Influenza A virus subtype H7N9 has received great attention since the first human case was reported at the beginning of this year. The viral characteristics have been quickly brought into research process. Here, the effect of temperature and live poultry market has been considered on the basis of analysis of existing data. As expected, both temperature and live poultry market has significant impact on the spread of this epidemic. Finally, the effect of government reactions and medical treatment on response was discussed. It indicated that sufficient and rapid reactions did not always lead to a perfect outcome which proved the influence of temperature and live poultry market potentially. Research highlights: The effect of temperature on H7N9 avian influenza is proved by analyzing the latitude and temperature of affected area. The effect of live poultry markets on H7N9 avian influenza is confirmed by analyzing the relationship between the number of live poultry markets in affected area and incidence of H7N9 human infection. The effect of government actions and medical treatment is studied and it turned out the result is not in accordance with actual efforts.

The serine protease gene from a thermophilic fungus Thermoascus aurantiacus var. levisporus, was cloned, sequenced, and expressed in Pichia pastoris and the recombinant protein was characterized. The full-length cDNA of 2,592 bp contains an ORF of 1,482 bp encoding 494 amino acids. Sequence analysis of the deduced amino acid sequence revealed high homology with subtilisin serine proteases. The putative enzyme contained catalytic domain with active sites formed by three residues of Asp183, His215, and Ser384. The molecular mass of the recombinant enzyme was estimated to be 59.1 kDa after overexpression in P. pastoris. The activity of recombinant protein was 115.58 U/mg. The protease exhibited its maximal activity at 50°C and pH 8.0 and kept thermostable at 60°C, and retained 60% activity after 60 min at 70°C. The protease activity was found to be inhibited by PMSF, but not by DTT or EDTA. The enzyme has broad substrate specificity such as gelatin, casein and pure milk, and exhibiting highest activity towards casein.
Exploring and calibrating local curvature effect of cortical bone for quantitative ultrasound (QUS)
Chen, Jiangang,Su, Zhongqing,Cheng, Li,Ta, De-An Techno-Press 2013 Structural Engineering and Mechanics, An Int'l Jou Vol.48 No.4
Apart from thinning of cortical layers, the local bone curvature, varying along bone periphery, modulates ultrasound waves as well, which is however often underestimated or overlooked in clinical quantitative ultrasound (QUS). A dedicated three-dimensional finite element modelling technique for cortical bones was established, for quantitatively exploring and calibrating the effect of local curvature of cortical bone on ultrasound. Using a correlation-based mode extraction technique, high-velocity group (HVG) and low-velocity group (LVG) wave modes in a human radius were examined. Experimental verification using acrylic cylinders and in vitro testing using a porcine femur were accomplished. Results coherently unravelled the cortical curvature exerts evident influence on bone-guided ultrasound when RoC/${\lambda}$ <1 for HVG mode and RoC/${\lambda}$ <2 for LVG mode (RoC/${\lambda}$: the ratio of local bone curvature radius to wavelength); the sensitivity of LVG mode to bone curvature is higher than HVG mode. It has also been demonstrated the local group velocity of an HVG or LVG mode at a particular skeletal site is equivalent to the velocity when propagating in a uniform cylinder having an outer radius identical to the radius of curvature at that site. This study provides a rule of thumb to compensate for the effect of bone curvature in QUS.

Cinnamate 4-hydroxylase (C4H) is a key enzyme of phenylpropanoid pathway, which synthesizes numerous secondary metabolites to participate in development and adaption. Two C4H isoforms, the 2192-bp BnC4H-1 and 2108-bp BnC4H-2, were cloned from oilseed rape (Brassica napus). They both have two introns and a 1518-bp open reading frame encoding a 505-amino-acid polypeptide. BnC4H-1 is 57.73 kDa with an isoelectric point of 9.11, while 57.75 kDa and 9.13 for BnC4H-2. They share only 80.6% identities on nucleotide level but 96.6% identities and 98.4% positives on protein level. Showing highest homologies to Arabidopsis thaliana C4H, they possess a conserved p450 domain and all P450-featured motifs, and are identical to typical C4Hs at substrate-recognition sites and active site residues. They are most probably associated with endoplasmic reticulum by one or both of the N- and C-terminal transmembrane helices. Phosphorylation may be a necessary post-translational modification. Their secondary structures are dominated by alpha helices and random coils. Most helices locate in the central region, while extended strands mainly distribute before and after this region. Southern blot indicated about 9 or more C4H paralogs in B. napus. In hypocotyl, cotyledon, stem, flower, bud, young- and middle-stage seed, they are co-dominantly expressed. In root and old seed, BnC4H-2 is dominant over BnC4H-1, with a reverse trend in leaf and pericarp. Paralogous C4H numbers in Brassicaceae genomes and possible roles of conserved motifs in 5' UTR and the 2nd intron are discussed.

Matric suction plays a key role in slope stability by conferring an apparent cohesion component to the unsaturated portion of the soil. This paper adopts the total cohesion method to investigate the contribution of apparent cohesion on the stability of silty slopes under hydrostatic conditions. Phase2 and Optum G2 numerical programs, based on strength reduction finite element analysis and finite element limit analysis methods, respectively, are used for numerical analysis. Generally, Phase2 and Optum G2 results are in good agreement with each other. Optum G2 yields slightly higher factor of safety results than Phase2, particularly for steep slopes β ≥ 30°. The results are presented in form of stability charts which are validated with a case from a previous study. Notably, the contribution of apparent cohesion to unsaturated shear strength is most pronounced when varying the water table. An examination of the slope failure mechanism reveals that the toe failure mechanism is the dominant failure mechanism. The depth of the failure surface is most sensitive to changes in the slope angle, cohesion and water table position. The influence of the air-entry value on the depth of the failure surface is contingent upon the location of the water table.

Background: Increasing epidemic of ischemic stroke (IS) makes it urgent to understand the pathogenesis and regulatory mechanism, previous studies have described microRNAs (miRNAs) is part of the brain's response to ischemia. Objective: The aim of this study was to screen potential biomarkers for the prediction and novel treatment of IS. Methods: Differentially expressed miRNAs were screened from three newly diagnosed IS patients and three controls by RNA sequencing technology. Furthermore, target prediction databases were then used to analysis the target genes of different expressed miRNAs, and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database were used to identify the functions and the main biochemical and signal pathways of differentially expressed target genes. Results: Our results revealed that 27 miRNAs were differentially expressed in IS, among which, hsa-miR-659-5p was the most highly increased and was first found to be associated with IS. In addition, KEGG pathway analyses showed that differentially expressed miRNAs were mainly significantly enriched in lysosome pathway, cytokine-cytokine receptor interaction pathway, spliceosome pathway, base excision repair pathway. Conclusions: miRNAs were involved in IS pathogenesis, and hsa-miR-659-5p, hsa-miR-151a-3p and hsa-miR-29c-5p as the three highest |log2FoldChange| regulation in this study, which may be the biomarkers of IS and need further study.
