        New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.

      • Molecular and structural characteristics of multidrug resistance-associated protein 7 in Chinese liver fluke Clonorchis sinensis

        <P>Multidrug resistance-associated protein 7 (MRP7, ABCC10) is a C subfamily member of the ATP-binding cassette (ABC) superfamily. MRP7 is a lipophilic anion transporter that pumps endogenous and xenobiotic substrates from the cytoplasm to the extracellular milieu. Here, we cloned and characterized CsMRP7 as a novel ABC transporter from the Chinese liver fluke, Clonorchis sinensis. Full-length cDNA of CsMRP7 was 5174 nt, encoded 1636 amino acids (aa), and harbored a 147-bp 5'-untranslated region (5'-UTR) and 116-bp 3'-UTR. Phylogenetic analysis confirmed that CsMRP7 was closer to the ABCC subfamily than the ABCB subfamily. Tertiary structures of the N-terminal region (1-322 aa) and core region (323-1621 aa) of CsMRP7 were generated by homology modeling using glucagon receptor (PDB ID: 5ee7_A) and P-glycoprotein (PDB ID: 4f4c_A) as templates, respectively. CsMRP7 nucleotide-binding domain 2 (NBD2) was conserved more than NBD1, which was the sites of ATP binding and hydrolysis. Like typical long MRPs, CsMRP7 has an additional membrane-spanning domain 0 (MSD0) and cytoplasmic loop, along with a common structural fold consisting of MSD1-NBD1-MSD2-NBD2 as a single polypeptide assembly. MSD0, MSD1, and MSD2 consisted of TM1-7, TM8-13, and TM14-19, respectively. The CsMRP7 transcript was more abundant in the metacercariae than in the adult worms. Truncated NBD1 (39 kDa) and NBD2 (44 kDa) were produced in bacteria and mouse immune sera were raised. CsMRP7 was localized in the apical side of the intestinal epithelium, sperm in the testes and seminal receptacle, receptacle membrane, and mesenchymal tissue around intestine in the adult worm. These results provide molecular information and insights into structural and functional characteristics of CsMRP7 and homologs of flukes.</P>

        R&D Status of High-current Accelerators at IFP

        High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.

      • Synthesis, characterization of the phenylquinoline-based on iridium(III) complexes for solution processable phosphorescent organic light-emitting diodes

        A new series of highly efficient Ir(III) complexes, (DPQ)<SUB>2</SUB>Ir(pic-N-O), (F<SUB>4</SUB>PPQ)<SUB>2</SUB>Ir(pic-N-O), (FPQ)<SUB>2</SUB>Ir(pic-N-O), and (CPQ)<SUB>2</SUB>Ir(pic-N-O) were synthesized for phosphorescent organic light-emitting diodes (PhOLEDs), and their photophysical, electrochemical, and electroluminescent (EL) properties were investigated. The Ir(III) complexes, including picolinic acid N-oxide (pic-N-O) ancillary ligand, are comprised with the various main ligands such as 2,4-diphenylquinoline (DPQ), 4-phenyl-2-(2,3,4,5-tetrafluorophenyl)quinoline (F<SUB>4</SUB>PPQ), 2-(9,9-diethyl-9H-fluoren-2-yl)-4-phenylquinoline (FPQ) and 9-ethyl-3-(4-phenylquinolin-2-yl)-9H-carbazole. Remarkably, high performance PhOLEDs using a solution-processable (DPQ)<SUB>2</SUB>Ir(pic-N-O) doped CBP host emission layer were fabricated to give a high luminance efficiency (LE) of 26.9cd/A, equivalent to an external quantum efficiency (EQE) of 14.2%. The calculated HOMO-LUMO energy gaps for (DPQ)<SUB>2</SUB>Ir(pic-N-O), (F<SUB>4</SUB>PPQ)<SUB>2</SUB>Ir(pic-N-O), (FPQ)<SUB>2</SUB>Ir(pic-N-O) and (CPQ)<SUB>2</SUB>Ir(pic-N-O) were in good agreement with the experimental results.

        Sustainable Development and Energy Geotechnology− Potential Roles for Geotechnical Engineering

        The world is facing unprecedented challenges related to energy resources, global climate change, material use, and waste generation. Failure to address these challenges will inhibit the growth of the developing world and will negatively impact the standard of living and security of future generations in all nations. The solutions to these challenges will require multidisciplinary research across the social and physical sciences and engineering. Although perhaps not always recognized, geotechnical engineering expertise is critical to the solution of many energy and sustainability-related problems. Hence, geotechnical engineers and academicians have opportunity and responsibility to contribute to the solution of these worldwide problems. Research will need to be extended to non-standard issues such as thermal properties of soils; sediment and rock response to extreme conditions and at very long time scales; coupled hydro-chemo-thermo-bio-mechanical processes; positive feedback systems; the development of discontinuities; biological modification of soil properties; spatial variability; and emergent phenomena. Clearly, the challenges facing geotechnical engineering in the future will require a much broader knowledge base than our traditional educational programs provide. The geotechnical engineering curricula, from undergraduate education through continuing professional education, must address the changing needs of a profession that will increasingly be engaged in alternative/renewable energy production; energy efficiency; sustainable design, enhanced and more efficient use of natural resources, waste management, and underground utilization.

