In the present study, we investigated an anti-inflammatory effect of ethyl gallate (EG) isolated from Galla Rhois as evaluated by inhibition of nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression, and a potential role of heme oxygenase-1 (HO-1) in the inhibition of NO production elicited by EG. Treatment of RAW264.7 macrophages with EG significantly inhibited the production of NO and iNOS expression stimulated by lipopolysaccharide (LPS). We also demonstrated that EG treatment increased HO-1 mRNA and protein expression, as assessed by quantitative RT-PCR and Western blot analysis. EG treatment also increased the levels of nuclear factor-erythroid 2-related factor 2, which is critical for transcriptional induction of HO-1. In addition, treatment with SnPP (tin protoporphyrin IX), a selective HO-1 inhibitor, counteracted the inhibitoryeffect of EG on nitrite production, suggesting that HO-1 is, at least in part, implicated in the inhibition of NO production induced by EG treatment. Taken together, these results indicate that EG isolated from Galla Rhois suppresses NO production in LPS-stimulated RAW 264.7 macrophages via HO-1induction.
Analysis of factors affecting crestal bone loss around the implants

Purpose : To determine whether peri-implant crestal bone loss could be affected by systemic disease, primary ISQ value, implantation method (submerged vs. non-submerged), surface treatment, and bone density Materials and methods : Patients who underwent fixture installation from June 24, 2005 to October 23, 2008 at Seoul National University Bundang Hospital were evaluated. A total of 157 patients (male: 52, female: 85) had 346 fixtures installed. Among them, 49 patients had periapical radiographs taken 1 year after prostheses were first set. A total of 97 fixtures were implanted. In particular, 30 fixtures were installed in patients with systemic diseases such as diabetes mellitus, cardiovascular disease, hypertension, and liver disease. The immediate stability of implants was measured with $Osstell^{tm}$. Implant surface treatment was classified into two groups (RBM, Cellnest (Anodized)), and bone density, into four groups (D1~D4). The bone resorption on the mesial and distal areas of fixtures was measured with periapical radiographs using the paralleling technique, and the mean value was calculated. The length determination program in IMPAX (AGFA, Belgium) was used. Results : At least 332 out of 346 (96%) installed GS II implants were successfully osseointegrated 1 year after prostheses were first set. The mean value of the bone resorption of the installed GS II implants was 0.44mm. The minimum value was 0mm, and the maximum value, 2.85mm. There was a statistically significant difference between the implantation methods (submerged, non-submerged) with regard to the amount of alveolar bone loss 1 year after prostheses were first set (p<0.05). Non-submerged implants showed less crestal bone loss. Note, however, that other variables had no correlation with crestal bone loss (p>0.05). Conclusion : There was a statistically significant difference between the 1-stage method and 2-stage method with regard to the amount of alveolar bone loss 1 year after prostheses were first set. Systemic disease, primary ISQ value, surface treatment, and bone density were not associated with alveolar bone loss. Other variables were assumed to have a correlation with alveolar bone loss.

<P><B>Background and Purpose</B></P><P>Progressive external ophthalmoplegia (PEO) with Mendelian inheritance is a heterogeneous group of diseases associated with multiple deletions of mitochondrial DNA (mtDNA), which results from the disturbed replication and maintenance of mtDNA secondary to the mutations of nuclear genes including <I>POLG</I>, <I>SLC25A4</I>, <I>C10ORF2</I>, <I>POLG2</I>, <I>OPA1</I>, and <I>RRM2B</I>. The aim of this study was to identify the genetic defects underlying the pathology and clinical features in two Korean kindreds with autosomal dominant PEO.</P><P><B>Methods</B></P><P>Two pathologically proven PEO patients with a clear autosomal dominant pattern of inheritance were selected. To exclude a large-scale rearrangement, a long-range polymerase chain reaction (PCR) was performed using DNA extracted from biopsied muscle tissue taken from each patient. All coding regions and exon-intron boundaries of <I>POLG</I>, <I>SLC25A4</I>, <I>C10ORF2</I>, and <I>POLG2</I> were amplified by PCR and directly sequenced.</P><P><B>Results</B></P><P>One patient showed multiple deletions of mtDNA on long-range PCR analysis, and two known heterozygous missense mutations in <I>SLC25A4</I> (p.Asp104Gly) and <I>C10ORF2</I> (p.Glu479Lys) were identified in each patient. The p.Asp104Gly mutation in <I>SLC25A4</I> was identified in the patient with an early onset, slowly progressive, pure PEO phenotype, while the p.Glu479Lys mutation in <I>C10ORF2</I> was identified in the other patient, with a late-onset disease and PEO plus phenotype.</P><P><B>Conclusions</B></P><P>Two mutations affecting nuclear genes were identified in Korean patients with autosomal dominant PEO. Further studies are necessary to identify the clear pathogenetic mechanisms and establish genotype-phenotype correlations in autosomal dominant PEO.</P>
Adiponectin as an Anti-fibrotic and Anti-inflammatory Adipokine in the Liver

