FSCB phosphorylation in mouse spermatozoa capacitation

( Shun Li Liu ),( Bing Ni ),( Xiang Wei Wang ),( Wen Qian Huo ),( Jun Zhang ),( Zhi Qiang Tian ),( Ze Min Huang ),( Yi Tian ),( Jun Tang ),( Yan Hua Zheng ),( Feng Shuo Jin ),( Yan Feng Li ) 생화학분자생물학회(구 한국생화학분자생물학회) 2011 BMB Reports Vol.44 No.8

It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation. Similarities and Differences in East Asian Confucian Culture: A Comparative Analysis

Min-hua Huang,Shu-Hsien Chang 숙명여자대학교 아시아여성연구원 2017 OMNES: The Journal of Multicultural Society Vol.7 No.2

In East Asia, Confucianism has long been regarded as the cultural basis for social norms and political order. In the past literature, many scholars have argued that the special nature of Confucian culture is an important factor hindering the modernization of East Asian countries. However, rapid modernization in the past two decades has shown that East Asia is not limited in its modernization by its cultural features. In this paper, the authors explore how the effects of modernization and democratization affect Confucian values in East Asian. Using data from the latest wave of the Asian Barometer Survey data in nine East and Southeast Asian countries, the authors find that modernization and democratization might attenuate Confucianism, but this trend is actually transformative instead of linear, meaning that Confucianism may be transformed into a post-modern culture that offers spiritual well-being and continues to guide the future progress of human civilization.

Digital Revolution or Digital Dominance? Regime Type, Internet Control, and Political Activism in East Asia

Min-Hua Huang,Wen Hong 한국국제정치학회 2017 The Korean Journal of International Studies Vol.15 No.2

Many studies have shown that Internet use may be a driver of real world political participation. However, we still have a lot to learn about the dynamics of this relationship. This article elaborates this relationship by exploring the effect of different types of Internet use (including social media) on participation in real world political activism. We also explain how two contextual factors – regime type (level of authoritarianism) and the level of government control over the Internet (level of Internet control) interact and affect the relationship between Internet use and political activism. Our findings in general corroborate the expected positive and significant relationship between Internet use and political activism. However, this relationship is highly contingent on how the level of authoritarianism and level of Internet control interact, producing two contrasting patterns of relationships between Internet use and political activism, labeled as “digital revolution” and “digital dominance.” We find that one-party authoritarian regimes are closer to the “digital dominance” type, meaning that the interaction between regime type and Internet control weakens the relationship between Internet use and political activism; however, in limited democracies or democracies with excessive Internet control or authoritarianism, citizens’ participation in political activism through online channels is emboldened, producing the “digital revolution” type. In democracies with little Internet control, citizens may have an incentive to participate in political activism through the Internet, but institutional rules that allow rotation of power might prevent either “digital dominance” or “digital revolution” from developing.

How Does Rising Internet Usage Affect Political Participation in East Asia? Explaining Divergent Effects

        ( Min-hua Huang ),( Ching-hsuan Su ),( Ruixia Han ),( Mark Weatherall ) 경남대학교 극동문제연구소 2017 ASIAN PERSPECTIVE Vol.41 No.4

The advance of information and communication technologies (ICTs) has fundamentally changed the way people communicate and interact with each other, and the rise of the Internet profoundly affects political participation. In applying the latest Asian Barometer surveys, we discovered Internet-driven divergent effects on political participation, suggesting rising Internet usage is simultaneously associated with decreasing electoral and increasing activist participation. Further analysis revealed that the divergent effects can be explained by severe frustration with the political system and economic conditions. When considering state polity characteristics, we found that the Internet-driven divergent effects in a democratic context refer to a coincidence of two Internet-related effects that decrease electoral and increase activist participation; in an authoritarian context, they indicate a correlation between greater Internet usage and a preference for activist over electoral participation.

