        In this paper, We propose the method of initial curve creation in levelset evolution for the edge of objects or the shape extractions. The shape of traditional initial curve is including edge. But we improve the speed of processing to draw many initial curve using special parameter which was solved by binary processing and sequence labeling. In particular, it is necessary to efficiently reduce noise, we use anisotropic diffusion filter. This filter is to reduce blurring that is the fault of traditional filter processing and to preserver an accurate edge part. The levelset method using inward and outward evolution extracts the edge of object drawn initial curve, it performs to calculate shrinking and expanding at the same time.

        The Korean hemorrhagic fever (KHF) is an acute febrile disease with characteristic of fever, bleeding tendency, and renal failure. There are many complications of Korean hemorrhagic fever such as infection, anemia, internal bleeding, hypopituitarism, respiratory, and neurologic complication. A few cases were reported on acute pancreatitis with hemorrhagic fever abroad, but there was no case about Korean hemorrhagic fever with acute pancreatitis in this country. We experienced a case of Korean hemorrhagic fever associated with suspected acute pancreatits. With review of articles, we report a case of 51 year-old woman with KHF, where acute pancreatitis developed during management, (Korean J Infect Dis 33:376∼379, 2001)

      • 저널베어링의 마찰성능 실험

        대형저속 디젤엔진에 탑재되는 저널베어링의 모델시험장치를 설계 및 제작하여 작용하중과 회전속도를 변화시켜가며 저널베어링의 마찰성능을 파악하였다. 모델베어링 직경은 60∼120 mm까지 넓은 범위에서 사용가능하도록 하였고, 측면에는 베어링 온도를 측정하기 위한 열전대를 설치하였으며, 유압잭크에 의해 1 ton의 하중까지 가할 수 있도록 하였다. 360。 회전운동하는 저널부와 최대 1800 rpm의 5 ps 모터 사이에는 토크미터를 설치하여 마찰 토크를 측정할 수 있도록 하였다. 모델베어링의 온도는 계속 상승하여 일정한 온도에 도달하는데, 가해지는 하중이 크고 회전속도가 빠를수록 일정한 온도에 도달하는 시간 간격이 작아짐을 알 수 있었다. 하중이 일정한 경우에 회전속도가 증가하면 마찰토크는 증가하였고, 회전속도가 일정한 경우도 하중이 증가하면 마찰토크는 증가하였다. A model testing system was designed and manufactured to understand the frictional characteristics of the journal bearing installed in a large-sized slow speed diesel engine. Model bearing from 60 mm to 120 mm in diameter can be tested in this system and thermocouples are located at the side of the model bearing to measure the temperature of the bearing. Bearing load up to 1 ton is applied by the oil-pressure jack. A torque meter was installed to measure the friction torque between the 360。 rotating journal and 5 PS motor(1800 rpm maximum). Temperature of the bearing rose continuously and reached a constant value. Time interval to reach a constant temperature was found to be less under a larger bearing load or a higher velocity condition. The friction torque increased as the bearing velocity increased under a constant loading condition and as the bearing load increased under a constant velocity condition.

        3.5% NaCl 水溶液中의 自由腐蝕條件下에서 SS41鋼과 複合組織鋼에 대한 回轉굽힘 腐蝕疲勞試驗을 행하여, 溫度影響이 腐蝕疲勞强度 및 腐蝕疲勞크랙 進展速度에 미치는 影響을 考察하였다. 溫度가 增加할수록, 腐蝕疲勞壽命은 減少하였으며 腐蝕疲勞크랙 進展速度는 加速化되었다. 303°K와 318°K의 溫度領域에서 S-N曲線과 腐蝕疲勞크랙 進展速度가 거의 一致하는 것은 陽極溶解의 制限때문이다.

