        Ziwuliuzhu Acupuncture Modulates Clock mRNA, Bmal1 mRNA and Melatonin in Insomnia Rats

        Background: In clinics, Ziwuliuzhu acupuncture is widely considered an effective method of treating insomnia; however, there is currently limited information available regarding its possible mechanisms. Although the method of Ziwuliuzhu acupuncture possesses a unique rhythmic pattern. Objectives: In this study, we have creatively combined the traditional Chinese medicine of Ziwuliuzhu with a modern biological rhythm to investigate the internal mechanism of insomnia. Methods: Pathological tissue from the hypothalamus was analyzed using hematoxylin– eosin staining. The level of TNF (tumor necrosis factor)-α in the SCN (suprachiasmatic nucleus) area of the hypothalamus was detected in situ using the TUNEL fluorescence staining assay. The concentration of hypothalamic melatonin was detected using the enzyme-linked immunosorbent assay (ELISA). The mRNA expression of Clock and Bmal1 was measured using RT-qPCR. Results: In the Ziwuliuzhu acupuncture groups, the structural damage in the hypothalamic neurons was alleviated compared to the model group and the expression of inflammatory factors was reduced. The mRNA expression levels of Clock and Bmal1 were significantly increased (p < 0.05). The concentration of melatonin was significantly increased (p < 0.001). Although there were no significant differences between the treatment groups (diazepam group, Nazi group, Najia group, and routine group) (p > 0.05). Conclusion: Ziwuliuzhu acupuncture alleviated neuronal damage and modulated the inflammatory reaction in the hypothalamus of rats with insomnia. Moreover, Ziwuliuzhu acupuncture increased the expression levels of Clock and Bmal1 mRNA, and MT content. This study has potentially highlighted one of

        Simultaneous Determination of Glutamate, Glycine, and Alanine in Human Plasma Using Precolumn Derivatization with 6-Aminoquinolyl- N-hydroxysuccinimidyl Carbamate and High-Performance Liquid Chromatography

        A simple, sensitive and reproducible high-performance liquid chromatography (HPLC) method has been validated for determining concentrations of glutamate, glycine, and alanine in human plasma. Proteins in plasma were precipitated with perchloric acid, followed by derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC). Simultaneous analysis of glutamate, glycine, and alanine is achieved using reversed-phase HPLC conditions and ultraviolet detection. Excellent linearity was observed for these three amino acids over their concentration ranges with correlation coefficients (r)>0.999. The intra- and inter-day precision were below 10%. This method utilizes quality control samples and demonstrates excellent plasma recovery and accuracy. The developed method has been successfully applied to measure plasma glutamate, glycine, and alanine in twenty volunteers.

      • Screening for MiRNAs Related to Laryngeal Squamous Carcinoma Stem Cell Radiation

        Objective: To use microarray chip technology for screening of stem cell radiation related miRNAs in laryngeal squamous cell carcinoma; study and explore the relationship of miRNAs with radiosensitivity of laryngeal squamous cells. Method: After conventional culture and amplification of the laryngeal squamous carcinoma cell line Hep-2, CD 133+ cells were screened out with combination of isolated culture of stem cell microspheres and FACS for preparation of laryngeal cancer stem cells. After radiation treatment, miRNAs of laryngeal squamous carcinoma stem cells before and after radiation were enriched and purified. After microarray hybridization with mammalian miRNA and scanning of fluorescence signal, the miRNAs of laryngeal squamous carcinoma stem cells before and after radiation was subject to differential screening and clustering analysis. Real-time quantitative RT-PCR was used to verify part of the differentially expressed miRNAs. Results: 70 miRNAs related to laryngeal cancer stem cell radiation with 2-fold difference in expression were screened out, in which 62 were down-regulated and 8 were up-regulated. Fluorescent quantitative RT-PCR results were consistent with miRNAs chip results. Conclusion: Some miRNAs may be involved in self-regulation with laryngeal squamous carcinoma stem cell radiation.

        Physicochemical characterization and cytotoxicity of chitosanmodified single walled carbon nanotubes as drug carriers

        The application of single-walled carbon nanotubes (SWCNTs) as drug carriers is limited by their poor dispersal in aqueous medium. This study aimed to prepare chitosan (CS)-modified SWCNTs (CS-SWCNTs) and to evaluate their physicochemical properties and cytotoxicity. Oxidized SWCNTs (O-SWCNTs) were prepared with the use of strong acid, and the effects of acidizing conditions on the oxidation degree of the O-SWCNTs were investigated. CS was then non-covalently modified on the surfaces of O-SWCNTs. O-SWCNTs and CS-SWCNTs were characterized through ultraviolet spectroscopy, Fourier transform-infrared spectroscopy, Raman spectroscopy, and transmission electron microscopy. The cytotoxic effects of the functionalized SWCNTs were determined through the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. O-SWCNTs with relatively complete structure were successfully synthesized through 5 h of treatment with 5 M acid. The amine group of the CS and the carboxyl group of O-SWCNTs interacted in CS-SWCNTs. The functionalized SWCNTs did not aggregate or precipitate in water and exerted no cytotoxic effects on A549 and MCF-7 tumor cells. The CS-SWCNTs possess the advantages of a simple preparation process, excellent water dispersibility, and biocompatibility for drug loading.

