Pretreatment with Lycopene Attenuates Oxidative Stress-Induced Apoptosis in Human Mesenchymal Stem Cells

( Ji Yong Kim ),( Jai Sung Lee ),( Yong Seok Han ),( Jun Hee Lee ),( Inhyu Bae ),( Yeo Min Yoon ),( Sang Mo Kwon ),( Sang Hun Lee ) 한국응용약물학회 2015 Biomolecules & Therapeutics(구 응용약물학회지) Vol.23 No.6

Human mesenchymal stem cells (MSCs) have been used in cell-based therapy to promote revascularization after peripheral or myocardial ischemia. High levels of reactive oxygen species (ROS) are involved in the senescence and apoptosis of MSCs, causing defective neovascularization. Here, we examined the effect of the natural antioxidant lycopene on oxidative stress-induced apoptosis in MSCs. Although H2O2 (200 mM) increased intracellular ROS levels in human MSCs, lycopene (10 μmM) pretreatment suppressed H2O2-induced ROS generation and increased survival. H2O2-induced ROS increased the levels of phosphorylated p38 mitogen activated protein kinase (MAPK), Jun-N-terminal kinase (JNK), ataxia telangiectasia mutated (ATM), and p53, which were inhibited by lycopene pretreatment. Furthermore, lycopene pretreatment decreased the expression of cleaved poly (ADP ribose) polymerase-1 (PARP-1) and caspase-3 and increased the expression of B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X protein (Bax), which were induced by H2O2 treatment. Moreover, lycopene significantly increased manganese superoxide dismutase (MnSOD) expression and decreased cellular ROS levels via the PI3K-Akt pathway. Our findings show that lycopene pretreatment prevents ischemic injury by suppressing apoptosis-associated signal pathway and enhancing anti-oxidant protein, suggesting that lycopene could be developed as a beneficial broad-spectrum agent for the successful MSC transplantation in ischemic diseases.

Vol.23 No.6

Electrically conductive polypyrrole-polyvinylpyrrolidone (PPy-PVP) nanofiber mats with a core-shell structure have been successfully fabricated by a two-step process: the formation of FeCl3-containing PVP nanofiber mat by electrospinning, and the vapor-phase polymerization (VPP) of pyrrole monomer on the mat in a sealed chamber at room temperature. Surface morphology and chemical composition of the PPy-PVP mat were characterized by SEM, EDX and FTIR analyses. The as-prepared nonwoven mat was composed of PPy-PVP nanofibers with an average diameter of 300 nm. The sheet conductivity of the nanofiber mat was measured to be approximately 0.01 S/cm by a four-point probe. We have also investigated gas-sensing properties of PPy-PVP nanofiber mat upon exposure to methanol vapor. The PPy-PVP nanofiber sensors were observed to have excellent methanol-sensing performance. The nanofiberbased core-shell nanostructure could give an opportunity to fabricate a highly sensitive and fast response sensor due to its high surfaceto-volume ratio.

Effect of Prunella vulgaris Labiatae Extract on Innate Immune Cells and Anti-metastatic Effect in Mice

        Jun Beom Lee,Tae Bong Kang,Sang-Hoon Choi,Uiyoung Lee,Ae-Jung Kim,Chang-Jin Jeong,Hak-Cheon Lee,Yong-Sun Cho,Jong-Gun Won,Jong-Cheol Lim,Taek Joon Yoon 한국식품과학회 2009 Food Science and Biotechnology Vol.18 No.1

Ability of water extract from Prunella vulgaris Labiatae to stimulate immune system and inhibit tumor metastasis in mice was assessed. In experimental lung metastasis, prophylactic intravenous (i.v.) administration of water extract from P. vulgaris significantly inhibited lung metastasis in a dose-dependant manner. Peritoneal macrophages stimulated with P. vulgaris produced various cytokines such as tumor necrosis factor (TNF)-α and interlukin (IL)-12 as well as induced tumoricidal activity. In an assay for natural killer (NK) cell activity, i.v. administration of P. vulgaris significantly augmented NK cytotoxicity. The depletion of NK cells by injection of rabbit anti-asialo GM1 serum abolished the inhibitory effect of P. vulgaris on lung metastasis of colon26-M3.1 cells. These data demonstrate that P. vulgaris activate innate immune system to inhibit the growth of foreign materials including tumor cells in mice.

Brachymetacarpia and Brachymetatarsia in Patients with Multiple Hereditary Exostosis

Yong Jin Cho,Jun Young Lee,Won Gyun Lee,Sung Taek Jung 대한정형외과학회 2020 Clinics in Orthopedic Surgery Vol.12 No.4

