pH-Responsive globular poly(ethylene glycol) for photodynamic tumor therapy

<P>In this study, we report the development of extremely small-sized globular poly(ethylene glycol) (gPEG) that can specifically recognize tumor acidic pH. gPEG coupled with chlorin e6 (Ce6, a photosensitizing drug) and 2,3-dimethylmaleic acid (DMA, as a pH-responsive moiety) (gPEG-Ce6-DMA, particle size: 3-4 nm in diameter) was easily dispersed in phosphate buffered saline (PBS) without any of the nanoparticle fabrication steps. We observed that gPEG-Ce6-DMA displayed pH-dependent zeta-potential changes due to coupling (at pH 7.4) or decoupling (at pH 6.8-6.0) of DMA. As a result, the uptake of gPEG-Ce6-DMA was significantly increased in tumors at acidic pH, likely due to the decoupling of DMA (backing cationic primary amines). As a result, the preferential cellular uptake of gPEG-Ce6-DMA at acidic pH allowed for a significant enhancement of in vitro/in vivo photodynamic tumor cell ablation under light illumination. We report the characteristics of three cell lines (designated, SNU-80, SNU-373 and SNU-790), which were established from two papillary carcinomas and one anaplastic carcinoma obtained from three Korean thyroid carcinoma patients. All cell lines grow as adherent cells. Electron microscopy characteristically showed cytoplasmic invaginations of nuclei and intranuclear cytoplasmic inclusions. SNU-80 and SNU-790 cells showed a positive reaction to anti-cytokeratin antibody, and SNU-790 cells positivity for CK-19. All lines were free of mycoplasma or bacteria and were proven unique by DNA fingerprinting analysis. The p15 and p16 genes are deleted in the SNU-790 line. Mutations of the p53 gene were found in two lines (SNU-80 and SNU-373), but no mutations in the RET or MEN1 genes were observed. Mutations of the BRAF gene were found in the SNU-80 (G468R) and the SNU-790 (V599E) cell lines, but no mutations in the K-ras gene were present. SNU-80 and SNU-790 cells showed a positive reaction to anti-cytokeratin antibody, and no evidence of the production of thyroglobulin or calcitonin was observed. The cell lines were unable to trap radioactive iodine but did not contain TSH receptor. In addition, we investigated the mRNA expression levels of Tg, TSHR, TTF-1, PAX-8, NIS, IL-6, and LIF, and of the α, β and γ retinoic acid receptors in these cell lines. IL-6 was down-regulated in all three cell lines by all-trans-retinoic acid treatment. RAR-α was expressed but RAR-β was not expressed in the three cell lines, and RAR-γ was not expressed in SNU-790. Interestingly, RAR-β (SNU-80 and SNU-373) and RAR-γ (SNU-790) was up-regulated by all-trans-retinoic acid treatment. We believe that these well-characterized thyroid carcinoma cell lines may be useful tools for investigations on the biological characteristics of thyroid carcinoma, particularly for investigations related to gene alterations, especially of the BRAF gene. These cell lines may also be useful for redifferentiation therapy studies on thyroid carcinoma using all-trans-retinoic acid.

<P>Middle East respiratory syndrome coronavirus (MERS-CoV) causes severe acute respiratory illness with fever, cough and shortness of breath. Up to date, it has resulted in 1826 human infections, including 649 deaths. Analogous to picornavirus 3C protease (3C(PrO)), 3C-like protease (3CL(PrO)) is critical for initiation of the MERS-CoV replication cycle and is thus regarded as a validated drug target. As presented here, our peptidomimetic inhibitors of enterovirus 3C(PrO) (6b. 6c and 6d) inhibited 3CLPr0 of MERS-CoV and severe acute respiratory syndrome coronavirus (SARS-CoV) with IC50 values ranging from 1.7 to 4.7 M and from 0.2 to 0.7 M, respectively. In MERS-CoV-infected cells, the inhibitors showed antiviral activity with EC50 values ranging from 0.6 to 1.4 1.1M, by downregulating the viral protein production in cells as well as reducing secretion of infectious viral particles into culture supernatants. They also suppressed other cc and 8-CoVs from human and feline origin. These compounds exhibited good selectivity index (over 70 against MERS-CoV) and could lead to the development of broad-spectrum antiviral drugs against emerging CoVs and picornaviruses. Recently, the tooling system that is essential to high speed machining-centers is changing from a BT tooling system to a HSK tooling system, because the bending stiffness and the machining accuracy of a HSK tooling system are higher than those of a BT tooling system. So we are developing a new HSK tooling system for high speed machining-centers presently. In this system, a gripper is a core part that clamps a tool-shank on a spindle shaft. In this paper, we selected the most stable shape in three models of the gripper. In order that, we modeled three different models of the gripper by using a 3D modeling/design program. Then we analyzed Stress Distribution Characteristics by using the Finite Element Analysis (FEA).

Selection of a right weather data set has been considered as one of important factors for a successful building energy audit process. A set of 30 year raw weather data base for six major cities has been developed to provide the weather data file for building energy audit and retrofit analysis in Korea. The program named as KWDP (KIER Weather Data Processor) uses the DB to produce a right data set for a specific building energy performance simulation program like DOE2.1E. A program called WMAKE has been developed to generate the right set of input parameters for DOE2.1E weather utility program. The set of the programs could provide the right weather data for specific building energy audit and retrofit analysis.
Dark green discoloration of the urine after prolonged propofol infusion: a case report

<P><B>Summary</B></P><P><B>What is known and Objective: </B> Propofol, a commonly used sedative, has on rare occasions, been reported to discolour urine green. However, in previous reports, it is uncertain that whether this colour change is dose dependent. We report on a patient who produced dark green discoloration of urine from prolonged propofol infusion, administered for intractable epilepsy.</P><P><B>Case summary: </B> The colour intensity of the patient’s urine was dependent on propofol infusion rate. Reducing propofol infusion rate lightened the colour of the urine, eventually back to normal.</P><P><B>What is new and Conclusion: </B> Green discoloration of the urine from propofol infusion is dose dependent. It is usually benign and reversible, as was the case for our patient.</P>
