        비만 개선 효과를 지닌 탄수화물 및 지방 흡수 억제 기능성 식이조성물 개발

        In a previous study, we developed a new food additive as an egg yolk antibody (IgY) against carbohydrate digestion enzymes for the regulation of blood glucose level and weight control. The IgY delayed and decreased the increment of blood glucose level after administration of sucrose in human being by 30% in 20∼30 min. We also developed a lipase inhibitor as a water extract of two kinds of herb, Platycodon grandiflorum and Solanum melongena. Twenty three volunteers were subjected to the intake of the egg yolk IgY plus the herbal extracts for 50 days. In average, the treated subjects appeared to lose 1.96 ㎏ of body weight and 3.4 ㎏ of body fat mass during the treated period. Furthermore, Panniculus adiposus and breech size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of egg yolk IgY and natural herbal extract improve the obesity by the decrement of body weight and body fat mass.

      • A Study on Dissolution of Chitin

        Several kinds of solvent system were applied to dissolve chitin which acknowledged as a raw material for biomedical application. Solvent systems were N-methylmorpholineoxide(NMMO) or mixture with water or mixtrure with dimethylsulfoxide(DMSO) or mixture with acetonitrile and mixture of trichloroacetic acid(TCA) with methylene chloride(MC). Among them mixture of TCA with MC appears the best solvent system for chitin. Chitin in this solvent showed clear sloution after few hours of dissolution.

        Glucose oxidase, alcohol oxidase and pyruvate oxidase have immobilized on oxygen transfer membranes of dissolved oxygen electrode for biosensor. 4% cellulose triacetate/polycaprolactone : 80/20(w/w) membrane by one-step aminated CDI method has effective immobilization ratio of 2.7 units/30㎟ glucose oxidase, 11.1units/30㎟ alcohol oxidase and 3.1 units/30㎟ pyruvate oxidase respectively, but polycarbonate has too low immobilization ratio of each enzymes. To improve of immobilized enzyme activity reacted with carbodiimide for spacer arm, which give mobile activity between enzyme and membrane, increased ca. 10% of enzymes activities. The storage activity of immobilized glucose oxidase remains 71% after 2 weeks in 4℃, and alcohol oxidase and pyruvate oxidase have under 40% after 8 days.

      • 식품의 치아우식유발력 측정방법에 관한 연구

        The acidogenic potential of a group of popular drink foods and total carbohydrate content of salivary expectorants following their consumption were assessed using an model of oral cavity with micro glass electrode on plaque from Steptococcos mutans. The standard 10% sucrose solution had decreased to pH4.2 after 60 minute by this method. The results suggested that test method of modulator for cariogenic potential was useful for determination of cariogenicity of foods.

      • 소나무잔나비버섯의 구강병원인균에 대한 항균효과

        Formitopsis pinicola has been traditionally used for medicine. The hot water extracts of Formitopsis pinicola has been used for reduce diabetes and inflammation. Therefore, the aim of this study was to evaluate in vitro the antimicrobial activity against some microorganisms for oral diseases. The fruiting body of Formitopsis pinicola were harvested in Nepal. The methanol extract Formitopsis pinicola(Fp-M) were evaporated and dissolved in dimethyl sulfoxide by 100㎎/㎖. Antimicrobial activity was carried out by thc minimal inhibitory concentrations (MICs) and short-term exposure studies with S. mutans (ATCC 25175). S. sanguis (ATCC 10556). A. viscoccus (ATCC 15987) C. albicans (ATCC 10231). The Fp-M was showing MICs ranging from 0.029(㎎/㎖) against S. sanguis and 0.05(㎎/㎖) against S. mutans 1.0 against C. albicans and 2.0 against A. viscosun respectively. 5-minute exposure of S. mutans, S. sanguis, A. viscoccus, and C. albicans to Fp-M (100㎎/㎖) resulted in a substantial delay at 2-16h in growth of oral microorganism. Fp-M exhibited potent antimicrobial activity against all of the studied microorganisms. It was suggested that F. pinicola (AY 854083) may be a valuable antimicrobial agent to prevent dental caries and periodontal disease.

        The purpose of this study was development of feed additives as antibiotic effect for poultry and swine. The fermentation method for feed additives was with Lactobacillus plantarum and Bacillus subtillus with curcumin, capsaicin and apple debris to reducing strong flavour. This additives was increasing of 250% antioxidant value and reducing death rate on poultry from 9.7% to 2.2% on field test.

      • Auricularia auricula 추출물의 streptococcus mutans에 대한 항균효과

        The purpose of this study was to development of dental hygiene product with extracts of Auricularia auricula fruiting body. The extracts Auricularia auricula were evaporated and dissolved in dimethyl sulfoxide by 100 ㎎/㎖. Antimicrobial activity was carried out with Streptococcus mutans (ATCC 25175). The ethyl ether extract from Auricularia auricula would be a useful for development of dental hygiene product. The effect of extract of Auricularia auricula fruiting body on Streptococcus mutans was as MIC 0.33 mg from diethyl ether extracts of 1 g mushroom, and Aal fraction after purified by TLC was identified in two bands from 4 bands and were partially analysed by HPLC.

