<P>Breast cancer is the most common cancer for women and is a major cause of mortality in women. Doxorubicin is a generally used chemotherapy drug for breast cancer. However, multidrug resistance of breast cancer interferes with the chemotherapy. We examined whether cucurbitacin D affects doxorubicin resistance of MCF7/ADR breast cancer cells. Cell viability was measured by MTT assay. Levels of p-STAT3, p-NF-κB, IκB, and caspases were measured by Western blot analysis. Nuclear staining of Stat3 and NF-κB was measured by immunocytochemistry. STAT3 and NF-κB transcriptional activity was detected by STAT3 and NF-κB luciferase reporter gene assays. Analysis of cell cycle arrest was performed by flow cytometry. Induction of apoptosis by cucurbitacin D was measured by Annexin V-FITC/propidium iodide assay. More than 90 % of MCF7/ADR cells lived upon treatment with doxorubicin for 24 h. However, upon treatment with cucurbitacin D, cell death was more than 60 %. Co-administration of cucurbitacin D and doxorubicin induced apoptosis, and G2/M cell cycle arrest, and inhibited upregulated Stat3 by doxorubicin on MCF7/ADR cells. Additionally, cucurbitacin D led to an increase in the IκBα level in the cytosol and a decrease in the p-NF-κB level in the nucleus. Finally, cucurbitacin D inhibited translocation of Stat3 and NF-κB and decreased transcriptional activity in the nucleus. Cucurbitacin D decreases cell proliferation and induces apoptosis by inhibiting Stat3 and NF-κB signaling in doxorubicin-resistant breast cancer cells. Cucurbitacin D could be used as a useful compound to treat adriamycin-resistant patients.</P>

통계상 국민 2명중 1명이 차를 소유하고 있는 오늘 날, 뉴스를 통해서 차량사고에 의한 인명사고 소식을 심심치 않게 접할 수 있다. 그 중 많은 비중을 차지하고 있는 사고는 승 하차시의 접촉 사고로, 최근 들어 증가하는 추세이다. 그래서 이러한 자동차 접촉사고에서의 자동차 탑승자의 안전을 지키기 위한 대책이 시급하다고 생각하였고, 이에 본 논문에서는 뒤의 다가오는 물체에 대한 속도측정을 통한 자동 차문 잠김을 대안으로 제시하며, 이를 통해서, 자동차 탑승자들의 안전을 보장하는 것에 보다 더 한걸음 나아가고자 한다.
Amounts of gingival crevicular fluid according to age and the relationship with oral micro-organism

Objectives: The objective of this study is to present, inquired the gingival crevicular fluid(GCF) by age and sex, and the relationship with other oral factors to use as base information of individual oral state. Methods: The study was done, classified with 6 group by age and 30 males and the same number of females were included, summing as 360 subjectives. Oral examination was done and amounts of GCF was measured with Periotron. Moisture paper strip was applied for 60 subjectives in order to estimate the correlationship coefficient between the value measured by Periotron and checked by moisture paper strip. Another 30 of adults subjectives were participated for oral micro-organism examination and correlation coefficient was estimated between the GCF amount and the amount/motile of each oral micro-organism. Results: GCF volumes at molar tooth by Periotron are ranged from 38.52 at adult to 65.53 at senile, and at child and senile the volume is significantly high. The GCF volume at front tooth are ranged from 38.57 in adult to 66.23 in senile, and in senile the volume is significantly high. GCF volume and clinical state of gingival has correlations, especially gingival index relationship being at molar part r=0.381, and frontal part r=0.465. There is relationship between GCF volume and amount of comma/spiral(r=0.394, p<0.05), filament(r=0.387, p<0.05), and bacilli(r=0.367, p<0.05) and motile of filament(r=0.378, p<0.05). There is relationship between GCF volume and value of moisture paper exam(r=0.315, p<0.05). Conclusion: Clinically, it is considered that clinical test of gingivitis can be used by measuring of the volume of GCF, one of the important informations of individual periodontal status. Analgesic Effects of Intrathecal Curcumin in the Rat Formalin Test

Background: Curcumin has been reported to have anti-inflammatory, antioxidant, antiviral, antifungal, antitumor, and antinociceptive activity when administered systemically. We investigated the analgesic efficacy of intrathecal curcumin in a rat model of inflammatory pain. Methods: Male Sprague Dawley rats were prepared for intrathecal catheterization. Pain was evoked by injection of formalin solution (5%, 50 μl) into the hind paw. Curcumin doses of 62.5, 125, 250, and 500 μg were delivered through an intrathecal catheter to examine the flinching responses. The ED50 values (half-maximal effective dose) with 95% confidence intervals of curcumin for both phases of the formalin test were calculated from the dose-response Lines fitted by Least-squares Linear regression on a log scale. Results: In rats with intrathecal administration of curcumin, the flinching responses were significantly decreased in both phases. The slope of the regression line was significantly different from zero only in phase 2, and the ED50 value (95% confidence interval) of curcumin was 511.4 μg (23.5?1126.5). There was no apparent abnormal behavior following the administration of curcumin. Conclusions: Intrathecal administration of curcumin decreased inflammatory pain in rats, and further investigation to elucidate the precise mechanism of spinal action of curcumin is warranted.