      • Facile green synthesis of isoindigo-based conjugated polymers using aldol polycondensation

        <P>The aldol reaction is a facile green synthetic method and has been widely used in the synthesis of small molecules. In this study, we attempted to prepare conjugated polymers using aldol polymerization for the first time. Two isoindigo-based conjugated polymers (PIID-DT and PBIBDF-DT) were synthesized successfully in excellent yields and with high molecular weights (MGPC n up to 53.9 kDa) using toluene as the solvent and an acid as the catalyst. The entire synthesis route is economical and environmentally friendly, and the traditional drawbacks such as the use of organometallic reagents, toxic tin monomers, and other environmentally harmful compounds often encountered in Stille and Suzuki cross-coupling reactions could be avoided. Moreover, the isoindigo-based polymers prepared by the newly established aldol polymerization were also evaluated as organic field-effect transistors with bottom-gate/ top-contact devices and exhibited excellent mobilities as high as 0.16 and 0.26 cm(2) V-1 s(-1) with a high I-on/I-off ratio of 10(5) for PIID-DT and PBIBDF-DT, respectively. Therefore, aldol polycondensation has environmentally friendly characteristics that can be applied in the green synthesis of isoindigo-based conjugated polymers.</P>

        Factors Affecting the Inosine Monophosphate Content of Muscles in Taihe Silkies Chickens

        In this paper, changes in the concentration of inosine monophosphate (IMP) of muscles in Taihe Silkies chickens from 2 to 28 weeks of age were studied. The results showed that: (1) with increasing age, IMP content of muscles decreased continuously. (2) the relationship between body weight and IMP content of musculus pectoralis major was significantly negative (p<0.01), so was body weight and IMP content of musculus peronaeus, and their coefficients of correlation were -0.45, -0.38 respectively; the relationship between IMP contents of musculus pectoralis major and that of musculus peronaeus was more significantly positive (p<0.01), and its coefficient of correlation was 0.59. (3) by multiple regression analysis, changes of IMP content of muscles depended on the weeks of age and body weight. (4) from 2 to 28 weeks of age, the heritabilities of IMP content of musculus pectoralis major were calculated between 0.33-0.48, and those of musculus peronaeus were calculated between 0.51-0.69.

        Genetic Analysis of Haimen Chicken Populations Using Decamer Random Markers

        Through a screening and selection approach method, decamer random markers were used in a technique called random amplified polymorphic DNA (RAPD) assay with 252 genomic DNAs isolated from four major Haimen chicken populations: Rugao (62), Jiangchun (62), Wan-Nan (63) and Cshiqishi (65). A total of 3-score decamer random primers (S241-S260, S1081-S1100 and S1341-S1360) were employed in the preliminary RAPD-polymerase chain reaction (RAPD-PCR) assay with 50 random template DNA samples from all the populations. Four (6.67%) of the primers that produced obvious polymorphic patterns, interpretable and reproducible bands were selected and used with both the individual DNAs from each population and with pooled DNA samples of the four populations in subsequent analyses. The selected primers produced a total of 131 fragments with molecular size ranging from 835 to 4,972 base pairs (bp) when used with the individual DNAs; 105 (80.15%) of these fragments were polymorphic. With the pooled DNAs, 47 stable and characteristic bands with molecular size ranging from 840 to 4,983 bp, of which 23 (48.94%) polymorphic, were also generated. The band-sharing coefficient (BSC) calculated for the individuals in the population and among populations of bulked samples was between 0.8247 (Rugao) and 0.9500 (Cshiqishi); for pairwise populations, it was between 0.7273 (Rugao vs. Wan-Nan) and 0.9367 (Jiangchun vs. Cshiqishi) chicken populations. Using the BSC for individual and pairwise populations, the Nei's standard genetic distances between the chicken populations were determined and ranged from 0.0043 (Jiangchun vs. Cshiqishi) to 0.1375 (Rugao vs. Cshiqishi). The reconstructed dendrogram linked the Jiangchun and Cshiqishi chickens as closely related populations, followed by Wan-Nan, while the Rugao was the most genetically distant among the populations.