      • Development of genome engineering technologies in cattle: from random to specific

        <P>The production of transgenic farm animals (e.g., cattle) via genome engineering for the gain or loss of gene functions is an important undertaking. In the initial stages of genome engineering, DNA micro-injection into one-cell stage embryos (zygotes) followed by embryo transfer into a recipient was performed because of the ease of the procedure. However, as this approach resulted in severe mosaicism and has a low efficiency, it is not typically employed in the cattle as priority, unlike in mice. To overcome the above issue with micro-injection in cattle, somatic cell nuclear transfer (SCNT) was introduced and successfully used to produce cloned livestock. The application of SCNT for the production of transgenic livestock represents a significant advancement, but its development speed is relatively slow because of abnormal reprogramming and low gene targeting efficiency. Recent genome editing technologies (e.g., ZFN, TALEN, and CRISPR-Cas9) have been rapidly adapted for applications in cattle and great results have been achieved in several fields such as disease models and bioreactors. In the future, genome engineering technologies will accelerate our understanding of genetic traits in bovine and will be readily adapted for bio-medical applications in cattle.</P>

      • Whole genome sequencing of a germline transmitted calf

        Recently, we published a microinjection method for generating transgenic cattle using the DNA transposon system and their analysis by next-generation sequencing (Yum et al. Sci Rep. 2016 Jun 21;6:27185). In that study, we generated transgenic cattle using two different types of DNA transposon system, sleeping beauty (SB) and piggybac (PB), carrying Yellow fluorescent protein with SB (SB-YFP, female) and green fluorescent protein with PB (PB-GFP, male) under the control of the ubiquitous CAG promoter, respectively. The female and male founder cattle have been grown up to date (the female age: 40 months old, the male age: 33 months old) without any health issues. In genomic instability and blood analysis, there was no significant differences between wild type and founder cattle. In the present study, we confirmed germ-line transmission of the transposon-mediated transgene integrations and ubiquitous and persistent expression of transgene in second generation of offspring (F1). The F1 was born without any assistance and expressed GFP in the eyes without UV light. The ubiquitous expression of GFP was detected in skin fibroblast from the ear tissue and confirmed by genomic DNA PCR, which suggest that the transgene from the PB-GFP was successfully transmitted. Unfortunately, no transgene from SB-YFP were identified. To confirm the transgene integration site, the genomic DNA from blood was extracted and performed next-generation sequencing (NGS). The GFP gene was integrated in chromosome 4 (two copies), and 6. As results, a total of two copies of paternal transgene transmitted into the F1. All the integrated position was not related with coding region and there was no significant difference in genomic variants between transgenic and non-transgenic cattle. To our knowledge, this is the first report of germ-line transmission through non-viral transgenic founder cattle. Those transgenic cattle will be valuable resource to many fields of biomedical research and agricultural science.

      • Application of GFP locus for gene Editing in Transgenic Cattle using CRISPR/Cas9 System

        CRISPR/Cas9-induced knock-out/-in can be occurred at specific locus in the genome by non-homologous end joining (NHEJ) or homology directed repair (HDR). Here, we demonstrate the targeted insertion into the specific loci of embryo fertilized by semen from transgenic cattle via CRISPR/Cas9 system. Recently, we published on the efficient generation of transgenic cattle using the DNA transposon system (Yum et al. Sci Rep. 2016 Jun 21;6:27185). In the study, eight transgenic cattle were born following transposon-mediated gene delivery system (Sleeping Beauty and Piggybac transposon system) via microinjection. In the analysis of their genome stability using next-generation sequencing, there was no significant difference in the number of genetic variants between transgenic and non-transgenic cattle. All the transgenic cattle have grown up to date (the oldest age: 33 months old, the youngest age: 15 months old) without any health issue. One of transgenic male cattle expressing GFP reached puberty and semen was collected. Over 200 frozen semen straws were produced and some were used for in vitro fertilization (IVF). On seven days after IVF, expression of GFP was observed at blastocyst stage and was seen in 80% of the embryos. Another application is to edit the GFP locus of the transgenic cattle because long-term and ubiquitous expression of transgene didn’t affect their health. In one cell stage embryos produced using GFP frozen-thawed semen, microinjection of sgRNA for GFP, Cas9, together with donor DNA that included RFP and homology arms to link the double-strand break of sgRNA target site into fertilized eggs resulted in expression of RFP. This indicated that the GFP locus of transgenic cattle shows potential candidates for stable insertion of the functional transgene. Knock-out/-in for editing GFP locus using CRISPR-Cas9 might be a valuable approach for the next generation of transgenic models by microinjection. In conclusion, we demonstrated P-112 that transgenic cattle via transposon system are healthy to date and germ-line competence was confirmed. The GFP locus will be used as the potential target site for future gene engineering via genome-editing technology. Finally, all those animals could be a valuable agricultural and veterinary science resource for studying the effects of gene manipulation on biomedical research and medicine.
