Effects of all-trans retinoic acid(retinoic acid) on DNA replication, H2B histone and DNA topoisopmerase I(Topo I) gene expression have been investigated in human promyelocytic leukemia cell line HL-60. DNA synthesis decreased at 24 hours after exposure of retinoic acid in the HL-60 cells. H2B histone mRNA rapidly reduced at 48 hours and Topo I and c-myc mRNA at 24 hours in retinoic acid-exposured HL-60 cells, respectively. The levels of c-myc, H2B histone and Topo I gene expression were reduced in proportion to the concentration of retinoic acid. The H2B histone mRNA in retinoic acid-exposured cells was elevated by cycloheximide treatment, while the level of Topo I mRNA was constant. These results suggest that regulations of H2B histone, c-myc and Topo I gene expression are different from one another, and repression of Topo I gene is closely correlated with c-myc gene during retinoic acid-induced differentiation of HL-60 cells.

In this paper, we have been studied the enhancement of SLAM image using average response methos. AR methos has been used to reduce the random noise in signal processing. In this the experiment, we have constructed the SLAM system of 10㎒ operating frequency and experienced for holed Al sample. In this experimental result, we have been found that SLAM using AR method was able to get the high-speed and enhanced image compared with SAM.

Effects of the differentiation inducers on DNA synthesis and c-myc gene expression have been investigated during the differentiation of human promyelocytic leukemia cell line HL-60. All-trans retinoic acid(retinoic acid) and dimethyl sulfoxide(DMSO) decreased DNA synthesis at 24 hours and 12-O-tetradecanoylphorbol 13-acetate(TPA) decreased at 12 hours after exposure, respectively. On the other hand control, no adding differentiation inducers, gradually increased DNA synthesis. The c-myc mRNA was sharply reduced at 24 hours in retinoic acid-, at 4 hours in DMSO-, and at 1 hour in TPA-exposured HL- 60 cells. The level of c-myc mRNAs was reduced in proportion to the concentration in all of three differentiation inducers. The level of c-myc mRNAs in the differentiation inducers-exposured cells were elevated by cycloheximide treatment. These results suggest that c-myc gene expression and differentiation mechanism are variable according to the kinds of the differentiation inducers in HL-60 cells.

The fate of diazinon in the intact rice plants and submerged paddy soil has been investigated with (??) diazinon, The labelled diazinon solution was applied to paddy water and distribution of radioactivities in the rice seedlings, paddy soil, volatile fraction and carbon dioxide has been ascertained at end the of incubation times of 0.5,1,4,6 and 9 days respectively. In addition, extract of plants and paddy soils were subjected to TLC separation for examination of possible transformation products of diazinon. The results may be summarized as follows; 1. Total recoveries of radiactivities were between 57.2~73.6 per cent. 2. Radioactivity in rice seedlings increased with incubation periods reaching one tenth of treated radioactivity at the end of 9 day incubation. 3. Non-extractable radioactivity in paddy soil increased with incubation periods. 4. Radioactive volatile fraction increased in the presence of rice seedlings. 5. Pyrimidinol was unique conversion product of diazinon in rice seedlings and paddy soils. 6. Pyrimidinol applied to paddy soil is readily absorbed by rice seedlings.

Five monoclonal antibodies (MAbs) and chicken immunoglobulin (IgY) were developed by immunizing with flagella purified from Listeria monocytogenes 4b and the five MAbs have been confirmed to be specific against three different epitopes of flagellin. The antibodies showed specific reaction to Listeria genus and no cross-reactivity with other bacteria tested in this experiment including E.coli O157:H7 and Salmonella enteritidis. Sandwich enzyme-linked immunosorbent assays (ELISA) using the MAbs and IgY were developed to detect Listeria species and the sensitivity and specificity of the developed ELISA have been analyzed. The detection limit of ELISA using MAb 2B1 and HRP labeled IgY was 1 × 105 cells/0.1 ml at 22oC and 1 × 106 cells/0.1 ml at 30oC. ELISA using the pair of MAbs (MAbs 2B1 and HRP labeled MAbs 7A3) detected up to 104 cells/0.1 ml at 22oC and 30oC. Detection limit of sandwich ELISA using IgY was 10 times lower than MAb pair. Using the developed ELISA, we could detect several Listeria contaminated in food samples after 48 h-culturing. In conclusion, both MAbs and IgY have been proved to be highly specific to detect Listeria flagella and the developed sandwich ELISA using these antibodies would be useful tool for screening Listeria spp. in food.

Lichen myxedematosus(LM), also known as papular mucinosis, is a rare, idiopathic disorder caused by the focal dermal deposition of mucin and occurs both as an isolated entity and manifestations of various systemic disorders. Cutaneous disease is common in patients infected with human immunodeficiency virus(HIV) and cutaneous mucinosis has been reported with increased frequency in HIV patients. A male patient with HIV infection was consulted to our clinic for evaluation of asymptomatic diffesely-scattered papules on his upper extremities and trunk. Histopathological finding was the deposition of mucin in the dermis and lichen myxedematosus type 2 was diagnosed. We present of a case of lichen myxedematosus in a HIV-infected male patient. We have investigated DNA synthesis and levels of H2B histone mRNA, and the binding pattern of nuclear proteins to various elements in the rat H2B histone gene upstream region with DNase I footprinting assay. Both DNA synthesis and H2B histone mRNA level were increased with maximal stimulation reached at 24 hrs and 36 hrs after partial hepatectomy, respectively. In DNase I footprinting analysis, the nuclear factors interacting with the three elements, TATA at - 19 bp (site A^R), site B at-29 bp, and CAAT at-69 bp (site C) were required during maximal increase of H2B histone mRNA level after partial hepatectomy. The DNase I protection pattern by nuclear extract of the cycloheximide-treated regenerating liver showed the same result with normal liver. These results suggest that transcriptional regulation of H2B histone gene during liver regeneration may be mediated by nuclear factors that are newly induced by paritial hepatectomy.
