Resonance Raman Study on Authrornyces Ramosus Peroxidase and its CO adducts

We observed the v (FeCO) and v (CO) on ARPCO for the first time and δ (FeCO) was calculated using normal coordinate calculation under a bending angle of δ= 165°. The v (FeCO) and v (CO) on ARPCO was obtained at 519 cm^(-1) and 1928 cm^(-1) for the ^(12)C^(16)O derivative, and shifted to 516 cm^(-1) and 1884 cm^(-1) with ^(13)C^(16)O, and again to 510 cm^(-1) and 1885 cm^(-1) with ^(12)C^(18)O, and then to 506 cm^(-1) and 1840 cm^(-1) with ^(13)C^(18)O, respectively. Among various heme-CO, the v(CO) is the lowest value and v (FeCO) is the highest value. The CO-isotope-sensitive- band at 583 cm^(-1) was detected as seen for other heme protein-CO.

High-performance Raman/ Absorption Simultaneous Determination Technique for Enzymatic Reaction Intermediate, Compound Ⅱ

High-performance resonance Raman/ absorption simultaneous determination technique for enzymatic reaction intermediate, compound Ⅱ, was performed upon addition of hydrogen peroxide to ferric Arthrornyces Ramosus Peroxidase (ARP) using microcirculating system. The Fe=O stretching, v(Fe=O), frequency of compound Ⅱ was observed at 788 cm^(-1) for the v(Fe=^(16)O) mode of ARP Ⅱ and at 750 cm^(-1) for the v (Fe=^(18)O) mode (△v = 35 cm^(-1)). The v(Fe=O) value found in compound Ⅱ of ARP is coincidently same as compound Ⅱ of HRP in alkaline pH. Also, it is shown that a trans ligand effect [v(Fe-N_(his))] is one of the important factors to influence to the v(Fe=O) frequency of compound Ⅱ.

Fast mixing device를 이용한 Protein sample의 Time-Resolved Resonance Raman Spectroscopy

The application of fast mixing devices on the heme protein samples was described. Obviously, it is important to determine whether protein samples are stable under conditions encountered with new devices. Human Hemoglobin (Hb) solution was introduced into Platinum sphere mixing (PSM) system at 20 lb/in² gas pressure. The results were checked by UV-Vis spectroscopy and resonance Raman spectroscopy, and the spectra are superimposable upon those obtained from the initial Hb solution, respectively. The ligand binding reaction (oxygenation) of deoxy Hb was foIIowed by PSM. Time-resolved resonance Raman spectroscopy shows that the oxygenation reaction of Hb can be measured with PSM device for resonance Raman studies. Also, resonance Raman spectrum of product of the reaction between met Hb and HzOz supports that this Hb-H₂O₂ complex is ferryl Hb.

Serotonin에 대한 면역분석법

A variety of labels have been used in the development of immunoassay. Compared to the other detection system, bioluminescence offers numerous advantages, such as high sensitivity, stability, and low detection limit. A sensitive competitive heterogeneous bioluminescence immunoassay for serotonin measurements was developed using a photoprotein, aequorin as a label. Avidin/biotin mediated assay was also performed for signal amplification and activity conservation of aequorin. Serotonin-avidin conjugate was prepared by Mannich reaction and biotin-aequorin conjugate was prepared through NHS-biotin. The resulting dose-response curve showed that the detection range for serotonin was 10^(-6) to 10^(-10)M, and the detection limit was 7.9 x 10^(-10)M. Cross reactivity test indicated that serotonin cross-reacted with 5-methoxytryptamine (28%) but not with 3-methylindole, melatonin, nor with 5-hydroxyindole-3-acetic acid.