Hepatic fibrosis is a dynamic process resulting from excessive deposition of extracellular matrix in the liver; uncontrolled progression of fibrosis can eventually lead to liver cirrhosis and/or hepatocellular carcinoma. The fibrogenic process is complex and modulated by a number of both hepatic and extra-hepatic biological factors. Growing evidence indicates that adipokines, a group of cytokines produced by adipose tissue, impart dynamic fibrosis. In particular, two key adipokines, adiponectin and leptin, directly regulate many biological responses closely associated with development and progression of hepatic fibrosis. Leptin acts as a pro-fibrogenic cytokine, while adiponectin possesses anti-fibrogenic and anti-inflamma-tory properties. Adiponectin, acting via its cognate recep-tors, adiponectin receptors 1 and 2, potently suppresses fibrosis and inflammation in liver via multiple mechanism. This review summarize recent findings concerning the role of adiponectin in fibrogenic process in liver and addresses the underlying molecular mechanisms in modu-lation of fibrosis.
Up-regulated claudin 7 expression in intestinal-type gastric carcinoma.

<P>The disruption of tight junction protein expression in cancer might account for invasiveness, loss of cohesion, and lack of differentiation. Our cDNA array data indicated that claudin 7 was up-regulated in gastric carcinoma. We investigated the expression patterns and clinical implications of claudin 7 in gastric cancer. By immunohistochemical staining and Western blot, claudin 7 was significantly more often expressed in intestinal metaplasia, adenoma and cancer than in normal gastric epithelium. Twenty-seven (47.4%) of 57 normal gastric epithelium samples did not express claudin 7, but 50 (86.2%) of 58 intestinal metaplasia, 11 (91.7%) of 12 adenoma tissues, and 129 (82.7%) of 156 cancer samples did. Claudin 7 was more often unexpressed in diffuse type gastric cancer than in intestinal type. Only 13 (11.2%) of 116 intestinal type samples did not express claudin 7, but 14 (41.2%) of 34 diffuse type samples showed no expression. Compared to normal gastric epithelium, intestinal type gastric cancer significantly more often expressed claudin 7, but diffuse type did not. The expression pattern of claudin 7 did not change as cancer progressed. In this study we show that claudin 7 expression changed with the gastric carcinogenic process and that this is implicated in cancer characteristics.</P>

<P>It is important to accurately estimate the flying height (FH) of the ultralow flying slider in hard disk drive (HDD). Especially, during the track-seeking motion, the FH can change by the motion from inner diameter (ID) to outer diameter (OD) and vice versa. The FH variation induced by the track-seeking motion happens over discrete track media (DTM). Therefore, the FH variation during track-seeking is an important issue and it certainly should be considered for DTM. In this research, we performed the track-seeking simulation using quasi-static approximation to reduce the computation time. The effects on groove depth, width, and pitch with the effective skew angle taken into account were investigated with the simulation. As a result, we calculated the absolute change in FH as a function of the effective skew angle and groove depth. Finally, we proposed that the model that considers the effective skew angle over DTM should be adopted in the track-seeking motion-studies.</P>

<P>In this paper, a thermally assisted magnetic recording (TAMR) head gimbal assembly (HGA) with an optical fiber and prism is proposed. The stiffness of a TAMR HGA is increased by an optical fiber and prism. Therefore, in this paper, operating shock analysis of a TAMR HGA considering the thermal effects during writing to record the data is numerically performed. Flying height (FH) analysis considering the thermal deformation by laser source also is investigated. As a result, judging from shock point of view, the shock characteristic of a TAMR HGA is better than that of conventional HGA. However, when considering the thermal effects during writing to record the data, the risk of contact for a TAMR HGA between media and slider is larger than that of conventional HGA. Hence, to solve these problems of a TAMR HGA, we should consider selection for material of media and TAMR slider, and design for head parts of TAMR slider.</P>
