Petroleum-degrading bacteria were enumerated from the coastal water near Samcheonpo City and Cheonsu Bay with the Most-Probable-Number method. Number of petroleum-degrading bacteria fluctuated from 2.0×10 exp (2) to 1.1×10 exp (5) cells/litre in the coastal water near Samcheonpo City, and 3.6×10 exp (1) to 1.1×10 exp (4) cells/litre in Cheonsu Bay. The proportions of petroleum-degrading bacteria to total viable counts were in the range of 2.1% to 0.00012%. In both study areas, ratio of petroleum-degrading bacteria at autumn showed the maximal value. Enterobacteriaceae, Vibrio, Bacillus, Pseudomonas, Flavobacterium, Acinetobacter, Micrococcus, Aerococcus and Aeromonas were isolated. Enterobacteriaceae, Vibrio and Pseudomonas were found dominantly. The observed results may be provided for understanding the level of potentiality of microbial petroleum-degradation in the research area.
다양한 당-아미노산 Model system에서의 Maillard Browning 반응 생성물의 항산화 효과

This study was conducted to observe the effect of various kinds of sugars (galactose, fructose, glucose, lactose, and sucrose) and amino acids (glycine, β-alanine, L-valine, L-tyrosin, L-aspartic acid, and L-glutamic acid ) on the Maillard reaction. Model systems were prepared at 0.1 M concentration and heated at 90 ℃ for 2 hour. The raction was evaluated by absorbance at 420 nm for browning and 517 nm for DPPH radical-scavenging activity. Model systems(1) derived from galactose showed a greater increase in browning intensity than others. The reactivity of reducing sugar and amino acid was increased in the following order: galactose > fructose > glucose > lactose > sucrose, Glycine > β-alanine > L-valine > L-tryrosin > L-aspartic acid > L-glutamic acid. The pH of Model systems(2) decreased gradually as the heating time increased up to 180 min. Melanoidin Model systems(2) derived from galactose showed the highest radical-scavenging activity. DPPH radical-scavenging activity of melanoidins increased as the heating time increased.
제주도 숨은물벵뒤 습지 서식 Alphaproteobacteria 및 Gammaproteobacteria 강에 속하는 신변이주의 특성

In this study, samples were collected from the Sumummulbangdui wetland at the Halla Mountain in Jeju Island in order to isolate novel bacterial strain. Bacterial strains belonging to the class Alphaproteobacteria, and Gammaproteobacteria were isolated after spreading samples onto solid agar media. The 16S rRNA gene sequences of the strains assigned to the two classes were compared to those of type strains of the species. The strains that showed less than 98.7% 16S rRNA gene sequence similarity to the validly published species were considered to be novel species candidates. A total of 19 strains were regarded as novel strains which can be regarded as novel species candidates. In the Alphaproteobacteria, 6 novel strains were affiliated with the genera Novosphingobium, and Rhizobium. A total of 13 novel strains belong to Gammaproteobacteria that assigned to the family Moraxellaceae, Pseudomonadaceae, and Enterobacteriaceae were identified. Cultural, physiological, chemotaxonomic characteristics and fatty acids compositions have been determined for the novel species candidates, and the characteristics are described in this study.

Although Sumunmulbangdui wetland at the Halla Mountain in Jeju Island, a kind of montane wetlands, has been considered to bear high biodiversity, no study has been reported on the bacterial diversity. In this study, soil and water samples were collected from the wetland in order to isolate novel bacterial species. Bacterial strains belonging to the phyla Bacteroidetes, Firmicutes, and Actinobacteria were isolated after spreading soil and water samples onto solid agar media. The 16S rRNA gene sequences of the strains assigned to the three phyla were compared to those of type strains of the species in the phyla. The strains that showed less than 98.7% 16S rRNA gene sequence similarity to the validly published species were considered to be novel species candidates. A total of 32 strains were regarded as novel species candidates in the phyla Bacteroidetes, Firmicutes, and Actinobacteria. Diversity of novel species candidates was very low; the candidates were confined to only few genera. In the Bacteroidetes, 13 novel candidate species were affiliated with the genera Mucilaginibacter, Sphingobacterium, Pedobacter, Flavobacterium, and Chryseobacterium. A total of 13 novel candidate species that assigned to the genera Paenibacillus, Lysinibacillus, and Bacillus were identified in the phylum Firmicutes. Only two candidate species that belonged to the genera Mycobacterium and Nocardia were excavated in the Actinobacteria. Cultural, physiological, and chemotaxonomic characteristics have been determined for the novel species candidates, and the characteristics are described in this study.

효모의 mating과정의 기작을 연구하기 위하여, α pheromone의 수용체를 coding하는 유전자인 STE2가 결여된 균주로 하여금 mating을 할 수 있게 하는 61개의 돌연변이를 획득하였다. 이들은 상보성시험과 연관시험 결과 3개 군의 유전자에 속했는데, 3개 군을 CDC70, CDC72 및 CDC73 이라고 명명하였다. 얻어진 돌연변이는 제한 온도에서는 세포분열이 start 단계에서 정지되는 온도 감수성이며 STE2 유전자 결여를 억제하는 능력을 보여 주었는데, 이는 유전자 CDC70, CDC72, CDC73 의 생성물이 수용체로부터 전달되는 signal의 중간 전달 물질이라는 사실을 가리킨다. In order to elucidate and characterize the signal transduction pathway(s) whereby yeast cells respond to mating pheromone, we have isolated mutants which are able to conjugate in the absence of the alpha-factor receptor. Sixty-one suppressors of a ste2-deletion mutation which also confer a ts conditional "start" arrest phenotype have been subjected to genetic analysis. The mutants could be assigned to three complementation groups designated CDC70, CDC72 and CDC73, which are unlinked to each other as well as to the previously identified start genes. Quantitation of mating ability of the cdc70, cdc72 and cdc73 mutations in a ste2-deletion background gives levels ranging from 0.1% to 0.3% of wild type, depending on the allele and the gene. The results indicate that the signals from mating pheromone might be mediated by the CDC70, CDC72 and CDC73 products.
