        Enhanced activation of NAD(P)H: quinone oxidoreductase 1 attenuates spontaneous hypertension by improvement of endothelial nitric oxide synthase coupling via tumor suppressor kinase liver kinase B1/adenosine 5&vprime;-monophosphate-activated protein kina

        AIMS:: Guanosine 5&vprime;-triphosphate cyclohydrolase-1 (GTPCH-1) is a rate-limiting enzyme in de-novo synthesis of tetrahydrobiopterin (BH4), an essential cofactor for endothelial nitric oxide synthase (eNOS) coupling. Adenosine 5&vprime;-monophosphate-activated protein kinase (AMPK) is crucial for GTPCH-1 preservation, and tumor suppressor kinase liver kinase B1 (LKB1), an upstream kinase of AMPK, is activated by NAD-dependent class III histone deacetylase sirtuin 1 (SIRT1)-mediated deacetylation. &bgr;-Lapachone has been shown to increase cellular NAD/NADH ratio via NAD(P)H:quinone oxidoreductase 1 (NQO1) activation. In this study, we have evaluated whether &bgr;-lapachone-induced NQO1 activation modulates blood pressure (BP) through preservation of GTPCH-1 in a hypertensive animal model. METHODS AND RESULTS:: Spontaneously hypertensive rats (SHRs), primary aortic endothelial cells, and endothelial cell line were used to investigate the hypotensive effect of &bgr;-lapachone and its action mechanism. &bgr;-Lapachone treatment dramatically lowered BP and vascular tension in SHRs and induced eNOS activation in endothelial cells. Consistent with these effects, &bgr;-lapachone treatment also elevated levels of both aortic cGMP and plasma nitric oxide in SHRs. Meanwhile, &bgr;-lapachone-treated SHRs showed significantly increased levels of aortic NAD, LKB1 deacetylation, and AMPK Thr phosphorylation followed by increased GTPCH-1 and tetrahydrobiopterin/dihydrobiopterin ratio. In-vitro study revealed that AMPK inhibition by overexpression of dominant-negative AMPK nearly abolished GTPCH-1 protein conservation. Enhanced LKB1 deacetylation and AMPK activation were also elicited by &bgr;-lapachone in endothelial cells. However, inhibition of LKB1 deacetylation by blocking of NQO1 or SIRT1 blunted AMPK activation by &bgr;-lapachone. CONCLUSION:: This is the first study demonstrating that eNOS coupling can be regulated by NQO1 activation via LKB1/AMPK/GTPCH-1 modulation, which is possibly correlated with relieving hypertension. These findings provide strong evidence to suggest that NQO1 might be a new therapeutic target for hypertension.

        Although there have been many clinical studies about the facial trauma in the recent as well as in the past, severity and frequency of facial trauma are reported multifarious. It seemed to be because of variety of social, cultural, and environmental factors, and the view point of investigators. In this study, we investigated about the patients visiting emergency room during recent 4 years(2004-2007), and compared with retrospective study during former 10 years(1992-1996, 1999-2003) in Dept. OMS Pusan National University Hospital. We assessed the cause, type, demographic, etc, and analyzed alterations of facial bone fractures during past 3 periods. The results were as follows. The total number of patients was increased. 429 patients treated for maxillofacial fractures between January 1992 and December 1996, 466 patients treated between January 1992 and December 1996 and 764 patients treated between January 2004 and December 2007. The male-to-female ratio were 4.6:1, 3.9:1, and 4.16:1, respectively. Patient in the age groups of 10-19 years and 20-29 years increased in the latter two periods. Assaults and falls are decreased gradually, but slip down s are increased. Traffic accidents are still major cause of facial trauma. The mandibular fractures showed the highest incidence thorought three periods. Symphysis fractures of mandible was increased and condyle fractures of mandible was drecreased in the third period.