        How Role Overload Affects Physical and Psychological Health of Low-ranking Government Employees at Different Ages: The Mediating Role of Burnout

        Background: The public now imposes higher demands on the government than in the past, which has created the role overload faced by low-ranking government employees in China. This research investigates the relationship between role overload and health among low-ranking government employees and explores the mediating effects of burnout. Methods: It draws on a survey of 2064 low-ranking government employees by probability proportionate to size sampling in China's Shandong Province. Structural equation modeling (SEM) methods are used to analyze the data. Results: Both role overload and burnout were found to have negative effects on low-ranking government employees' health; however, the associations varied among the three age groups (less than 36, between 36 and 45, and over 45). Those over 45 reported the highest level of both physical and psychological health, while the youngest age group (less than 36) reported the lowest level of health. Role overload has a direct influence on health among government employees over 45 but not among those below 45. Burnout's mediating effects between role overload and health are significant among all age groups, but most significant among the youngest civil servants below 36. Conclusions: The findings evidenced that both role overload and burnout affect low-ranking government employees' self-reported physical and psychological health. In addition, the effect of age differences in coping with role stressors and burnout should be considered.

        Interaction and micellar behavior of ternary mixture of amphoteric amino sulfonate surfactant with traditional anionic and nonionic surfactants: Effect of hydrophilicity

        The micellization and the molecular interaction behaviors for two ternary mixtures constituted by anamphoteric sodium 3-(n-dodecyl ethylenediamino)-2-hydroxypropyl sulfonate (C12AS), an anionicsodium dodecylbenzene sulfonate (SDBS) and a nonionic octylphenol polyoxyethylated ether with thenumber (n) of oxyethylene glycol ethers OP-n (n = 10 or 7) in aqueous solution were investigated usingthe tensiometry and the effect of hydrophilicity on them was also discussed. In the framework of pseudophaseseparation model and based on the regular solution theory, the related micellization parametersincluding the mixed critical micelle concentration (cmc) values in the ideal and real cases, the activitycoefficients and the compositions in mixed micelle, etc. and thermodynamic parameters were estimatedby the Clint’s model and the Rubingh’ model. The mixed cmc value is dependent on the composition inaqueous solution and influenced by the hydrophilicity of nonionic surfactant. With increasing the nonionicin ternary mixture, the mixed cmc value is initially decreased and then slightly rise. An increasingin the hydrophilicity of nonionic will make the minimum value of mixed cmc be increased from1.299 mM to 1.705 mM. The resulting phenomena can be explained reasonably by the electrostatic effect,the steric hindrance, the hydrogen bonding, etc. Thermodynamic data indicate that the contribution ofentropy or enthalpy plays a vital role on the spontaneous process of micellization and the share ofentropy or enthalpy in free energy change is dependent largely on the amount of nonionic surfactantand the hydrophilicity. In ternary mixtures of C12AS/SDBS/OP-10, an increase in an amount of OP-10 willinduce the change from the enthalpy-driven micellization process to the entropically favorable process. Once an abundant amount of OP-7 is added, while, the enthalpy will make a main contribution on themicellization process, which can be described by the drop in the share of entropy at the composition(0.6970/0.0000/0.3030) of ternary mixture from 0.6478 to 0.4901. These findings will help with understandingthe molecular interaction behavior for the ternary surfactant mixture and the effect of the additionof nonionic surfactant and its hydrophilicity.

        Structural property of soil is a new concept studied in science and study of this issue is a new problem, which involves soil mechanical effect, mechanical variability, and engineering design. Soil such as saline soil, red soil, loess, Xiashu loess, soft soil, or solidified dredger fill, which had been studied in previous research by our research group, has high pre-consolidation pressure (Pc) and high over-consolidation ratio (OCR). Considering the needs of intensity determination in engineering design, soil structure force, structural strength and other issues of structural Soils were studied in this paper. By analyzing experimental data, affecting factors of pre-consolidation pressure (Pc) was discussed. A new cognition about pre-consolidation pressure (Pc), determination method of structural strength, and genetic mechanism of structural force were proposed in this paper. Soil mass structural behavior and the mechanical effect can be one of the important fields of soil structural quantitative research.         Simulation of the Transmission Characteristics of a Hexagonal Photonic Crystal Slab Etched into an Ion-implanted LiNbO_3 Waveguide

        A photonic crystal slab that was constructed as a 2D hexagonal photonic crystal with a finite depth and was etched into an ion-implanted LiNbO_3 waveguide was simulated by using the finitedifference time-domain (FDTD) method. Both the ΓK-oriented and ΓM-oriented propagations were simulated with the fundamental TE mode of the LiNbO_3 waveguide as the incident beam. Despite the weak vertical confinement, the transmittance spectra show that they both have quite deep stop gaps. These gaps show a good overlap. At wavelengths within the overlap region, both the line defects that we created in the ΓK and the ΓM directions worked very well as channel waveguides.
