The western flower thrips, Frankliniella occidentalis, is the most economically important agronomic pest withinThysanoptera because it is both a direct pest of horticulture crops and an efficient vector of plant viruses. Sixty-seven polymorphic SSR loci were identified in the contigs (containing redundant ESTs) generated byassembling 13,839 F. occidentalis ESTs from the public sequence database. Nineteen SSR markers exhibited polymorphismamong 860 samples from43 F. occidentalis populations,with alleles per SSR marker ranging fromtwoto eight, the effective number of alleles (Ne) range from 0.73 to 2.64; the observed (Ho) and expected (He) heterozygositiesranged from 0.09 to 0.77 and 0.12 to 0.96, respectively. The PIC values were from 0.24 to 0.73. AMOVA revealed most genetic variation resided within, rather than between, greenhouse and field isolates. The Mantel test showed no significant differences between genetic and geographical distances. We demonstratedthe value of mining the redundant sequences in public sequence databases for the development of polymorphicSSR markers, which can be used for better understanding population variation and spreading of the invasive pestF. occidentalis.

To clarify the effects of sublethal concentration of insecticide on the growth, development and reproduction of Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), the sublethal and transgenerational effects of the LC 30 of six insecticides on adults and progeny (F 1 and F 2 generations) were studied using age-stage, two-sex life tables. Exposure to six insecticides had significant negative impacts on the life-history traits of B. dorsalis and led to reduced adult longevity and fecundity in F 0 , a longer larval stage duration in F 1 , and reduced fertility and survival. The intrinsic rate of increase (r), finite rate of increase (λ), and net reproductive rate (R 0 ) were reduced in F 1 , with the greatest reduction observed in the chlorpyrifos treatment. In the F 2 generation, the adverse effects were weakened, only the cyantraniliprole treatment had lower values than the control, and the fecundity of other treatments returned to normal. Obvious “hormesis” phenomena were not observed in any of the generations. Based on the population hazard index, chlorpyrifos had the best control effect in the early stage, and cyan traniliprole had the best control effect in the long term. These results indicate that the LC 30 of the six insecticides exert different transgenerational effects on the B. dorsalis population.

Objective: Jining Grey goat is a local Chinese goat breed that is well known for its high fertility and excellent meat quality but shows low meat production performance. Numerous studies have focused on revealing the genetic mechanism of its high fertility, but its highlighting meat quality and muscle growth mechanism still need to be studied. Methods: In this research, an integrative analysis of the genomics and transcriptomics of Jining Grey goats compared with Boer goats was performed to identify candidate genes and pathways related to the mechanisms of meat quality and muscle development. Results: Our results overlap among five genes (ABHD2, FN1, PGM2L1, PRKAG3, RAVER2) and detected a set of candidate genes associated with fatty acid metabolism (PRKAG3, HADHB, FASN, ACADM), amino acid metabolism (KMT2C, PLOD3, NSD2, SETDB1, STT3B, MAN1A2, BCKDHB, NAT8L, P4HA3) and muscle development (MSTN, PPARGC1A, ANKRD2). Several pathways have also been detected, such as the FoxO signaling pathway and Apelin signaling pathway that play roles in lipid metabolism, lysine degradation, N-glycan biosynthesis, valine, leucine and isoleucine degradation that involving with amino acid metabolism. Conclusion: The comparative genomic and transcriptomic analysis of Jining Grey goat and Boer goat revealed the mechanisms underlying the meat quality and meat productive performance of goats. These results provide valuable information for future breeding of goats.

