      • Bacillus sp. Ye-1213이 생산하는 chitinase의 동정, 분리정제 및 특성

        국내 해변 연안의 해변의 토양으로부터 chitinase 활성이 우수한 균주를 분리하고 동정하여 Bacillus sp. YE-1213으로 명명하였다. Chitinase를 생산하기 위한 최적 배양조건은 1.0%(w/v, dry base) Colloidal chitin, 1.0%(w/v) Tryptone, 0.05%(w/v) MgSo₄·7H₂O, pH 8.0, 30℃, 배양시간 48시간이었다. 효소는 (NH₄)₂SO₄분별침전, affinity adsorption, Sephadex G-200 gel filtration의 과정을 거쳐 정제한 결과 비활성 81.32units/mg, 정제도 16.7배, 수율은 15.8%이었다. 정제된 효소는 최적 반응조건은 pH 6.0, 40℃ 이었으며, 온도에 대한 효소안전성은 40℃까지는 매우 안정하였고 pH에 대한 안정성은 pH 5.0-9.0에서 안정하였다. 정제된 효소의 Km 값은 1.74mg/ml이었고 PCMB, I₂, ?? ion, ?? ion 에 의해 저해되었으며 ?? ion, ?? ion 및 ?? ion 에 의해 1.21배, 1.12배, 1.18배의 효소활성이 증가하였다. A bacterium with the highest chitinase activity selected from the soil and identified as a Bacillus sp. YE-1213. The optimal condition for the production of chitinase was as follows; 1.0% (w/v, dry base) colloidal chitin, 1.0%(w/v) tryptone, 0.05%(w/v) MgSO₄·7H₂O, pH8.0, 30℃, 48 hrs. The chitinase was purified about 16.7 folds by ammonium sulfate fraction, affinity adsorption and Sephadex G-200 gel filtration. Optimal temperature and pH for the enzyme activity were 40℃ and pH 6.0. The enzyme was stable between pH 5.0-9.0 and up to 40℃. Its activity was activated by ??, ??, and ?? while inhibited by PCMB, I₂, ?? and ??.

      • 영상분할통신을 이용한 화상진료시스템의 설계

        In general cases, the conventional internet connected to a terrestrial network is transmit too large medical images. To overcome this low speed transmitting rate problem of the internet, we have studied about an image division communication system as a fast telemedicine system. The Image division communication system was 5 10 times faster than the conventional terrestrial internet link. Also, we have developed a Web-based telemedicine system that can access every permitted server of hospital via the internet. Studied image division communication corrected problem of other reflex quality decline in erratic transmission of reflex by transmission speed imbalance that is problem of single communication techniques that used in existing reflex transmission. Also, could keep quality state of fixed reflex gouge abnormal transmission speed. Visual Basic and C++, ASP programming techniques were used to make our system and it can access and retrieve medical information and image through only public web browse such as internet explorer without additional specific tools. To increase the transmitting speed of our telemedicine system, JPEG method was used. In conclusion, we were able to develop a fast and public telemedicine system using the proposed image division communication system and Web technology. Image division communication system technology increased the speed of the conventional internet and Web technology extended the scope of use for telemedicine system from intrahospital to public us.

        지체장애인 편의시설의 실태조사 및 현황 분석에 관한 연구 : 대구광역시 종합사회복지관을 중심으로 Focusing of The Welfare Facilities in Daegu

        This study is to search for appearance of facilities based on facility improvement and then analyzes degree of satisfaction of user and level of facilities by comparing. The way of improvement of welfare facilities by searching can be summarized as follow : Facilities for access : For the handicapped, circulation of lamp should be separated from parking area. Parking area for the handicapped should be made safe against general people and also should be made guide equipments for the handicapped parking area. Facilities for shifting : Hand-rails that exist corridors and stairs should be founded with law of facility improvement. Welfare facilities that founded elevators should be slowed down closing time of elevator door for the handicapped, and welfare facilities that founded wheelchair lifts should be enhanced application by educating. Facilities for sanitation. : Avaliable close-stool space and avaliable door width of toilet should be founded by law of improvement facility. Floor tiles of toilet should be finished off non-slipping tiles. In conclusion, criteria of system are most important of all when facilities for the handicapped are founded. And it would be extremely ideal, if facilities can be designed appropriately for the handicapped with this criteria of system.

        This study is to search for appearance of facilities based on facility improvement and then analyzes degree of satisfaction of user and level of facilities by comparing. This study is focused of welfare facilities in Daegu. Conclusions as follow : 1 Satisfaction of the facility is different whether welfare facilities were founded before establishment by law of facility improvement or not. 2 Most welfare facilities is weak an elevator equipment. So the right to access for the handicapped act has been limiting so much. 3 Expansion of parking areas for disabled person are urgent.

        Multimedia information includes image information, sound information, character information and various informations. Difficulty follows to look for image which user wants by existent retrieval method. Therefore, introduce MSE(Mean Square Error) by index method for efficient practical use of video information in this research and propose scene change detection retrieval method of video. This image data for diagonal line direction color value of schedule pixel draw, (By j^(*)i matrix to Matrix A, i is frame number, j stored cost that express diagonal line Image height of frame to Matrix A). So that can grasp whole structure of video, do sampling in still image, Made user can grasp whole structure of video, do sampling in still image. Made user grasp scene change point of total video on one eye. If MSE of each frame exceeds critical value, search the frame by scene change.

