<P>IL-2 is crucial to T cell homeostasis, especially of CD4<SUP>+</SUP> T regulatory cells and memory CD8<SUP>+</SUP> cells, as evidenced by vigorous proliferation of these cells in vivo following injections of superagonist IL-2/anti-IL-2 antibody complexes. The mechanism of IL-2/anti-IL-2 antibody complexes is unknown owing to a lack of understanding of IL-2 homeostasis. We show that IL-2 receptor α (CD25) plays a crucial role in IL-2 homeostasis. Thus, prolongation of IL-2 half-life and blocking of CD25 using antibodies or CD25-deficient mice led in combination, but not alone, to vigorous IL-2-mediated T cell proliferation, similar to IL-2/anti-IL-2 antibody complexes. These data suggest an unpredicted role for CD25 in IL-2 homeostasis.</P>

CARMA LARGE AREA STAR FORMATION SURVEY: DENSE GAS IN THE YOUNG L1451 REGION OF PERSEUS

<P>We present a 3 mm spectral line and continuum survey of L1451 in the Perseus Molecular Cloud. These observations are from the CARMA Large Area Star Formation Survey (CLASSy), which also imaged Barnard. 1, NGC 1333, Serpens Main, and Serpens South. L1451 is the survey region with the lowest level of star formation activity-it contains no confirmed protostars. HCO+, HCN, and N2H+ (J = 1 -> 0). are all detected throughout the region, with HCO+ being the most spatially widespread, and molecular emission seen toward 90% of the area above N(H-2) column densities of 1.9 x 10(21) cm(-2). HCO+ has the broadest velocity dispersion, near 0.3 km s(-1) on average, compared with similar to 0.15 km s(-1) for the other molecules, thus representing a range of subsonic to supersonic gas motions. Our non-binary dendrogram analysis reveals that the dense gas traced by each molecule has a similar hierarchical structure, and that gas surrounding the candidate first hydrostatic core (FHSC), L1451-mm, and other previously detected single-dish continuum clumps has similar hierarchical structure; this suggests that different subregions of L1451 are fragmenting on the pathway to forming young stars. We determined that the three-dimensional morphology of the largest detectable dense-gas structures was relatively ellipsoidal compared with other CLASSy regions, which appeared more flattened at the largest scales. A virial analysis shows that the most centrally condensed dust structures are likely unstable against collapse. Additionally, we identify a new spherical, centrally condensed N2H+ feature that could be a new FHSC candidate. The overall results suggest that L1451 is a young region starting to form its generation of stars within turbulent, hierarchical structures.</P>
The Immune Landscape of Cancer

<P><B>Summary</B></P> <P>We performed an extensive immunogenomic analysis of more than 10,000 tumors comprising 33 diverse cancer types by utilizing data compiled by TCGA. Across cancer types, we identified six immune subtypes—wound healing, IFN-γ dominant, inflammatory, lymphocyte depleted, immunologically quiet, and TGF-β dominant—characterized by differences in macrophage or lymphocyte signatures, Th1:Th2 cell ratio, extent of intratumoral heterogeneity, aneuploidy, extent of neoantigen load, overall cell proliferation, expression of immunomodulatory genes, and prognosis. Specific driver mutations correlated with lower (<I>CTNNB1</I>, <I>NRAS</I>, or <I>IDH1</I>) or higher (<I>BRAF</I>, <I>TP53</I>, or <I>CASP8</I>) leukocyte levels across all cancers. Multiple control modalities of the intracellular and extracellular networks (transcription, microRNAs, copy number, and epigenetic processes) were involved in tumor-immune cell interactions, both across and within immune subtypes. <P>The commercial polymers poly(ethylene imine) (PEI), poly(ethylene imine epichlorohydrin), and poly(dimethylamine-co-epichlorohydrin) were purified and fractionated by ultrafiltration. Their metal-ion-binding properties with respect to different ligand groups and the effect of the concentration on the retention properties were investigated. The amine ligands of the polymers formed the most stable complexes with the metal ions. In general, there was an effect of the pH and polymer fraction size on the retention properties. As the pH and polymer fraction size increased, the affinity to bind metal ions also increased. PEI had the highest metal-retention values, particularly at higher pHs, at which the amine groups were nonprotonated and could coordinate easily with the metal ions. Only Pb(II) was poorly retained. The affinity for all the metal ions, except Pb(II), increased significantly at pH 5. The metal-ion retention decreased quickly as the filtration factor increased, except for Cu(II), Co(II), Ni(II), Cd(II), and Zn(II) ions, which were retained by over 40% at a filtration factor of 4. <P>Two new supramolecular architectures based on zinc phthalocyanine (Pc) and imidazolyl-substituted perylenediimide (PDI), ZnPc/DImPDI/ZnPc 1 and ZnPc/ImPDI 2, have been prepared. A strong electron-donor, ZnPc-8, which contained eight tert-octylphenoxy groups was synthesized to ensure high solubility, thereby reducing aggregation in solution and providing ??-donor features while avoiding regioisomeric mixtures. Also, PDI units were functionalized with tert-octylphenoxy groups at the bay positions, which provide solubility to avoid aggregation in solution, together with one and two imidazole moieties in the amide position, PDI-6 and PDI-4, respectively, to be able to strongly coordinate with the ZnPc complex. Supramolecular complexation studies by (1)H NMR spectroscopy and ESI-MS demonstrate a high coordinative binding constant between imidazole-substituted PDI-4 or PDI-6 and ZnPc-8. The same results were confirmed by UV/Vis and fluorescence titration studies. UV/Vis titration studies revealed the formation of a 1:1 complex ZnPc/ImPDI 2 for the systems ZnPc-8 and PDI-6 and a 2:1 complex ZnPc/DImPDI/ZnPc 1 for the interaction of ZnPc-8 and PDI-4. The binding constant in both cases was determined to be on the order of 10(5) M(-1). Femtosecond laser flash photolysis measurements provided a direct proof of the charge-separated state within both supramolecular assemblies by observing the transient absorption band at 820 nm due to the zinc phthalocyanine radical cation. The lifetimes of charge-separated states are (9.8±3) ns for triad 1 and (3±1) ns for dyad 2. As far as we know, this is the first time that a radical ion pair has been detected in a supramolecular assembled ZnPc-PDI system and has obtained the longest lifetime of a charge-separated state published for ZnPc-PDI assemblies.</P>
Graded Otx2 activities demonstrate dose-sensitive eye and retina phenotypes

