        YPEL3 expression induces cellular senescence via the Hippo signaling pathway in human breast cancer cells

        The Hippo pathway is a signaling pathway that controls organ size in animals by regulating cell proliferation and apoptosis. Yes-associated protein 1 (YAP1), an oncogene associated with the development and progression of breast cancer, is downregulated by the Hippo pathway and is associated with the development and progression of breast cancer. Yippee-like 3 (YPEL3) is a target gene of the tumor suppressor protein p53, and its activation has been shown to inhibit cell growth, induce cellular senescence, and suppress tumor cell metastasis. In this study, we found that YAP1 inhibits the expression of YPEL3 expression in breast cancer cells. Furthermore, a decrease in lamin B1, a marker protein of cellular senescence, coupled with the activation of senescence-associated β-galactosidase indicated that upregulating YPEL3 levels through YAP1 downregulation can induce cellular senescence. Additionally, elevated YPEL3 levels resulted in higher levels of oxygen consumption rate in mitochondria, thus promoting apoptosis. This suggests that YPEL3 plays a crucial role in regulating oxidative stress and cell apoptosis in breast cancer cells. Therefore, the interaction between YAP1 and YPEL3 represents a novel mechanism of cellular senescence mediated by the Hippo signaling pathway. Collectively, our findings suggest that the Hippo signaling pathway plays an important role in regulating cellular senescence, which could have implications for the development of new therapeutic strategies for diseases such as cancer.

        Induction of synergistic apoptosis by tetramethoxystilbene and nutlin-3a in human cervical cancer cells

        2,4,3’,5′-Tetramethoxystilbene (TMS) is a selective inhibitor of cytochrome P450 1B1 to block the conversion from estradiol to 4-OH-estradiol. Several studies suggested that TMS may act as a potent anti-cancer agent for hormone-related cancer including cervical cancer. Nutlin-3a is a cis-imidazoline analog that interferes with the interaction between mouse double minute 2 homolog (MDM2) and the tumor suppressor p53. The purpose of the study was to compare the cytotoxic effect of TMS and nutlin-3a treatment individually and in combination in HeLa cells. To assess the potential synergistic effects between TMS and nutlin-3a, low concentrations of TMS and nutlin-3a were simultaneously treated in HeLa cells. Based on cell viability, apoptosis assays, and the increase in cleaved caspase-3 and poly (ADP-ribose) polymerase cleavage, it was demonstrated that the combination with TMS and nutlin-3a exerts a synergistic effect on cancer cell death. Isobologram analysis of HeLa cells noted synergism between TMS and nutlin-3a. The combined treatment increased the expression of mitochondrial pro-apoptotic factors such as Bax and Bak, and decreased the expression of the XIAP. In addition, combination treatment significantly enhanced the translocation of AIF to the nucleus in HeLa cells. In conclusion, the results demonstrate that the combination of TMS and nutlin-3a induces synergistic apoptosis in HeLa cells, suggesting the possibility that this combination can be applied as a novel therapeutic strategy for cervical cancer.

      • Papain 유도 호흡기 염증모델에서 인삼패독산(人蔘敗毒散)의 항염증 효과

        Objectives: Insampaedok-san and its main components have been used for treatment of fever, joint pain, cough, headache and pain. Using a respiratory inflammation model, we intend to investigate the anti-inflammatory effect and its mechanism of Insampaedok-san. Methods: We induced the actue respiratory inflammatory mouse model by papain treatment. BALB/C mice (female, 8 weeks old) were divided into three groups as follows: saline control (SC): papain treated group (vehicle): papain and Insampaedok-san (200mg/kg) treated group (n=4). To demonstrate the ani-inflammatory effect of Insampaedok-san water extracts, inflammatory cell infiltration was evaluated in bronchoalveolar lavage fluid (BALF) and nasal lavage fluid (NALF). In addition, immunogloblin (Ig)-E level was measured by using ELISA. Results: Insampaedok-san extracts administration decreased inflammatory cell infiltration in BALF and NALF. Especially, eosinophil infiltration was dramatically reduced. Furthermore, total Ig-E level was diminished in BALF and serum by Insampaedok-san administration. Conclusions: Our findings revealed Insampaedok-san extracts have anti-inflammatory effects by inhibiting inflammatory cell, especially eosinophil infiltration and Ig-E level.

