Comprehensive Review of Golgi Staining Methods for Nervous Tissue

Golgi staining has been modified and developed since Camillo Golgi introduced the black reaction in 1873. This study focuses on the commonly used Golgi staining methods and presents comprehensive data regarding three Golgi staining methods along with their strong and weak points. The Golgi-Cox method uses mercuric chloride for brain tissue impregnation and is a reliable technique for analyzing the complete dendritic tree of cortical neurons. However, specimens tend to shrink during the staining steps. Recent combination of the Golgi-Cox method and immunofluorescence provides additional options for neuroscientists. Rapid Golgi staining requires osmium tetroxide for the post-fixation process. It homogenously stains whole structures of neurons and provides their detailed anatomical morphology. This staining is influenced by the age of the specimen, temperature of the laboratory, and duration of each procedure. The Golgi-Kopsch method uses formaldehyde and glutaraldehyde instead of osmium tetroxide and can be used regardless of the age of the specimen and the duration after fixation. This method is suitable for research using human brain fixed for a long time or for specimens obtained from old-aged animals. Selecting a Golgi staining protocol that is appropriate for the specimen type and research purpose is important to achieve best results.

Gap junctional intercellular communication (GJIC) plays a key role during development, process of tissue differentiation, and in maintenance of adult tissue homeostasis. Neural stem cells leading to formation of cell clusters termed “neurospheres”, can differentiate into neurons, oligodendrocytes, and astrocytes. We investigated the expression levels and distribution of connexin43 (Cx43) and connexin32 (Cx32), abundant gap junctional protein in neural cells and in neurospheres isolated from rat fetus embryonic day (ED) 17. During differentiation of neurospheres, expression of Cx43 and 32 were increased time-dependently within 72 h, and then decreased at 7 day in western blot analysis. TPA-induced inhibition<br/> of GJIC was confirmed by decreased fluorescence by SL/DT assay, and induced hyperphosphorylation of Cx43 while no changes in Cx32 levels in western blot assay. Our results indicate that GJIC may be a crucial role in the differentiation of neuronal stem cell. And this GJIC can be inhibited by TPA through the hyperphosphorylation of Cx43.
Evaluating the Efficacy of Anti-wrinkle Products in USA

Introducing to the market place, safe and effective product is an important responsibility of clinical investigators as well as regulatory agencies in all developed countries. Products claiming to improve skin wrinkles are no exceptions. To date, Renova(R) (all-trans retinoic acid), Avage(R) (tazarotene), and Botox(R) (botulinum toxin) are the only agents FDA approved to ameliorate wrinkles associated with photoaged skin in the USA. For all three, clinical evaluation of wrinkle severity was the primary endpoint required for the approval process. No sophisticated instrument measurements of wrinkles were required, nor used in the pivotal studies. The Division of Dermatologic & Dental Products of the US FDA (Director, Jonathan Wilkin, MD) is not against the use of mechanical instruments in assessing wrinkle severity. Its position on this issue however, remains that any such device must be grounded in patients' or product users' perspective, which means that the evaluation instrument must be clinically relevant and clinically perceptible. Sophisticated devices that can detect minimal improvement, but imperceptible to the users are considered useless in the eyes of the US FDA. Two instruments that have been tried in some antiwrinkle studies in the USA are silicone replicas and Primos. Despite their sophistications, they have clear limitations; thus have never replaced clinical evaluations in these studies. At most, they have served as secondary measures to provide corroborative data on the clinical efficacy of antiwrinkle products. For the foreseeable future, at least in the USA, careful clinical assessment of wrinkles will continue to serve as the critical benchmark to determine whether an antiwrinkle product has enough efficacy to benefit its users. We must not lose sight of the fact that sophisticated devices are only to serve in generating supportive evidence, and not the primary evidence, in any clinical studies.

Purpose - Facing the FTA era, Jeju Special Self-Governing Province is focusing on the improvement and the expansion of support policies to industrial companies and trade basis systems. However, there are only few studies about human resource development for Jeju Special Self-Governing Province. Moreover, it is difficult to conduct trade-related business efficiently because Jeju is suffering from the lack of the infrastructures of human resources for trade. In this thesis, theoretical approach and empirical approach are used to study about human resource development program of trade for Jeju Special Self-Governing Province. Research design, data, and methodology - In the theoretical approach, economic trends and employment trends of Jeju special self-governing province are investigated about statistical data from 2004 to 2016. In the empirical approach, a questionnaire survey is performed to analyze the employment status of trade professionals. The questionnaire survey targets are export companies and university students in Jeju. The results of the investigations show the export companies highly require more trade professionals and the university students want the jobs which meet their conditions. The major reasons for this employment mismatch are the lack of practical business abilities and the disagreement over pay conditions. Results - The analysis on the current state of the employment and the sense of employment between the exporters and undergraduates in Jeju area shows the major key point is the awareness gap between employer and job hunter , and the political institutional problems. Conclusions - To overcome these problems, four methods for human resource development are proposed. First, Jeju local government should expand the support policies of human resource development which include educational trade programs for university students. Second, the improvement of consciousness about export mind of Jeju local government. Third, university education curriculums should be improved and practically experienced experts shoulde be invited to inspire the university students. Fourth, industry- government-university collaborations should be enhanced to maintain the human resource development program during the long-term. This study on human resource development program of the trade is expected to contribute to the growth of trade infra of Jeju special self-governing province.
Effect of Incorporation Levels of Green Manure Crops on Rice Yield and Soil Chemical Properties

This study was conducted to investigate rice yield and soil chemical properties affected by incorporation levels of green barley (GB) and hairy vetch (HV). The GB and HV were applied to the pots at 500, 1,000, 1,500, and $2,000kg\;10^{-1}$ (GB500, GB1000, GB1500, GB2000, HV500, HV1000, HV1500, and HV2000, respectively), and inorganic fertilizer ($N-P-K=9-4.5-5.7kg\;10a^{-1}$) treatment as control. After rice harvesting, chemical properties of soil such as pH, OM, T-N, and available $P_2O_5$ in GB and HV treated treatments were improved over those in Control treatment. The rice yield ranged from 433 to $512kg\;10a^{-1}$ for GB treatments and 490 to $532kg\;10a^{-1}$ for HV treatments, indicating that rice yield was affected by incorporation levels of GB and HV. The rice yields in GB2000, HV1000, HV1500 and HV2000 treatments increased by 3.3, 3.1, 6.4 and 7.4% compared with Control treatment, respectively. Therefore, minimum incorporation level of GB and HV for increasing rice yield was $2,000kg\;10a^{-1}$ of GB and $1,000kg\;10a^{-1}$ of HV.
