        Impact of selected fungi from an artificial diet on the growth and development of Drosophila suzukii (Diptera: Drosophilidae)

        Drosophila suzukii (Diptera: Drosophilidae) is one of the very few Drosophila species which are able to lay eggs and feed on healthy ripening fruit. Adults and larvae are all able to obtain the nutrition in the decaying food, which will generate many microorganisms. However, the relationship between D. suzukii and microorganism is poorly understood. In this study, 13 species of fungi were identified in an artificial diet fed by D. suzukii. Short-term and continuously life table experiments were conducted to determine the impact of three of these fungus species including Geotrichum candidum, Talaromyces minioluteus and Actinomucor elegans on the growth and development of Drosophila suzukii. Results revealed that, compared to the control, G. candidum, T.minioluteus and A. elegans increased the mortality of D. suzukii adults in the short time, while extended the developmental time of pupal D. suzukii by 18.00%, 16.22% and 26.44%, respectively in the life table experiment. T. minioluteus reduced the total longevity of D. suzukii by 15.52%, while A. elegans enhanced the total longevity by 25.96%. G. candidum and A. elegans increased the fecundity bymore than two folds. T. minioluteus elongated the mean generation time (T) by 31.34%, whereas G. candidum shortened it by 15.26% but increased the net reproductive rate (R0) by 217.76%, intrinsic rate of increase (r) by 88.89% and finite rate of increase (λ) by 9.17%. It was concluded that G. candidumand A. elegans significantly had the beneficial effect on the growth and reproduction of D. suzukii, however, T. minioluteus had the negative effect on D. suzukii. Our results could provide a new integrated pest management strategy for D. suzukii which would be discussed in this study.

        Effects of multi-strain probiotic supplementation on intestinal microbiota, tight junctions, and inflammation in young broiler chickens challenged with Salmonella enterica subsp. enterica

        Objective: This study assessed the effects of probiotics on cecal microbiota, gene expression of intestinal tight junction proteins, and immune response in the cecal tonsil of broiler chickens challenged with Salmonella enterica subsp. enterica. Methods: One-day-old broiler chickens (n = 240) were randomly allocated to four treatments: negative control (Cont), multi-strain probiotic-treated group (Pro), Salmonella-infected group (Sal), and multi-strain probiotic-treated and Salmonella-infected group (ProSal). All chickens except those in the Cont and Pro groups were gavaged with 1×108 cfu/mL of S. enterica subsp. enterica 4 days after hatching. Results: Our results indicated that body weight, weight gain, and feed conversion ratio of birds were significantly reduced (p<0.05) by Salmonella challenge. Chickens challenged with Salmonella decreased cecal microbial diversity. Chickens in the Sal group exhibited abundant Proteobacteria than those in the Cont, Pro, and ProSal groups. Salmonella infection downregulated gene expression of Occludin, zonula occludens-1 (ZO1), and Mucin 2 in the jejunum and Occludin and Claudin in the ileum. Moreover, the Sal group increased gene expression of interferon-γ (IFN-γ), interleukin-6 (IL-6), IL-1β, and lipopolysaccharide-induced tumor necrosis factor-alpha factor (LITAF) and reduced levels of transforming growth factor-β4 and IL-10 compared with the other groups (p<0.05). However, chickens receiving probiotic diets increased Lactobacillaceae abundance and reduced Enterobacteriaceae abundance in the ceca. Moreover, supplementation with probiotics increased the mRNA expression of Occludin, ZO1, and Mucin 2 in the ileum (p<0.05). In addition, probiotic supplementation downregulated the mRNA levels of IFN-γ (p<0.05) and LITAF (p = 0.075) and upregulated IL-10 (p = 0.084) expression in the cecal tonsil. Conclusion: The administration of multi-strain probiotics modulated intestinal microbiota, gene expression of tight junction proteins, and immunomodulatory activity in broiler chickens.

        Improvement of ulcerative colitis control by searching and restricting of inflammatory trigger factors in daily clinical practice

        Background/Aims: Exacerbating factors of ulcerative colitis (UC) are multiple and complex with individual influence. We aimed to evaluate the efficacy of disease control by searching and restricting inflammation trigger factors of UC relapse individually in daily clinical practice. Methods: Both patients with UC history or new diagnosis were asked to avoid dairy products at first doctor visit. Individual-reported potential trigger factors were restricted when UC flared up (Mayo endoscopy score ≥1) from remission status. The remission rate, duration to remission and medication were analyzed between the groups of factor restriction complete, incomplete and unknown. Results: The total remission rate was 91.7% of 108 patients with complete restriction of dairy product. The duration to remission of UC history group was significantly longer than that of new diagnosis group (88.5 days vs. 43.4 days, P=0.006) in patients with initial endoscopic score 2–3, but no difference in patients with score 1. After first remission, the inflammation trigger factors in 161 relapse episodes of 72 patients were multiple and personal. Milk/dairy products, herb medicine/Chinese tonic food and dietary supplement were the common factors, followed by psychological issues, non-dietary factors (smoking cessation, cosmetic products) and discontinuation of medication by patients themselves. Factor unknown accounted for 14.1% of patients. The benefits of factor complete restriction included shorter duration to remission (P<0.001), less steroid and biological agent use (P=0.022) when compared to incomplete restriction or factor unknown group. Conclusions: Restriction of dairy diet first then searching and restricting trigger factors personally if UC relapse can improve the disease control and downgrade the medication usage of UC patients in daily clinical practice.

