Molecular Cloning and Bioinformatic Analysis of SPATA4 Gene

Full-length cDNA sequences of four novel SPATA4 genes in chimpanzee, cow, chicken and ascidian were identified by bioinformatic analysis using mouse or human SPATA4 cDNA fragment as electronic probe. All these genes have 6 exons and have similar protein molecular weight and do not localize in sex chromosome. The mouse SPATA4 sequence is identified as significantly changed in cryptorchidism, which shares no significant homology with any known protein in swissprot databases except for the homologous genes in various vertebrates. Our searching results showed that all SPATA4 proteins have a putative conserved domain DUF1042. The percentages of putative SPATA4 protein sequence identity ranging from 30% to 99%. The high similarity was also found in 1 kb promoter regions of human, mouse and rat SPATA4 gene. The similarities of the sequences upstream of SPATA4 promoter also have a high proportion. The results of searching SymAtlas (http://symatlas.gnf.org/SymAtlas/) showed that human SPATA4 has a high expression in testis, especially in testis interstitial, leydig cell, seminiferous tubule and germ cell. Mouse SPATA4 was observed exclusively in adult mouse testis and almost no signal was detected in other tissues. The pI values of the protein are negative, ranging from 9.44 to 10.15. The subcellular location of the protein is usually in the nucleus. And the signal peptide possibilities for SPATA4 are always zero. Using the SNPs data in NCBI, we found 33 SNPs in human SPATA4 gene genomic DNA region, with the distribution of 29 SNPs in the introns. CpG island searching gives the data about CpG island, which shows that the regions of the CpG island have a high similarity with each other, though the length of the CpG island is different from each other.This research is a fundamental work in the fields of the bioinformational analysis, and also put forward a new way for the bioinformatic analysis of other genes.

We present for the first time the analysis and fabrication of a novel Tin-Nickel mixed salt electrolytic coloured processing and the performance of coloured films for Al-12.7Si-0.7Mg alloy. This alloy is a novel alloy containing high silicon aluminum alloy extrusion profile which presents excellent mechanical properties as well as broad market prospects. Nevertheless, this kind of material is urgent in need of surface treatment technology. The orthogonal design and single factor tests were applied to optimize for electrolytic coloured technological conditions. By controlling operation conditions, the uniform electrolytic coloured films with different color were obtained. Analysis of microstructure showed that tin particles had been deposited in the coloured film. The coloured films, about 10 μm thick, were uniform, dense and firmly attached to the substrate. After the coloured samples were maintained at 400ºC for 1 h, or quenched from 300ºC to room temperature, the coloured films did not change, demonstrating excellent thermostability and thermal shock resistance. Acid and alkali corrosion tests and potentiodynamic polarization showed that corrosion resistance of coloured sample was much better than those of untreated samples. After 240 h neutral salt spray test, protection ratings and appearance ratings of coloured films were Grade 9.

Mitochondrial dysfunction plays a major role in the development of intervertebral disc degeneration (IDD). Sirtuin 5 (SIRT5) participates in the maintenance of mitochondrial homeostasis through its desuccinylase activity. However, it is still unclear whether succinylation or SIRT5 is involved in the impairment of mitochondria and development of IDD induced by excessive mechanical stress. Our 4D label-free quantitative proteomic results showed decreased expression of the desuccinylase SIRT5 in rat nucleus pulposus (NP) tissues under mechanical loading. Overexpression of Sirt5 effectively alleviated, whereas knockdown of Sirt5 aggravated, the apoptosis and dysfunction of NP cells under mechanical stress, consistent with the more severe IDD phenotype of Sirt5 KO mice than wild-type mice that underwent lumbar spine instability (LSI) surgery. Moreover, immunoprecipitation-coupled mass spectrometry (IP-MS) results suggested that AIFM1 was a downstream target of SIRT5, which was verified by a Co-IP assay. We further demonstrated that reduced SIRT5 expression resulted in the increased succinylation of AIFM1, which in turn abolished the interaction between AIFM1 and CHCHD4 and thus led to the reduced electron transfer chain (ETC) complex subunits in NP cells. Reduced ETC complex subunits resulted in mitochondrial dysfunction and the subsequent occurrence of IDD under mechanical stress. Finally, we validated the efficacy of treatments targeting disrupted mitochondrial protein importation by upregulating SIRT5 expression or methylene blue (MB) administration in the compression-induced rat IDD model. In conclusion, our study provides new insights into the occurrence and development of IDD and offers promising therapeutic approaches for IDD.
RFID Bi-Directional Authentication Protocol Based on Dynamic Coupled Integer Tent Map

To meet the special needs of RFID, it is proposed to use a coupled dynamic integer tent map lattices model (DCML) to implement a Hash function and a random number generation scheme. The scheme is featured with fast operation. Based on this, this paper proposes RFID bi-directional authentication protocol. This has strictly proved by BAN logic. This protocol can resolve eavesdropping, illegal access, location tracking, impersonation and replay attack.
Flexible Cu2O/Cu film self-powered photodetector for high visible light selection and fast response

Spectral selection is one of the key performance parameters for visible light photodetectors (VLPD). However, as the absorbing layer of the VLPD, the semiconductor has strong ultraviolet light absorption, leading to poor visible light selectivity of the VLPD. This work fabricated a flexible photovoltaic VLPD based on Cu2O/Cu Schottky junction by annealing Cu foil. Under zero bias, the Cu2O/Cu VLPD showed the responsivity of 14 mA/W, the response time of 1 ms, and the detectivity of 8.8 × 1011 Jones to 620 nm light. Moreover, the detection band of the device is in the range of 500–640 nm, indicating excellent visible light selectivity. Optical and electrical characterization shows that the high visible selectivity of the device is mainly attributed to the fact that the penetration depth of visible light in Cu2O film matches the position of the Cu2O/Cu Schottky junction. This, in turn, induces the effective separation of the photogenerated carriers excited by the visible light. In addition, the bend detection performance of the device is demonstrated. This work provides a reference for improving the spectral selectivity of visible photodetectors.
Enhanced energy storage properties in PbZrO3 thin films via the incorporation of NiO

In this study, NiO–PbZrO3 composite films were deposited on SiO2/Si substrates buffered with LaNiO3 films via the sol-gel coating technique. The effects of NiO addition in PZO thin films on the microstructure, dielectric properties, leakage mechanism, ferroelectric properties and energy storage properties have been discussed. The dielectric constant increased with the addition of NiO, while the leakage current density decreased. Compared with pure PZO films, the maximum polarization of the composite films was improved. For the composite films prepared using the NiO precursor solution with 0.05 mol/L, the recoverable energy storage density of the NiO-PZO composite film is up to 19.6 J/cm3 under the electric field of 1038 kV/cm, which is 30% higher than that of the pure PZO film under the same conditions. Also, the energy storage efficiency of the composite film reaches 48%. Accordingly, we demonstrate a simple and convenient method by adding NiO to fabricate thin films with high energy storage performance.