        Clinical Implication of Candida Score in Multidrug-Resistant Pneumonia with Airway Candida Colonization

        Background The growth of Candida in respiratory secretions is usually considered colonization, and antifungal therapy is rarely required. The role of Candida colonization in the progression of bacterial pneumonia remains controversial. The aim of this study was to identify the clinical implication of Candida score by analyzinge the relationship with multidrug-resistant (MDR) pneumonia and prognosis in patients with airway Candida colonization. Materials and Methods This study was a retrospective review of patients with airway Candida colonization by bronchial washing or bronchoalveolar lavage. The Candida score was calculated according to the four factors (severe sepsis, surgery at baseline, total parenteral nutrition, and multifocal Candida colonization). Pneumonia related mortality or hopeless discharge expecting death was defined as a poor outcome. Results A total of 148 patients were enrolled in the study. In a multivariate analysis model, Candida score was identified as an independent predictor of poor outcomes (odds ratio 2.23; 95% confidential interval 1.57 – 3.17; P <0.001) in pneumonia patients with airway Candida colonization. With a Candida score of three or higher compared with low score group, it was associated with bacterial pneumonia, especially methicillin-resistant Staphylococcus aureus (MRSA) infection (0.0% vs. 15.2%, P = 0.004). In addition, patients with a high Candida score had a longer hospital stay (13 vs. 38 days, P <0.001), longer duration of intensive care (7 vs. 18 days, P <0.001), and higher pneumonia-related mortality (0.0% vs. 45.5%, P <0.001) as compared to the low Candida score group. The Candida score showed a positive correlation with other pneumonia severity scales such as CURB-65 (Confusion, Urea, Respiratory rate, Blood pressure, and age ≥65 years) (r = 0.461, P <0.001), Pneumonia Severity Index (r = 0.397, P <0.001), and predisposition, insult, response, and organ dysfunction (PIRO) score (r = 0.425, P <0.001). Conclusion This study revealed that Candida is no longer a bystander of airway colonization, and that it affects the progression of bacterial pneumonia, including multidrug-resistant pathogens, particularly MRSA infection. Also Candida score can be used to predict the prognosis of patients with pneumonia.         드론을 활용한 다양한 산업분야의 성장과 낙관적인 경제 효과가 전망됨에 따라 앞으로도 급격한 드론 시장의 발전이 예상된다. 하지만, 드론 운용을 위한 교통관리 체계 미흡으로 안전, 사생활 침해 등의 문제가 대두되고 있다. 본 논문에서는 저고도 공역에서 안전하고 효율적인 무인기 운용을 위해 수립되고 UTM (Unmanned Aircraft System Traffic Management) 운용개념과 발전 로드맵을 해외 사례를 통해 파악해 보았다. 이를 통해, 우리나라 또한 실정에 맞는 저고도 무인기 교통관리 체계를 구축하여 드론의 적극적 활용을 기대해본다. With the growth of the various fields utilizing drones and the optimistic prospect of their economic effects, rapid development of the drone markets is expected. However, many problems such as security and privacy problems have been issued due to lack of traffic management for drone operations. In this paper, we surveyed the concept of operations for UTM (Unmanned Aircraft System Traffic Management) proposed by NASA and EASA for the secure and effective operations of drones in the low altitude airspace. The survey results will be used for the references to build Korean UTM system, which is a core infrastructure for the safe and efficient operations of drones in Korea.

        배우자 사별을 한 여성 독거노인의 삶의 적응경험에 관한 연구

        본 연구는 여성 독거노인이 배우자 사별 후의 적응과정과 삶에 대한 태도와 인식에 대해 알아보고 배우자 사별을 한 여성 독거노인을 위한 사회복지 실천적 방안을 모색하는데 목적이 있다. 이를 위해 배우자 사별을 한 여성 독거노인 8명을 대상으로 심층면접을 실시하였으며, Colaizzi의 현상학적 분석방법을 이용하여 5개의 범주와 11개의 하위범주를 추출하였다. 연구결과를 통하여 배우자를 사별한 여성 독거노인을 위한 실천적 방안을 제시하면 다음과 같다. 첫째, 상담 프로그램의 개발과 심리치료서비스를 제공할 수 있는 전문기관의 연계가 필요하다. 둘째, 안정된 노후 생활을 잘 준비하고 적응하도록 가족 및 이웃의 지원체계가 필요하며, 사별에 대비한 가족교육, 사회교육이 필요하다. 셋째, 배우자를 사별 한 여성 독거노인들이 서로의 고통을 이해하고 공감할 수 있는 자조모임이 활성화 되어야 하며, 이를 위한 적합한 환경과 프로그램 개발이 이루어져야 할 것이다. This research aims to develop social welfare practical services for elderly women who end up living alone after their spouse died based on how old women adapt and how their attitude and perception about life change. As a method of study, eight senior citizens who live alone after spouses died were interviewed in depth, and Colaizzi’s phenomenological analysis method was used to extract five categories and 14 sub-categories. The result of the research suggests practical measures for elderly women who have lost their spouses. First, the development of counseling programs are required and the formation of links between specialized agencies capable of providing psychological treatment services are needed. Second, Family and neighbor support systems are asked and relevant educations are required for prepare adaptation and stable retirement life of elderly women. Third, a self-help group among elderly women who live alone should be activated to understand and sympathize with each other's pain and proper environment and program development should be implemented before.