        해상 실험에 의한 침하식 해삼 양성 기구의생물학적 성능 평가

        The sea cucumber, Apostichopus japonicus, is becoming an important aquaculture species in China, Japan and Korea. The purpose of this study is to evaluate the biological performance of a tubular type of subsurface cage for sea cucumber grow-out by in-situ tests. The cage structure was constructed of PP (polypropylene) frames and PP tubes (Φ130 mm×24 pieces). A set of tests were conducted at a depth of 15 m near Seosaeng, Ulsan, Korea for 295 days (23 July, 2012 to 13 May, 2013). A total of 155 sea cucumbers were used and fed a mixed diet containing mud, mineral, wheat, fish meal, etc (3% of their body weight). At the end of the experiments, sea cucumber showed a higher survival rate in feed shelter (77.14%) rather than no feed shelter (64.71%). The specific growth rate (SGR) of sea cucumbers in feed condition (0.04% day-1) was higher than that of no feed culture animals (-0.49% day-1) during total experimental periods. The result was able to find a no significance difference in survival rate but a significance difference in SGR under feed and no feed treatments.

      • 실내 수조에서 수온 변화에 따른 해삼(Apostichopus japonicus, Selenka)의 운동 패턴

        A series of experiments on the movement patterns of sea cucumber (Apostichopus japonicus) to the change of water temperature (10, 13, 16, 19 and 21℃) were carried out in the indoor water tank. The experiments consisted of the preliminary test (Case 1) and formal experiments (Case 2). In case 1 test, the moving patterns of sea cucumbers inside model cage set up in water tank was examined with naked eye observation. In addition, the moving speed and distance of total sea cucumbers in water tank were analyzed based on the image of video camera and PVS program in the case 2 test. The initial body wet weights of sea cucumber were 33.1±11.3 g in case 1 and 52.9±15.3 g in case 2 , respectively. The movement patterns of sea cucumbers were observed using video camera for 12 hours a day. The movement pattern of sea cucumber showed the highest activity at 10℃ of water temperature. The maximum moving distance and speed of sea cucumber were about 0.78m and approximately 0.028 cm/s at 10℃, respectively. The minimum moving distance and speed of sea cucumber were about 0.01m at 19℃ and approximately 0.0048cm/s at 21℃, respectively. It was observed that the optimum water temperature for the movement of sea cucumber was about 10℃.

        염료의 색도 제거능력이 우수한 백색부후균 분리 및 특성 연구

        한국에서 자생하는 버섯으로부터 염료의 색도 제거능력이 우수한 균주를 분리하기 위하여 각종 염료를 함유한 평판배지 상에서 성장성과 색도 변화를 관찰한 결과 우수한 균체 성장과 색도 제거능력을 지닌 3가지 균주를 최종 선별하였으며, 선별된 균주의 특성을 조사하여 Trametes sp. CJ-105, Pleurotus sp. CJ-212, Fomes sp. CJ-315로 동정하였다. 이 균주들은 각각 remazol brilliant blue R 및 Acid red 264를 함유한 액체배지에서 배양한 결과, 배양 2∼8일째에 거의 색도가 제거되었고 리그닌 분해효소인 laccase의 활성이 다량 존재하였다. 선별된 균주의 색도 제거능력과 laccase, LiP, MnP의 활성은 상관관계가 있는 것으로 나타났으며, 염료의 색도 제거는 리그닌 PAH 및 PCP의 분해에도 좋은 활성을 가지며 난분해 물질의 분해에도 리그닌 분해효소의 활성이 상관관계가 있음을 확인하였다. 따라서, 선별된 균주가 향후 염색폐수의 색도 제거, 난분해 물질의 분해 또는 매립지 침출수의 처리등에 이용될 수 있을 것으로 기대된다. Several white-rot fungi collected from the mountains of Korea were evaluated for their ability to decolorize azo, polymeric, and reactive dyes. Strains CJ-105, CJ-212 and CJ-315, identified as Trametes sp., Pleurotus sp. and Formes sp., respectively, showed higher potential for decolorization of those dyes in either solid or liquid media. For Trametes sp. CJ-105, 100 ppm of Remazol Brilliant Blue R and 500 ppm of Acid Red 264 were completely decolorized after 2 days under liquid culture. The dominating ligninolytic enzyme existing in the culture broth was laccase (E.C. 1.10.3.2). Also, Pleurotus sp. CJ-212 and Fomes sp. CJ-315 showed similar patterns in decolorization of Remazol Brilliant Blue R and Acid Red 264. The extent of decolorization of the dyes in liquid culture was found to be proportional to the activities of the ligninolytic enzymes produced by each strain. In addition to that Trametes sp. CJ-105 was highly effective in degradation of polycyclic aromatic hydrocarbons and pentachlorophenol by the activity of the ligninolytic enzymes produced. In this study, we found that white-rot fungi, Trametes sp. CJ-105(KFCC 10941), Pleurotus sp. CJ-212(KFCC 10943) and Fomes sp. CJ-315(KFCC 10942), were effective in decolorizing a wide range of structurally different synthetic dyes, as well as some chemical compounds which are known to be hardly degradable.