<P>In the human, mutations of <I>OTX2</I> (<I>Orthodenticle homeobox 2</I> transcription factor) translate into eye malformations of variable expressivity (even between the two eyes of the same individual) and incomplete penetrance, suggesting the existence of subtle thresholds in OTX2 activity. We have addressed this issue by analyzing retinal structure and function in six mutant mice with graded Otx2 activity: <I>Otx2<SUP>+/+</SUP></I>, <I>Otx2<SUP>+/AA</SUP></I>, <I>Otx2<SUP>+/GFP</SUP></I>, <I>Otx2<SUP>AA/AA</SUP></I>, <I>Otx2<SUP>AA/GFP</SUP></I> and <I>Otx2<SUP>GFP/GFP</SUP></I>. Null mice (<I>Otx2<SUP>GFP/GFP</SUP></I>) fail to develop the head and are embryonic lethal, and compound heterozygous <I>Otx2<SUP>AA/GFP</SUP></I> mice show a truncated head and die at birth. All other genotypes develop until adulthood. We analyzed eye structure and visual physiology in the genotypes that develop until adulthood and report that phenotype severity parallels Otx2 activity. <I>Otx2<SUP>+/AA</SUP></I> are only mildly affected whereas <I>Otx2<SUP>+/GFP</SUP></I> are more affected than <I>Otx2<SUP>+/AA</SUP></I> but less than <I>Otx2<SUP>AA/AA</SUP></I> mice. <I>Otx2<SUP>AA/AA</SUP></I> mice later manifest the most severe defects, with variable expressivity. Electrophysiological and histological analyses of the mouse retina revealed progressive death of bipolar cells and cone photoreceptors that is both Otx2 activity- and age-dependent with the same ranking of phenotypic severity. This study demonstrates the importance of gene dosage in the development of age-dependent pathologies and underscores the fact that small gene dosage differences can cause significant pathological states.</P>

<P>The commercial, water-soluble poly(sodium anetole sulfonate) (PAnS), poly(sodium vinylsulfonate) (PVS), and poly(sodium 4-styrene sulfonate) (PStyS) were investigated as metal ion binding reagents using the liquid-phase-polymer-based retention (LPR) technique. The retention profiles of the following metal ions: Co(II), Ni(II), Ca(II), Mg(II), Pb(II), Cd(II), Zn(II), Cr(III), and Cu(II) were obtained by the LPR technique at pH 3, 5, and 7, and by changing the filtration factor Z from 0 to 10. As the pH increased, the affinity towards the metal ions increased slightly. By increasing the filtration factor Z, the affinity for the metal ions did not decrease significantly. This means that the ligand–metal interaction was strong, and it is not possible to break it by washing with water at the same pH of the filtration cell. Copyright © 2006 John Wiley & Sons, Ltd.</P>

<P>Hybrid porous solids, with their tunable structures, their multifunctional properties and their numerous applications, are currently topical, particularly in the domain of adsorption and storage of greenhouse gases. Most of the data reported so far concern the performances of these solids in this domain, particularly in terms of adsorbed amounts of gas but do not explain at the atomic level why and how adsorption and storage occur.