        Brain CT에서 차폐 재료 변화에 따른 수정체 선량 비교 연구

        우리나라 국민의 연간 진단용 방사선 검사 건수는 매년 증가하고 있으며, 특히 의료 피폭의 절반이상을 차지하는 CT검사에 대한 각별한 주의가 요구된다. 본 연구에서는 현재 CT검사 중 가장 많은 부분을 차지하는 두부 CT에서 CT Number, Noise, Uniformity를 유지하면서 수정체 부분의 입사표면선량(ESD)을 줄이기 위해, 현재 사용하고 있는 Bismuth 차폐체와 Aluminum 6mm, Silicone 22mm를 비교 실험하였다. 실험 결과 IOML에 평행한 나선형 scan과 고식적 scan에서 차폐체를 사용하지 않을 때의 선량보다 각각 Bismuth 26.41%, 17.52%, Aluminum 18.24%, 9.39%, Silicone 19.47%, 14.39% 감소되었다. 물의 CT Number, Noise, Uniformity 항목에서 Bismuth 차폐체의 경우 팬텀영상 검사 기준을 초과하였으며, Aluminum, Silicone의 경우는 기준내에 포함되었다. 화질의 변화를 줄이면서 차폐의 효과를 보고자 할 때 가공성이 좋고 구입하기 쉬운 Silicone을 이용한 차폐체가 도움이 되리라 생각한다. The cases of radiographic inspection for medical diagnosis in Korea have been continuously increasing year after year, which pays particularly more attention to CT which occupies over the half of medical radiation exposure. To find an effective alternative for reducing radiation exposure, the researchers conducted comparative experiments using some shields made of bismuth, aluminum 6mm, and silicone 22mm. These shielding materials have been used to reduce the entrance surface dose (ESD) on lenses, maintaining the CT number, noise, and uniformity in brain CT scanning which forms the largest part in CT scanning these days. These experiments showed that the doses in the spiral scan parallel to IOML and the conventional scan in Bismuth were 26.41% and 17.52%, respectively; in Aluminum 18.24% and 9.39%; in Silicone 19.47% and 14.39% lower than compared with those in the cases without any shields. In the items of the CT number, noise, and uniformity, the bismuth shield satisfied exceedingly the standards of the phantom image test while aluminum and silicone were within. To keep the graphic quality and get good shielding effect, we recommend the silicone shield which can be manufactured and purchased with ease.

        Background: Until today, there is currently no data available on the efficacy, safety, and tolerability of pegylated interferon alfa 2a (PEG-IFNa-2a) in Korean patients with chronic hepatitis B (CHB), who are known to have mostly HBV genotype C. Methods: We collected data from 18 institutes of 451 Korean patients with CHB who were treated with PEG-IFNa-2a as a first line therapy (370 patients with HBeAg positive and 81 with HBeAg negative). Treatment responses at the end of treatment (ET) and at 6 months post-treatment (PT6) were compared between patients treated for 24 weeks vs. 48 weeks, and adverse events were evaluated. Results: In HBeAg-positive patients, the patients who had received PEG-IFNa-2a for 48 weeks than 24 weeks achieved significantly higher virological response (HBV DNA < 2,000 IU/mL) (48 vs. 24 weeks at ET, 44.4% vs. 27.2%, p = 0.005; at PT6, 46.7% vs. 17.2%, p=0.001). Complete virological response (HBV DNA < 60 IU/mL) at ET was also achieved in more patients with a longer treatment (48 vs. 24 weeks 22.0% vs. 13.0%, p=0.005). HBeAg seroconversion rate at ET was 18.1% with 48 weeks treatment, which is significantly higher than 10.1% (p=0.012) with 24 weeks treatment. This finding was continued to PT6 (23.7% vs. 17.2%, p = 0.028). The rate of ALT normalization was increased from 61.4% at ET to 76.8% at PT6 on 48 weeks-treatment. HBsAg seroconversion was not common (48 vs. 24 weeks at ET, 0.4% vs. 0%; at PT6, 0.8% vs. 0%). In HBeAg-negative patients, virologic response at ET was higher than that in HBeAg-positive patients following 48 weeks treatment (serum HBV DNA< 60 and < 2,000 IU/mL; 60.8% and 87.8%, respectively). HBsAg seroconversion was 1.4% at ET and it was maintained at PT6. Adverse events were typical of those associated with PEG-IFNa-2a. Conclusion: In Korean CHB patients, PEG-IFNa-2a showed substantial treatment response and good tolerability. In patients with HBeAg-positive CHB, the longer treatment of PEG-IFNa-2a (48 weeks rather than 24 weeks) was more efficacious with similar safety profiles.

        고속철도는 최근 급격한 관련 기술의 발전으로 가장 효율적이고 이동성이 뛰어난 교통 수단 중의 하나가 되었다. 국내의 경우, 2004년 철도안전법 제정 이후, 철도시스템 개발시에는 RAMS 프로세스 절차에 따른 활동을 요구하고 있다. 본 연구에서는 철도운영관점에서 철도차량 운영과 관련된 효율적 신뢰성 관리 지표를 개발하기 위한 연구를 수행하였으며, 이를 위하여 국내·외 규격 및 타 운영사들의 신뢰성 관리 지표를 비교 분석하였다. Recently, rapid development of technology in high-speed railway industry has become one of the most efficient and excellent transport mobility. In case of domestic, after Railway Safety Act enacted in 2004, the railway system requires the activity of the RAMS procedures during development of this system. In this paper, we developed an efficient & practical Reliability management index according to the type of railway rolling stocks in railway operating company perspective and have case studies about the application plan for this index.

        Synthesis and Characterization of Blue Light-Emitting Polymers Containing Phenylenevinylene Units

        A series of new polymers containing phenylenevinylene (PV) units were synthesized by Wittig polycondensation reaction and properties of the synthesized polymers were characterized by FT-IR and UV-visible spectroscopy, and their light-emitting properties were studied. All of the synthesized polymers were soluble in organic solvents and showed good film quality. The absorption maxima and band edges of the polymers were moved to shorter wavelength region by reducing the electron donating alkoxy groups incorporated in phenylenevinylene unit. The photo-induced emission spectra were obtained and all of the polymers revealed their emission in blue region. The observed emission maxima of the polymers were ranged from 480-495 nm.