Root-knot nematode disease is a widespread and catastrophic disease of tobacco. However, little is known about the relationship between rhizosphere bacterial community and root-knot nematode disease. This study used 16S rRNA gene sequencing and PICRUSt to assess bacterial community structure and function changes in rhizosphere soil from Meloidogyne incognita-infected tobacco plants. We studied the rhizosphere bacterial community structure of M. incognita-infected and uninfected tobacco plants through a paired comparison design in two regions of tobacco planting area, Yuxi and Jiuxiang of Yunnan Province, southwest China. According to the findings, M. incognita infection can alter the bacterial population in the soil. Uninfested soil has more operational taxonomic unit numbers and richness than infested soil. Principal Coordinate Analysis revealed clear separations between bacterial communities from infested and uninfested soil, indicating that different infection conditions resulted in significantly different bacterial community structures in soils. Firmicutes was prevalent in infested soil, but Chloroflexi and Acidobacteria were prevalent in uninfested soil. Sphingomonas, Streptomyces, and Bradyrhizobium were the dominant bacteria genera, and their abundance were higher in infested soil. By PICRUSt analysis, some metabolism-related functions and signal transduction functions of the rhizosphere bacterial community in the M. incognita infection-tobacco plants had a higher relative abundance than those uninfected. As a result, rhizosphere soils from tobacco plants infected with M. incognita showed considerable bacterial community structure and function alterations.

A facile and versatile route to prepare porous polymer microparticles with tunable pore size and density through the combination of phase separation and emulsion-solvent evaporation method is demonstrated. When volatile organic solvent (e.g., chloroform) diffuses through the aqueous phase containing poly(vinyl alcohol) (PVA) and evaporates, n-hexadecane (HD) and polystyrene (PS) in oil-in-water emulsion droplets occur to phase separate due to the incompatibility between PS and HD, ultimately yielding microparticles with porous structures. Interestingly, density of the pores (pore number) on the shell of microparticles can be tailored from one to hundreds by simply varying the HD concentration and/or the rate of solvent evaporation. Moreover, this versatile approach for preparing porous microparticles with tunable pore size and density can be applied to other types of hydrophobic polymers, organic solvents, and alkanes, which will find potential applications in the fields of pharmaceutical, catalyst carrier, separation, and diagnostics.

Objective: More than 75% of ovarian cancer patients are diagnosed at advanced stages and die of tumor cell metastasis. This study aimed to identify new epigenetic and transcriptomic alterations associated with ovarian cancer metastasis. Methods: Two cell sublines with low- and high-metastasis potentials were derived from the ovarian cancer cell line A2780. Genome-wide DNA methylome and transcriptome profiling were carried out in these two sublines by Reduced Representation Bisulfite Sequencing and RNA-seq technologies. Cell-based assays were conducted to support the clinical findings. Results: There are distinct DNA methylation and gene expression patterns between the two cell sublines with low- and high-metastasis potentials. Integrated analysis identified 33 methylation-induced genes potentially involved in ovarian cancer metastasis. The DNA methylation patterns of two of them (i.e., SFRP1 and LIPG) were further validated in human specimens, indicating that they were hypermethylated and downregulated in peritoneal metastatic ovarian carcinoma compared to primary ovarian carcinoma. Patients with lower SFRP1 and LIPG expression tend to have a worse prognosis. Functionally, knockdown of SFRP1 and LIPG promoted cell growth and migration, whereas their overexpression resulted in the opposite effects. In particular, knockdown of SFRP1 could phosphorylate GSK3β and increase β-catenin expression, leading to deregulated activation of the Wnt/β-catenin signaling. Conclusion: Many systemic and important epigenetic and transcriptomic alterations occur in the progression of ovarian cancer. In particular, epigenetic silencing of SFRP1 and LIPG is a potential driver event in ovarian cancer metastasis. They can be used as prognostic biomarkers and therapeutic targets for ovarian cancer patients.
Mechanical behavior of composite gel periodic structures with the pattern transformation

When the periodic cellular structure is loaded or swelling beyond the critical value, the structure may undergo a pattern transformation owing to the local elastic instabilities, thus leading to structural collapse and the structure changing to a new configuration. Based on this deformation-triggered pattern, we have proposed the novel composite gel materials. This designed material is a type of architectural material possessing special mechanical properties. In this study, the mechanical behavior of the composite gel periodic structure with various gel inclusions is studied further through numerical simulations. When pattern transformation occurs, it results in a different elastic relationship compared with the material at untransformed state. Based on the obtained nominal stress versus nominal strain behavior, the Poisson‟s ratio and corresponding deformed structure patterns, we investigate the performance of designed composite materials and the effects of the uniformly distributed gel inclusions on composite materials. A better understanding of the characteristics of these composite gel materials is a key to develop its potential applications on new soft machines.