Background: Multiple hereditary exostosis is a common autosomal dominant inherited musculoskeletal disorder that manifests with multiple osteochondromas. The clinical manifestations and pathological characteristics of osteochondromas found in the long bone and genetic alterations related to multiple hereditary exostosis have been widely reported. In this study, we investigated the characteristics of brachymetacarpia and brachymetatarsia associated with multiple hereditary exostosis. Methods: Of the 133 patients with a diagnosis of multiple hereditary exostosis who were recruited from 2005 to 2018, 101 patients who underwent plain radiography after 10 years of age were included. There were 55 male (54.5%) and 46 female (45.5%) patients. Brachymetacarpia or brachymetatarsia was diagnosed when disruption of the Lièvre parabola connecting the metacarpal or metatarsal heads was observed on plain radiographs. Three orthopedic surgeons individually reviewed hand and foot plain radiographs. Results: Of the 101 patients, 41 patients (40.6%) had more than 1 brachymetacarpia (88 cases) or brachymetatarsia (81 cases). Among 41 cases, 22 (53.7%) were male and 19 (46.3%) were female. The mean age at the time of radiographic evaluation of the hands and feet was 14.6 years (range, 10–63 years). Shortening was most commonly found in the 3rd and 4th metacarpal or metatarsal bones. Conclusions: We found a relatively high incidence of brachymetacarpia and brachymetatarsia in our patients. Physicians should suspect the presence of brachymetacarpia and brachymetatarsia when treating patients with multiple hereditary exostosis.

Current Management Status of Mercury Emissions from Coal Combustion Facilities

        Sung Jun Lee,Deepak Pudasainee,Yong-Chil Seo 한국대기환경학회 2008 한국대기환경학회지 Vol.24 No.E1

Mercury (Hg), which is mainly emitted from coal-fired power plants, remains one of the most toxic compounds to both humans and ecosystems. Hg pollution is not a local or regional issue, but a global issue. Hg compounds emitted from anthropogenic sources such as coal-fired power plants, incinerators, and boilers, can be transported over long distances. Since the last decade, many European countries, Canada, and especially the United States, have focused on technology to control Hg emissions. Korea has also recently showed an interest in managing Hg pollution from various combustion sources. Previous studies indicate that coal-fired power plants are one of the major sources of Hg in Korea. However, lack of Hg emission data and feasible emission controls have been major obstacles in Hg study. In order to achieve effective Hg control, understanding the characteristics of current Hg sampling methods and control technologies is essential. There is no one proven technology that fits all Hg emission sources, because Hg emission and control efficiency depend on fuel type, configuration of air pollution control devices, flue gas composition, among others. Therefore, a broad knowledge of Hg sampling and control technologies is necessary to select the most suitable method for each Hg-emitting source. In this paper, various Hg sampling methods, including wet chemistry, dry sorbents trap, field, and laboratory demonstrated control technologies, and international regulations, are introduced, with a focus on coal-fired power plants.

Do Clinical Manifestations Influence the Efficiency of Computed Tomography and Ultrasonography in Diagnosing Acute Appendicitis?

( Sung Bin Chon ),( Won Sup Oh ),( Seung Joon Lee ),( Yong Hwan Jeon ),( Chan Woo Park ),( Joong Bum Moon ),( Jun Hwi Cho ) 대한응급의학회 2011 대한응급의학회지 Vol.22 No.4

Purpose: Radiological readings of possible or suspicious appendicitis are often unhelpful for clinicians, and normal readings may be misleading if the patient has appendicitis. We conducted a retrospective study to determine whether the efficiency of computed tomography (CT) or ultrasonography (USG) in diagnosing acute appendicitis is altered by clinical manifestations. Methods: Data were collected from all the pathologicallyproven acute appendicitis patients who underwent CT or USG and subsequently had appendectomy between January 2009 and March 2010 at Kangwon National University Hospital. We classified radiological findings of compatible with and probable appendicitis as highly efficient and possible or suspicious appendicitis and normal appendix as less efficient. The following clinical manifestations were included: duration of the chief complaint prior to performing CT or USG, right lower quadrant pain, pain migration, body temperature, abdominal tenderness, rebound tenderness, muscle guarding, wall rigidity, white blood cell count, percentage of polymorphonuclear cells, and C-reactive protein level. Results: A total of 202 patients underwent appendectomies after imaging studies (37.2±20.3-years-of-age; male-tofemale ratio, 1.08). Of these, 154(76.2%) received CT, 44 (21.8%) received USG, and four (2.0%) received magnetic resonance imaging. Radiological findings were highly efficient in 155 cases (76.7%) but less efficient in 47 cases (23.3%). Multiple logistic regression analysis demonstrated that the absence of pain migration was related to less efficient results (adjusted odds ratio, 3.26; 95% confidence interval, 1.16-9.12). Conclusion: The ``efficient`` sensitivity of CT or USG in diagnosing acute appendicitis is low in the absence of a history of pain migration.

Effect of Process Condition on Tensile Properties of Carbon Fiber

        Sung-Ho Lee,Ji-Hoon Kim,Bon-Cheol Ku,Jun-Kyong Kim,Yong-Sik Chung 한국탄소학회 2011 Carbon Letters Vol.12 No.1

For polyacrylonitrile (PAN) based carbon fiber (CF) process, we developed a lab scale wet spinning line and a continuous tailor-made stabilization system with ten columns for controlling temperature profile. PAN precursor was spun with a different spinning rate. PAN spun fibers were stabilized with a total duration of 45 to 110 min at a given temperature profile. Furthermore, a stabilization temperature profile was varied with the last column temperature from 230 to 275℃. Stabilized fibers were carbonized in nitrogen atmosphere at 1200℃ in a furnace. Morphologies of spun and CFs were observed using optical and scanning electron microscopy, respectively. Tensile properties of resulting CFs were measured. The results revealed that process conditions such as spinning rate, stabilization time, and temperature profile affect microstructure and tensile properties of CFs significantly.
