        Estimation of the net energy requirement for maintenance in broilers

        Objective: The net energy requirement for the maintenance (NEm) of broilers was determined using regression models by the indirect calorimetry method (ICM) or the comparative slaughter method (CSM). Methods: A 2×4 factorial arrangement of treatments including the evaluation method (ICM or CSM) and feed intake (25%, 50%, 75%, or 100% of ad libitum recommended) was employed in this experiment. In the ICM, 96 male Arbor Acres (AA) birds aged d 15 were used with 4 birds per replicate and 6 replicates in each treatment. In the CSM, 116 male AA birds aged d 15 were used. Among these 116 birds, 20 were selected as for initial data and 96 were assigned to 4 treatments with 6 replicate cages and 4 birds each. The linear regression between retained energy (RE) and metabolizable energy intake (MEI) or the logarithmic regression between heat production (HP) and MEI were used to calculate the metabolizable or net energy requirement for maintenance (MEm) or NEm, respectively. Results: The evaluation method did not detect any differences in the metabolizable energy (ME), net energy (NE), and NE:ME of diet, and in the MEI, HP, and RE of broilers. The MEI, HP, and RE of broilers decreased (p<0.01) as the feed intake decreased. No evaluation method× feed intake interaction was observed on these parameters. The MEm and NEm estimated from the linear relationship were 594 and 386 kJ/kg of body weight (BW)0.75/d in the ICM, and 618 and 404 kJ/kg of BW0.75/d in the CSM, respectively. The MEm and NEm estimated by logarithmic regression were 607 and 448 kJ/kg of BW0.75/d in the ICM, and were 619 and 462 kJ/kg of BW0.75/d in the CSM, respectively. Conclusion: The NEm values obtained in this study provide references for estimating the NE values of broiler diets.

      • Clinicopathological Features and Prognosis of Gastroenteropancreatic Neuroendocrine Tumors: Analysis from a Single-institution

        Background: The gastroenteropancreatic neuroendocrine neoplasm (GEP-NEN) is the most common type of neuroendocrine neoplasm. We summarized data in our centre to investigate the clinicopathological features, diagnostic methods, therapeutic approaches and prognosis for this neoplasm to increase knowledge of this disease in Asian populations. Method: A total of 122 patients treated at Sun Yet-san Memorial Hospital of Sun Yat-sen University between January 2000 and December 2011 were analyzed retrospectively. Results: Pancreas was the most common site of involvement (65/122, 53.3%); this disease has no special symptoms; positive rates of chromogranin A (CgA) and synaptophysin (Syn) were 81.1% and 87.7%, respectively. The positive rate of Syn had statistical difference among the three grades, but not CgA. Some 68 patients had G1 tumors, 32 G2 tumors and 22 G3 tumors, and Chi-square test showed that higher grading was correlated with worse prognosis (${\chi}^2=32.825$, P=0.0001). A total of 32 patients presented with distant metastasis, and 8 cases emerged during following up. Cox proportional hazards regression modeling showed that the tumor grade (P=0.01), lymphatic metastasis (P=0.025) and distant metastasis (P=0.031) were predictors of unfavorable prognosis. The overall 5-year survival rate was 39.6%, the 5-year survival rate of G1 was 55.7%, and the G2 and G3 were 34.2% and 0%, respectively. Conclusions: The incidence of gastroenteropancreatic neuroendocrine tumors has risen over the last 12 years. All grades of these diseases metastasize readily, and further research regarding the treatment of patients after radical surgery is needed to prolong disease-free survival.

      • Inhibition of Tumor Growth in Vitro by a Combination of Extracts from Rosa Roxburghii Tratt and Fagopyrum Cymosum

        Objective: Traditional Chinese herbal medicines have a very long history. Rosa roxburghii Tratt and Fagopyrum cymosum are two examples of plants which are reputed to have benefits in improving immune responses, enhancing digestive ability and demonstrating anti-aging effects. Some evidence indicates that herbal medicine soups containing extracts from the two in combination have efficacy in treating malignant tumors. However, the underlying mechanisms are far from well understood. The present study was therefore undertaken to evaluate anticancer effects and explore molecular mechanisms in vitro. Methods: Proliferation and apoptosis were assessed with three carcinoma cell lines (human esophageal squamous carcinoma CaEs-17, human gastric carcinoma SGC-7901 and pulmonary carcinoma A549) by MTT assay and flow cytometry, respectively, after exposure to extract from Rosa roxburghii Tratt (CL) and extract from Fagopyrum cymosum (FR). $IC_{30}$ of CL and FR were obtained by MTT assay. Tumor cells were divided into four groups : control with no exposure to CL or FR; CL with $IC_{30}$ CL; FR with $IC_{30}$ FR; CL+FR group with 1/2 ($IC_{30}$ CL + $IC_{30}$ FR). RT-PCR and Western blot analysis were used to detect the expression of Ki-67, Bax and Bcl-2 at mRNA and protein levels. Results: Compared with the CL or FR groups, the combination of CL+FR showed significant inhibition of cell growth and increase in apoptosis; the mRNA and protein expression levels of Ki-67 and Bcl-2 in CL+FR group were all greatly decreased, while the expression of Bax was markedly increased. Conclusions: These results indicate that the synergistic antitumor effects of combination of CL and FR are related to inhibition of proliferation and induction of apoptosis.