        새로운 유형의 스마트 화분의 제품 설계 설계에 관한 내용을 소개한다. 본 내용은 스마트 화분에서 실내환경 정보 모니터링에 관해서 처음 소개한다. 스마트 화분의 요구를 충족하기 위해 많은 제품들이 출시괴었다. 이 제품들의 장점과 단점들을 분석한 후에 하드웨어 설계에서 소프트웨어 시스템 개발에 이르기까지 IoT 기반의 스마트 화분을 설계하였다. 시스템의 하드웨어는 Arduino 플랫폼을 기반으로 설계하였으며, 스마트폰 어플과 원격 모니터링 및 소셜 환경을 제공할 수 있도록 하였다. 본 논문은 스마트 화분의 설계 내용을 자세히 기술하고, 반려 식물을 재배하는 사람들에게 더 좋은 환경 제공과 여가 생활의 편리함과 즐거움을 제공하는데 목적이 있다. This paper introduces the product design of a new type of smart flower pot. The article first introduced the trend of indoor planting. In order to meet this demand, companies have launched many smart flowerpot products. After analyzing the advantages and disadvantages of these products, I began to design an Iot-based smart flowerpot product, from hardware design to software system development. The hardware in the system mainly uses the Arduino platform, and cooperates with the smart terminal APP to realize remote monitoring and social operations. The article details the characteristics of the new product, and hopes to design this smart flowerpot to bring a better experience for planters, bringing convenience and fun to leisure life.

      • Annual Financial Impact of Thyroidectomies for Nodular Thyroid Disease in China

        A large proportion of patients with thyroid nodules in China undergo thyroidectomy in order to get confirmatory histology diagnosis. The financial impact of this modality remains to be investigated. To evaluate rationality of performing thyroidectomy without a routine FNA preoperatively from the economic perspective, we conducted a retrospective, observational study of all archival thyroidectomies with records of cost per stay (CPS), cost per day (CPD) and length of stay (LOS) from 2008 to 2013 in the First Affiliated Hospital of Nanjing Medical University. We compared all the parameters between cancer and non-cancer thyroidectomies. We recruited 6, 140 thyroidectomies with valid records of CPS, CPD and LOS in this period. The CPS of cancer thyroidectomy was significantly higher than non-cancer thyroidectomy. The percentage of cancer thyroidectomy increased from 26.5% to 41.6%. The percentage of annual cost of cancer thyroidectomies rose from 30.2% to 45.2%. The LOS for cancer and non-cancer thyroidectomy decreased while the CPD increased in the past six years. The estimated national cost in 2012 for all thyroidectomies would be USD 1.86 billion with USD 1.09 billion for non-cancer thyroidectomies. We have witnessed great improvement in the healthcare for patients with thyroid nodules in China. However, given limited healthcare resources, currently thyroid FNA for more precise preoperative diagnosis may help to curb the rapidly increasing demand in healthcare costs in the future for nodular thyroid disease in China.

      • Prognostic Implications for High Expression of MiR-25 in Lung Adenocarcinomas of Female Non-smokers

        Background: Adenocarcinoma (ADC) is the most common histological type of lung cancer and its proportion is rising, especially in Asian non-smoking women. Recent studies suggest miR-25 may have diverse effects on the pathogenesis of different types of cancer. However, the role of miR-25 in lung cancer is still unknown. The aim of this study was to investigate the potential clinical value of miR-25 in non-smoking women with lung ADC. Patients and Methods: Quantitative RT-PCR was performed to evaluate the expression of miR-25 in 100 lung ADC tumor tissues and matched plasma samples and Pearson correlation tests were used to analyze the relationship between values. Associations of miR-25 expression with clinicopathological features were determined using the Student's t-test. To determine prognostic value, overall survival (OS) was evaluated using the Kaplan-Meier method. Univariate and multivariate analyses were performed using the Cox proportional hazard model. Results: Expression of miR-25 in tissue was found to be associated with lymph node metastasis (P=0.021) and disease stage (P=0.012). Moreover, high miR-25 expression was also associated with poorer overall survival of women with lung ADC (P=0.008). Conclusion: Tissue miR-25 expression may be associated with tumor progression and have prognostic implications in female lung ADC patients.