        Chitin is a complex polysaccaride which is the potential biodegradable biopolymer with characteristics of biocompatability and blood compatability, and it has a very good potential for a lot of medical applications. In this study, a colorless and transparent CHITULOSE FILM which made from the mixture of chitin and cellulose was used. When the degrees of diffusion of water and organic compounds were tested, those of cellulose film were lower compared to those of film containing chitin. The degree of diffusion of CHITULOSE FILM showed high values for ionic organic compounds compared to nonionic organic compounds.

        In the korean soybean seedlings O-methyltranferase and ononitol epimerase lead to accumulation of methylated inositols, D-ononiton and D-pinitol, while serve as osmorprotectants. This paper describes analysis for myo-inositol, D-ononitol, D-pinitol and glucose by TLC for metabolites in salt stressed soybean seedlings. The crude enzymes, myo-inosito-6-O-methyltranferase and ononitol epimerase, extract from soybean seedlings, which analysis by DEAE-cellulose column. This enzymes react with glucose to produce polyol metabolites.

        Streptomyces sp. Y9343이 生産하는 齒面細菌膜 分解酵素의 精製와 特性

        치면세균막을 제거하거나 형성을 억제하여 치아우식증 예방제를 개발할 목적으로 α-1,3 glucanase를 분비하는 새로운 균주로 부터 분리 정제하여 그 특성을 조사하였다. 불용성 glucan을 유일탄소원으로 하는 agar plate를 제조하여 토양으로 부터 α-1,3 glucanase 분비균주를 탐색한 결과 Streptomyces sp. Y9343을 얻었다. 액체배양시 최적 효소생산조건은 탄소원으로 1% soluble starch와 indcer로 0.5% insoluble glucan을 첨가하였을 때 가장 효율적이었다. α-1,3 glucanase는 황산 암모늄 염석, DEAE-CEllulose 이온교환 크로마토그래피, Sephadex G-75 겔 여과 등에 의하여 32.1배까지 정제되었고 수율은 0.53%이었으며, 이 때의 활성도는 7840.9 U/mg protein이었다. 정제된 α-1,3 glucanase를 SDS-PAGE로 분석한 결과, 단일체임을 확인하였으며, 이 때 분자량은 22,500이었다. 효소의 최적 pH는 6.5이었다. 효소의 최저온도는 37℃이었고, 열에 대한 안정성은 70℃ 이상에서 40%~60%의 효소활성이 상실함을 보였다. Detergent의 영향은 SDS에 의해 83%, Tween 20에 의해서는 약 27% 정도의 활성저해를 받았다. 효소활성의 금속이온에 의한 영향은 Co^2+, Mn^2_+에 의해 각각 81.8, 69.7%의 활성의 증가를 보였고 이들의 최적농도는 10mM이었으며, 반면에 Hg^2+에 의해서는 93.9%의 효소활성의 저해를 나타내었다. 또한 초기속도(30분 이내)에 금속이온에 의한 영향이 큰 것으로 나타났다. α-1,3 glucanase의 불용성 glucan에 대한 K_m 값은 2.50mM이었고, V_max는 0.0431 mM/min이었다. α-1,3 glucanase의 기질특이성을 조사한 결과, 반응 30분 후 IG와 soluble starch에는 각각73, 100%의 높은 분해력을 보였으며, raw starch, dextran T-10에 대해서는 낮은 분해력을 보였다. 한편, 인조치면세균막을 S. mutans로부터 시험관 벽에 제조한 후, α-1,3 glucanase를 처리한 결과 2시간 이내에 완전히 분해 제거되는 것을 알 수 있어 강력한 치아우식예방제로 개발될 수 있음을 보여 주었다. Streptococcus mutans has been implicated as primary causative agents of dental caries by insoluble glucan (IG) in human and experimental animals. An attempt was made to search for the α-1,3 glucanase that degrades IG produced by S. mutans. α-1,3 glucanase was detected in the culture supernatant of microorganisms, which are isolated from soils on agar medium containing IG as a sole carbon source. This Streptomyces sp. hydrolysed IG produced by immobilized S. mutans and was named as Y9343. This enzyme required α-1,3 glucan (IG) as an inducer. The optimum conditions for enzyme production were studied. The enzyme was purified by 30~70% (NH_4)_2SO_4 precipitation, anion exchange chromatography on DEAE-cellulose and gel filration on Sepadex G-75. The purified enzyme has a specific activity of 7840.0 U/mg protein giving 32.1-fold purification and final yield of 0.53%. The molecular weight was estimated to be about 22.5 kDa by SDS-PAGE. The optimum pH and temperature for enzyme reactiorr were 6.5 and 37℃, respectively and the enzyme was relatively stable at the temperature below 60℃. The activity of purified enzyme was enhanced by adding Co^2+, Mn^2+, and Mg^2+ into the medium, whereas inhiited by adding Hg^2+, Zn^2+ and SDS. The K_m and V_max value of α-1,3 glucanase for IG were estimated to be 2.50 mM and 0.0431 mM/min, respectively. The thin layer chromatographic analysis of hydrolysates from IG with α-1,3 glucanase showed that glucose was the main product of reaction. This enzyme activity was about 14 times higher than marketing dextranase as preventive agent against artificial dental caries by S. mutans in TH medium including 5% sucrose after 30 minutes.