        One New Pregnane Glycoside from the Seeds of Cultivated Brucea javanica

        A new pregnane glycoside, named (20R)-O-(3)-β-D-glucopyranosyl-(1→2)-α-L-arabinopyranosyl-pregn-5-en-3β,20-diol (1), and seven known compounds, brusatol (2), bruceine B (3), bruceine D (4), yadanziolide A (5), bruceine E (6), yadanzioside G (7), and yadanzioside B (8), were isolated from the cultivated dry seeds of Brucea javanica. The structure of 1 was elucidated on the basis of 1D- and 2D-NMR spectroscopic analyses. Their inhibitory effects on tumor cells were also tested. Compound 1 was slightly active against HL-60, SMMC-7721, A-549, and MCF-7 tumor cells. Compounds 2 and 3 demonstrated significant inhibitory activities against all tested cells. These results indicate that cultivated B. javanica could replace the wild plant as an antitumor plant resource.

        MiR-17-5p modulates osteoblastic differentiation and cell proliferation by targeting SMAD7 in non-traumatic osteonecrosis

        MicroRNAs (miRNAs) have recently been recognized to have a role in human orthopedic disorders. The objective of our study was to explore the expression profile and biological function of miRNA-17-5p (miR-17-5p), which is well known to be related to cancer cell proliferation and invasion, in osteoblastic differentiation and in cell proliferation. The expression levels of miR-17- 5p in the femoral head mesenchymal stem cells of 20 patients with non-traumatic osteonecrosis (ON) and 10 patients with osteoarthritis (OA) were examined by quantitative reverse transcription-PCR (qRT–PCR). Furthermore, the interaction between miR-17-5p and SMAD7 was observed. We found that in non-traumatic ON samples the level of mature miR-17-5p wassignificantly lower than that of OA samples (P¼0.0002). By targeting SMAD7, miR-17-5p promoted nuclear translocation of b-catenin, enhanced expression of COL1A1 and finally facilitated the proliferation and differentiation of HMSC-bm cells. We also demonstrated that restoring expression of SMAD7 in HMSC-bm cells partially reversed the function of miR-17-5p. Together, our data suggested a theory that dysfunction of a network containing miR-17-5p, SMAD7 and b-catenin could contribute to ON pathogenesis. The present study prompts the potential clinical value of miR-17-5p in non-traumatic ON.

        Post-fi re Behavior of Welded Hollow Spherical Joints Subjected to Eccentric Loads

        Welded hollow spherical joints are widely used as a connection pattern in space lattice structures. Understanding the postfi re residual behavior of welded hollow spherical joints is crucial for fi re damage assessment of the space lattice structures. However, the post-fi re behavior of welded hollow spherical joints has not been explored in existing studies. In this paper, experimental and numerical studies were conducted to investigate the residual behavior of eccentrically loaded welded hollow spherical joints after fi re exposure. Eccentric compressive tests were performed on fi ve joint specimens after exposure to the ISO-834 standard fi re (including both heating and cooling phases), and three highest fi re temperatures, i.e., 600, 800, and 1000 °C, were considered. The temperature distributions in the specimens during the heating and cooling process and the related mechanical behavior of the specimens, such as load versus longitudinal displacement and rotation responses, load-bearing capacities, and strain distributions, were obtained and analyzed. Finite element analysis (FEA), including both heat transfer and mechanical analysis, were also developed using the ABAQUS software. Having validated the FE models against the experimental results, a design method was proposed on the basis of parametric studies to predict the residual load-bearing capacity of eccentrically loaded welded hollow spherical joints after fi re exposure.

        Experimental Study on Shear Behavior of BFRP-reinforced Recycled Aggregate Concrete Deep Beams without Stirrups

        This paper describes an experimental investigation to evaluate the shear performance of basalt Fiber-reinforced Polymer (BFRP)- reinforced recycled aggregate concrete deep beams without stirrups. A total of 13 beams were tested under four-point loading until failure: nine were reinforced with BFRP bars, and four were reinforced with steel bars. The parameters of this experiment included the shear-span-to-depth ratio (a/d), reinforcement ratio of the longitudinal bar (ρ), sectional effective depth (d) and compressive strength of the recycled concrete ( ). The shear behavior of the beams was described in terms of the crack type, failure mode, shear strength and load-deformation relationship. The effects of the experimental variables on shear capacities and deflections of the tested beams were also investigated. The experimental results showed that the shear capacity of BFRP-reinforced beams was smaller than that of steel-reinforced beams. Additionally, the deformation of BFRP-reinforced beams was larger than that of steel-reinforced ones. The shear capacity of BFRP-reinforced recycled concrete beams without stirrups showed a linear relationship with the sectional effective depth d and was proportional to (d/a)4/3, (ρ)1/5 and ( )1/3. Meanwhile, four existing codes were used to predict the shear strength of test beams and the CSA806-02 gave the better results.