        Improvement of Fumigaclavine C Production in a Two-stage Culture of Aspergillus fumigatus with Molasses as a Cost-effective Ingredient

        Fumigaclavine C (FC), which is produced by Aspergillus fumigatus, is a conidiation-associated ergot alkaloid with significant medical benefits. However, its application is restricted by low yields from submerged cultures. In this study, the technical feasibility of using molasses as a cost-effective ingredient for FC production in a two-stage culture of A. fumigatus was evaluated. The results indicated that molasses supplementation significantly enhanced FC accumulation by promoting conidiation and up-regulating hydroxymethylglutaryl-CoA reductase activity. Via the optimization of the two-stage process in the presence of molasses, FC production in shake flasks reached 226.9 mg/L, which was approximately three times that in the original medium (75.9 mg/L). The use of molasses as a cost-effective ingredient for FC fermentation was also successfully reproduced in a lab-scale bioreactor system in which the maximum FC production reached 215.0 mg/L. The FC production obtained in this study is the highest ever reported. This increased efficiency will enable large-scale production of FC and extend the application of molasses as a low-cost substrate for producing other conidiation-related secondary metabolites.

      • Suppression of Ku80 Correlates with Radiosensitivity and Telomere Shortening in the U2OS Telomerase-negative Osteosarcoma Cell Line

        Ku70/80 heterodimer is a central element in the nonhomologous end joining (NHEJ) DNA repair pathway, Ku80 playing a key role in regulating the multiple functions of Ku proteins. It has been found that the Ku80 protein located at telomeres is a major contributor to radiosensitivity in some telomerase positive human cancer cells. However, in ALT human osteosarcoma cells, the precise function in radiosensitivity and telomere maintenance is still unknown. The aim of this study was to investigate the effects of Ku80 depletion in the U2OS ALT cell line cell line. Suppression of Ku80 expression was performed using a vector-based shRNA and stable Ku80 knockdown in cells was verified by Western blotting. U2OS cells treated with shRNA-Ku80 showed lower radiobiological parameters (D0, Dq and SF2) in clonogenic assays. Furthermore, shRNA-Ku80 vector transfected cells displayed shortening of the telomere length and showed less expression of TRF2 protein. These results demonstrated that down-regulation of Ku80 can sensitize ALT cells U2OS to radiation, and this radiosensitization is related to telomere length shortening.

        대기압 플라즈마 제트의 기체 유량에 대한 방전 특성

        The influence of gas flow on the plasma generation in the atmospheric plasma jet is described with the theory of hydrodynamics. The plasma discharge is affected by the gas-flow streams with Reynolds number (Re) as well as the gas pressure with Bernoulli's theorem according to the gas flow rate inserted into the glass tube. The length of plasma column is varied with the flow types such as the laminar flow of Re<2,000 and the turbulent flow of Re>4,000 as it has been known in a general fluid experiments. In the laminar flow, the plasma column length is increased as the increase of flow rate. Since the pressure in the glass tube becomes low as the increase of flow velocity by the Bernoulli's theorem, the breakdown voltage of plasma discharge is reduced by the Paschen's law. Therefore, the plasma length is increased as the increasing flow rate with the fixed operation voltage. In the transition of laminar and turbulent flows, the plasma length is decreased. When the flow becomes turbulent as the flow rate is increasing, the plasma length becomes short and the discharge is shut down ultimately. In the discharge of laminar flow, the diameter of plasma beam exposed on the substrate surface is kept less than the glass diameter, since the gas flow is kept to the distinct distance from the nozzle of glass tube. 대기압 플라즈마 제트 장치의 유량 변화에 대한 플라즈마 방전 특성을 실험적으로 조사하고 이를 유체역학적으로 해석하였다. 유리관에 주입되는 아르곤 기체의 유량 변화에 대한 레이놀즈 수(Re)로 결정되는 기체 흐름의 형태 변화와 베르누이 정리에 의한 압력 변화가 플라즈마 발생에 영향을 준다. 유리관 내부에 발생하는 플라즈마의 길이 변화의 실험을 통하여, 아르곤 기체에 대한 레이놀즈 수가 Re<2,000이면 층류이고, Re>4,000이면 난류가 형성된다. 이는 일반 유체에서 알려진 결과와 일치한다. 층류에서 유량의 증가로 플라즈마의 길이가 증가한다. 층류와 난류의 전환 영역에서 플라즈마의 길이는 줄어든다. 난류 영역에서는 방전 기체의 흐름이 불규칙함으로서 방전 경로가 흐트러져 플라즈마 칼럼의 길이가 매우 짧아지고 급기야 플라즈마가 소멸된다. 층류에서 주입 유량의 증가로 유리관 내의 유속이 증가하면, 베르누이 정리에 의하여 유리관 내부의 압력이 낮아진다. 관내의 압력이 낮아지면, 파센 법칙에 의하여 관내의 전기장의 세기가 증가하여 방전 전압이 낮아진다. 따라서 주입 유량이 증가하면, 동일한 구동 전압에서 유리관에 발생하는 플라즈마의 길이는 길어진다. 층류의 방전은 유리관 밖에서도 층류의 흐름이 일정 길이로 유지되므로 시료 표면에 조사되는 플라즈마 빔의 직경은 유리관의 직경 이하로 유지된다.
