability-increasing (BPI) fold-containing protein, family A, member 1 (BPIFA1). SPLUNC1 is an abundant protein in human airways, but its function remains poorly understood. The lipid ligands of SPLUNC1 as well as other PLUNC family members are largely unknown, although some reports provide evidence that lipopolysaccharide (LPS) could be a lipid ligand. Unlike previous hypotheses, we found significant structural differences between SPLUNC1 and BPI. Recombinant SPLUNC1 produced in HEK 293 cells harbored several molecular species of sphingomyelin and phosphatidylcholine as its ligands. Significantly, <I>in vitro</I> lipid-binding studies failed to demonstrate interactions between SPLUNC1 and LPS, lipoteichoic acid, or polymyxin B. Instead, one of the major and most important pulmonary surfactant phospholipids, dipalmitoylphosphatidylcholine (DPPC), bound to SPLUNC1 with high affinity and specificity. We found that SPLUNC1 could be the first protein receptor for DPPC. These discoveries provide insight into the specific determinants governing the interaction between SPLUNC1 and lipids and also shed light on novel functions that SPLUNC1 and other PLUNC family members perform in host defense.—Ning, F., Wang, C., Berry, K. Simulation and experiment study for aero-engine turbine blade grinding descaling with single grain

Xiangrui Zeng,Liwen Wang,Xin Lu 대한기계학회 2022 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.36 No.3

In order to explore the mechanism of aero-engine turbine blade grinding descaling, simulation and experiment were carried out on PW4084 first-stage high-pressure turbine rotor blade grinding descaling with single diamond grain. The essential parameters of grinding descaling simulation model were determined by experiment and theoretical analysis. Based on the JH-2 brittle material constitutive model, the grinding descaling simulation with single grain was conducted by FEM. The grinding crack micromorphology and the influence of different grinding parameters on grinding forces were analyzed, then the correctness of simulation results were verified by experiment. The results show that the grinding cracks are direct and brightness, little plastic ridges, with the larger size of broken edge and small partially broken. As the grinding speed increases, the normal force and tangential force reduce. As the grinding depth and grain negative rake increases, the normal force and tangential force increase.