        Effect of Temperature on Resistivity of CFRP Materials with Added Carbon Powder or Nano-silica

        This paper presents an experimental investigation on the effect of temperature on resistivity of Carbon Fiber Reinforced Polymer (CFRP) materials. A series of tests were conducted on three types of CFRP materials, namely pure CFRP material, CFRP with carbon powder (4% in weight) and CFRP with nano-silica (4% in weight). Test results showed that adding carbon powder into the epoxy resin decreases the initial electric resistance R0 and initial volume resistivity ρ0 while adding nano-silica increases R0 and ρ0 compared to pure CFRP material. Preheating cycle test results showed that the volume resistivity of all three types of specimens linearly increases with increasing temperature. CFRP with added nano-silica exhibits higher temperature sensitivity than CFRP with added carbon powder compared to the lowest temperature sensitivity for pure CFRP material. In addition, temperature cycle test results showed that CFRP specimens have approximately stable values of volume resistivity. Both CFRP specimens with added carbon powder or nano-silica exhibit a recognizable trend of first decrease and then increase in volume resistivity with increasing temperature both during heating and cooling cycles. CFRP with added carbon powder mainly shows Negative Temperature Coefficient (NTC) effect in the temperature range of −40 to 40oC and Positive Temperature Coefficient (PTC) effect from 40 to 80oC. CFRP with added nano-silica mainly exhibits PTC effect in the temperature domain of −15 to 80oC and NTC effect from −40 to −15oC. A mathematical-physical model with respect to the thermal effect was presented based on the Eshelby-Mori-Tanaka (EMT) approach and mesomechanics method. The results obtained with the model agree well with the test results considering the temperature domain of PTC effect, which indicates that the proposed model is effective in characterizing the variation of fractional change in resistance (ΔR/R0) at varying temperature.

      • Interactive Effect of Bisphenol A (BPA) Exposure with -22G/C Polymorphism in LOX Gene on the Risk of Osteosarcoma

        Background: Osteosarcomas have many established risk factors, both genetic and environmental, but by themselves these explain only part of the total cancer incidence. Bisphenol A (BPA) is an environmental estrogen associated with risk of several kinds of tumour. The lysyl oxidase gene (LOX) may also contribute to risk of tumours including osteosarcomas. Here, we investigated possible interactions of BPA and a LOX polymorphism on the risk of osteosarcoma. Method: The present hospital-based case-control study included 106 cancer patients and 112 controls from a Chinese population. Internal burden of BPA exposure was assessed using high-performance liquid chromatography-mass spectrometry (HPLC-MS) method. Genotypes were determined using PCR-RFLP methods. Results: Compared with those in low BPA exposure group, subjects with BPA more than or equal to median value had significant increased risk of osteosarcoma among subjects who carried GC or CC genotypes. A significant interaction with BPA level and the -22G/C polymorphism was observed for osteosarcoma overall, osteosarcoma affecting knee and osteosarcoma affecting hip, as $P_{forinteraction}$ = 0.036 for osteosarcoma overall; $P_{forinteraction}$ = 0.024 for osteosarcoma affecting knee; and $P_{forinteraction}$ = 0.017 for osteosarcoma affecting hip. Conclusions: The results suggest that BPA exposure interacts with the -22G/C polymorphism of the LOX gene to increase the risk of osteosarcoma.

        Preparation of solid acid catalyst packing AAO/SBA-15-SO3H and application for dehydration of xylose to furfural

        The solid acid catalyst packing AAO/SBA-15-SO3H was prepared by the co-condensation and grafting method with porous anodic aluminum oxide (AAO) as support. FT-IR, SEM and TEM were applied to characterize the prepared samples. Results showed that catalysts prepared by two methods both contained active centers, and SBA-15 nanorod arrays grow inside a porous alumina membrane AAO and are perpendicular to the substrate. Their catalytic performances were tested for dehydration of xylose to furfural. The conversion of xylose and selectivity of furfural were 90% and 74% on the AAO/SBA-15-SO3H(C) catalyst prepared by the co-condensation method, respectively. The deactivation and regeneration of the AAO/SBA-15-SO3H(C) catalyst for the dehydration of xylose were also investigated,the activity of catalyst treated by 30 wt.% H2O2 almost was recovered.