      • Antiatherogenic Effect of Antioxidant Polyphenols from <i>Phellinus baumii </i> in Apolipoprotein E-Deficient Mice

        <P>Abstract</P><P><I>Aims:</I> The present study was carried out to investigate the antiatherosclerotic effect of antioxidant polyphenols from <I>Phellinus baumii</I> (PBE) in apolipoprotein E-deficient (apoE–/–) mice. <I>Methods and Results:</I> apoE–/– mice were randomly divided into three groups: mice on a normal chow diet comprised the normal group, mice on an atherogenic diet plus vehicle were the control group, and mice on an atherogenic diet plus PBE (500 mg/kg) comprised the PB500 group. After 8 weeks of treatment, the plasma lipids and cytokine levels were measured. Although no significant differences were found in cholesterol levels among groups, the triglyceride level was significantly decreased in the PBE-treated group compared with the control group. Plasma tumor necrosis factor (TNF)-α and interleukin (IL)-6 levels were reduced by PBE treatment. Real-time PCR analysis of the aorta showed that PBE significantly prevented the upregulation of the vascular cell adhesion molecule (VCAM)-1, intercellular adhesion molecule (ICAM)-1, TNF-α, IL-6, and IL-1β expression. Furthermore, reduced macrophage infiltration, lipid accumulation and atherosclerotic lesions were observed in the aortic sinus and en face of the whole aorta in PBE-fed apoE–/– mice compared with atherogenic diet-fed control mice. <I>Conclusions:</I> Collectively, the findings of the present study suggest that the antiatherosclerotic effect of PBE is probably related to the inhibition of adhesion molecule and cytokine expression resulting in amelioration of lesion development.</P><P>Copyright © 2011 S.         리틀·주니어 야구복 상의 기준패턴 개발

        최근 리틀·주니어 야구 활동인구의 증가에 따라 국내 아동·청소년의 체형 특성 및 야구의 운동특성을 고려하여 야구복 상의의 사이즈스펙 및 기준패턴을 개발하였다. 리틀·주니어 야구 유니폼 디자인을 위한 연구의 대상 범위를 한국의 만7세~만15세 소년으로 설정하였다. 연구대상 범위의 중심 연령인 만11세를 기준 연령으로 설정하고, 5차 사이즈 코리아 직접 측정 장치를 이용하여 리틀·주니어 체형을 고려한 사이즈스펙을 개발하여 인체치수를 기반으로 한 그레이딩 편차를 설정하였다. 먼저 리틀·주니어 야구복을 생산하고 있는 국내 3개사 제품의 만 11세용 업체패턴을 비교분석하였다. 1차 연구패턴은 오픈형 셋인 소매 타입을 기준으로 가슴둘레 여유분 21cm 그리고 옷길이 67.2cm로 개발하였다. 3차원 인체형상을 이용하여 1차 가상 피팅 방법으로 전문가 5인이 착의평가 결과를 이용한 외관평가를 실시하였다. 그 결과 2차 가상 피팅 방법으로 제작한 기준패턴을 개발하였다. 그 결과 가슴둘레 여유분량이 16cm인 기준패턴이 개발되었다. 따라서 본 연구에서 개발 된 리틀·주니어를 위한 야구복 상의 사이즈스펙과 기준패턴은 작은 시장 규모와 영세한 업체가 밀집되어있는 야구복 생산업체에 제공함으로써 국내 리틀·주니어 야구복 시장의 기술기반 마련에 기초가 될 것이다. Amidst the recent increase of little & junior baseball population, this study aims at developing the size specification and prototype pattern of baseball jerseys considering the body type characteristics of Korean children & teenagers and the athletic characteristics of baseball. The target range of research for little & junior baseball uniforms design was set at 7~15 year old boys in Korea. With 11 years as the prototype age when physical changes suddenly begin, the direct measuring instrument of 5th Size Korea was used to develop the size specification considering little & junior body types, and the grading increment based. Currently, the 1<sup>st</sup> research pattern appropriate for the prototype size has been manufactured through comparative analysis of the business pattern for 11 year olds from 3 local producer of little & junior baseball uniforms. The 1st research pattern was developed with extra chest size 21cm and garment length 67.2cm based on the sleeve type of open-type set. Using the 3-dimensional human model, the 1<sup>st</sup> Virtual Fitting Simulation was created and fitting test was conducted by 5 experts. Accordingly, the final prototype pattern was designed, and the 2<sup>nd</sup> Virtual Fitting Simulation was conducted. According to its results, the final prototype pattern that set the extra chest size as 16cm was developed. Hence, the size specification and prototype pattern of baseball jerseys for little & junior developed in this study shall be provided to baseball uniform manufacturers of limited market size where small scale businesses are concentrated in establishing the foundation for technological basis in local little & junior baseball uniform market.