다염소 유기 오염물질의 탈염소화반응 - 1,2,4-트리클로로벤젠

환경 잔류성이 높고 인체에 주요한 암 유발 물질인 1, 2, 4-TCB를 전기화학적인 환원 반응을 통하여 탈염소화반응을 수행하였고, 그 결과는 HPLC로 분석하였으며, 다음과 같은 결과를 얻었다. 작업 전극의 종류에 따른 탈염소화 반응에서는 탄소 전극보다는 금속 전극인 납 전극에서 반응은 더 잘 일어났다. 그러나 탄소 전극을 다공성 탄소 전극으로 대체하였을 때에는 금속인 납 전극을 사용하였을 때보다 반응속도가 더 컸다. 이는 탄소 전극이라도 다공성으로 제작하여 그 표면적을 넓힐 경우 훨씬 좋은 반응 결과를 기대 할 수 있었으며, 다공성 탄소 전극을 사용함으로써 납 전극을 사용함에 따라 야기되는 2차 오염을 방지할 수 있었다. 최종 환원 전위인 -2.7V를 사용하여 1mM 1, 2, 4-TCB (0.1M TEAP) 의 탈염소화 반응에서 반응물인 1, 2, 4-TCB는 환원전위를 걸어준 60분이 경과한 후 모두 환원되었다. 중간체로 생성되는 1, 4-DCB와 1, 3-DCB 그리고 1, 2-DCB 는 모두 15분에서 최대치를 보여주었으며, 1, 2, 4-TCB와 같이 60분에서 모두 환원되었다. MCB의 경우는 70분에서 최대치를 나타냈으며, 반 이 시작한지 140분 만에 탈염소화 반응이 완료되었다. 최종 환원 전위인 -2.7V 이하를 사용한 경우도 (-2.6V, -2.5V) 반응속도는 느렸지만, 완전한 탈염소화 반응이 일어남을 보였다. 이는 반응 조건의 최적화를 통하여 환원전위 이하에서도 탈염소화 반응의 가능성이 연구되었다. 전해질의 농도를 0.1M에서 0.04M로 낮춤에 따라 예측한 대로 반응속도는 느렸다. 또한, 다 공성 전극을 사용하였을 때 0.04M 이하의 TEAP에서는 200분이 경과하여도 탈염소화 반응이 완결되지 못함을 볼 수 있다. 이는 낮은 농도의 전해질은 전자를 이동시키지 못하므로 전극과 시료간의 저항이 커져서 탈염소화 반응이 일어나지 않음을 나타내고 있다. 탈염소화 반응의 반응메카니즘은 첫 번째 전자 첨가로 음이온 라디칼을 형성하고 이로써 염소이온의 제거가 일어나며, 그 자리에 두 번째 전자가 첨가되어 탄소음이온을 형성하며 H^-의 첨가로 반응은 완결됨을 볼 수 있다. 또한, 1, 2, 4-TCB의 탈염소화 반응에 있어서의 중간체는 1, 4-DCB와 1, 3-DCB 그리고 1, 2-DCB이 각각 58% : 25% : 17%의 비로서 존재한다. 1, 2, 4-TCB의 탈염소화 반응의 증간체로 1, 4-DCB가 주생성물로 생성되는 이유는 전기분해로 인해 염소기가 제거되고 두 번째 전자첨가로 생성되는 1, 4-DCB음이온의 공명구조가 1, 3-DCB음이은 공명구조보다 안정하기 때문으로 생각된다. 이와 같은 탈염소화 반응은 전극의 조성, 첨가제(전해질)의 개발 그리고 용액의 조건 등의 최적화를 통하여 환원전위 이하에서도 탈염소화 반응의 가능성을 보여주었다. Polychlorinated aromatic hydrocarbons represent a major environmental problem. These materials, which were extensively used as transformer oils and heat-exchanger fluids since 1929, are major components of the hazardous waste disposal problem of the Environmental Protection Agency. The present report describes an electrolytic system for the reduction of chlorinated aromatic hydrocarbons, such as 1,2,4-trichlorobenzene (in the absence of oxygen) to their parent hydrocarbon. During the course of electrolysis, a DMF (0.1M TEAP) solution of 1,2,4-trichlorobenzene(1mM) was electrolyzed at -2.7V vs SCE at a porous glassy carbon(or glassy carbon or lead) electrode under nitrogen atmosphere. Porous glassy carbon cathode was even better than other non-porous glassy carbon, lead(Pb) cathode to dechlorination of 1,2,4-trichlorobenzene.

Mono-, Di-fluoro 5, 10, 15, 20-tetraphenylporphyrin Iron(Ⅲ)의 분광학적 연구

Resonance Raman(RR) spectra of 5,10, 15, 20-tetraphenylporphyriniron(III) difluoride, [Fe(TPP)F_2], which was formed by the reaction between Fe(TPP)Cl and tetrabutylammonium fluoride(TBAF 3H_2O) in various non-aqueous solvents, were obtained with the 441.6nm(He-Cd laser) excitation. [Fe(TPP)F_2] is six-coordinate iron porphyrin with two polar Fe-F bonds. The porphyrin skeletal modes can be correlated with previously recorded mode of Ni(TPP) and [FeTPP]_2O. And these assignments were confirmed by observed wavenumber shifts due to ??Fe/^54Fe and TPP/TOO-d_8/TPP-N_15 isotopic substitutions. Difluoroiron complex is an iron(III) high-spin complex with the oxidation sensitive band at 1347cm^-1 for v_4 and core size/spin state sensitive band at 1541cm^-1 for v_2.

« La guerre des cuillères » : précarité sociale et politique en Corée

Cet article analyse les conséquences politiques de la précarité sociale en Corée du Sud, où un tiers des emplois salariés sont précaires. A partir d’une enquête par sondage effectuée fin 2018, au sortir d’une crise politique majeure qui voit la destitution pour corruption de la présidente Park Geun-hye et l’élection triomphale du candidat progressiste Moon Jae-in, il explore le degré d’intégration politique, les préférences partisanes, et les attitudes populistes dans l’électorat. Une série de régressions logistiques montre que toutes choses égales par ailleurs, la précarité sociale n’a quasiment pas d’impact sur les choix partisans ni sur les sentiments anti-élites, elle encourage surtout la désaffection politique et l’abstention. Les principaux clivages politiques sont moins socioéconomiques que générationnels et régionaux. This article analyses the political consequences of social precariousness in South Korea, where one third of salaried jobs are precarious. Drawing from a survey conducted end 2018, after a major crisis when President Park Geun-hye was impeached for corruption and the progressive candidate Moon Jae-in triumphantly elected, it explores the degree of political integration, party preferences, and populist attitudes among voters. A series of logistic regressions show that, all else being equal, the impact of social precariousness on party preferences and anti-elites feelings is quasi null, it essentially favors political disaffection and abstention. The main political cleavages are less socio economic than generational and regional.