MRAS Genetic Variation is Associated with Atherothrombotic Stroke in the Han Chinese Population

        Hua Liu,Xiao Qin Huang,Min Yang,Xun Ming Ji,Xin Du,Jian Zheng 대한신경과학회 2013 Journal of Clinical Neurology Vol.9 No.4

Background and Purpose Atherothrombotic cerebral infarction [atherothrombotic stroke (ATS)] shares common risk factors and pathophysiological mechanisms with coronary artery disease (CAD), and both diseases appear to have common susceptibility loci. The muscle RAS oncogene homolog gene (MRAS) has been identified as a susceptibility locus for CAD and is implicated in atherosclerosis. The aim of this study was to elucidate whether the single-nucleotide polymorphisms (SNPs) and haplotypes of MRAS are associated with increased risk of ATS in a population of Han Chinese. Methods A case-controlled association study was conducted in which only patients with ATS (identified as a major subtype in the Korean modification of the Trial of Org 10172 in Acute Stroke Treatment classification) were enrolled. Subgroup analyses were carried out to determine whether the effect of the MRAS polymorphism was specific to age and gender among the subjects. Results In total, 194 ATS and 186 control subjects were included in the present study. Two tagging SNPs were identified in MRAS (rs40593 and rs3755751). A multivariate regression analysis revealed a positive association between rs40593 and ATS under dominant and additive models after adjustment for covariates. Subgroup analyses revealed that there were no gender differences with respect to allele or genotype frequencies between the groups. The AG genotype for rs40593(p=0.028), the CT genotype for rs3755751 (p=0.036), and G-allele carriers (AG plus GG) for rs40593 (p=0.015) exhibited a significant protective effect among those aged ≥45 years. For the haplotype analysis, ATS subjects aged ≥45 years had a higher frequency of the ACAC haplotype (76.0%) than the controls (68.1%; p<0.05); that haplotype was associated with an increased risk of ATS. Conclusions The obtained data suggest a positive association between MRAS and ATS among the Han Chinese. Further studies should be performed with larger sample and among different ethnic populations, and gene-gene or gene-environment interactions should be considered.

The cytoplasmic loops of AgrC contribute to the quorum-sensing activity of Staphylococcus aureus

        Huang Qian,Xie Yihui,Yang Ziyu,Cheng Danhong,He Lei,Wang Hua,Liu Qian,Li Min 한국미생물학회 2021 The journal of microbiology Vol.59 No.1

In Staphylococcus aureus, the accessory gene regulator (agr) quorum-sensing system is thought to play an important role in biofilm formation. The histidine kinase AgrC is one of the agr system components and activated by the self-generated auto-inducing peptide (AIP), which is released continuously into the extracellular environment during bacterial growth. The extracellular loops (Extra-loops) of AgrC are crucial for AIP binding. Here, we reported that the cytoplasmic loops (Cyto-loops) of AgrC are also involved in Agr activity. We identified S. aureus ST398 clinical isolates containing a naturally occurring single amino acid substitution (lysine to isoleucine) at position 73 of an AgrC Cyto-loop that exhibited significantly stronger biofilm formation and decreased Agr activity compared to the wild-type strain. A constructed strain containing the K73I point mutation in AgrC Cyto-loop continued to show a growth dependent induction of the agr system, although the growth dependent induction was delayed by about 6 h compared to the wild-type. In addition, a series of strains containing deletion mutants of the AgrC Cyto- and Extra-loops were constructed and revealed that the removal of the two Cyto-loops and Extra-loops 2 and 3 totally abolished the Agr activity and the growth-dependence on the agr system induction. Remarkably, the Extra-loop 1 deletion did not affect the Agr activity. In conclusion, the AgrC Cyto-loops play a crucial role in the S. aureus quorum-sensing activity.

      • Palladium nanoparticles decorated mesoporous carbon spheres as catalyst for reduction of 4-nitrophenol.

Huang, Xin Hua,Moon, Byeong Kyu,Byeon, Seong Jin,Heo, Min Seon,Kim, Ii American Scientific Publishers 2014 Journal of Nanoscience and Nanotechnology Vol.14 No.11

<P>Two kinds of polyaromatics with mesoporous have been synthesized from aromatic hydrocarbons using anhydrous zinc chloride as the Friedel-Crafts catalyst and chloromethyl methyl ether as a cross-linker, after the Pd nanoparticles (PdNPs) decorated on the mesoporous carbon spheres (Pd@CSs) have been prepared by simply mixing the as-prepared polyaromatics (polynaphthalene or polypyrene) with PdCl2, reducing the Pd2+ to Pd0 by using NaBH4, followed by thermal treatment at 600 degrees C for 5 h in nitrogen atmosphere. The synthesized PdNPs have a uniform size distribution with an average size smaller than 15 nm and they can be loaded on the highly mesoporous carbon microspheres. Structural of the resulting Pd@CSs were carried out using FE-SEM, HR-TEM, X-ray differaction, dispersive X-ray spectroscopy. The resulting Pd@CSs have been investigated as a catalyst for the reduction of 4-nitrophenol to 4-aminophenol, showing the Pd@CSs have high catalytic reactivity and recyclability.</P>
