        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        miR-638 is a new biomarker for outcome prediction of non-small cell lung cancer patients receiving chemotherapy

        MicroRNAs (miRNAs), a class of small non-coding RNAs, mediate gene expression by either cleaving target mRNAs or inhibiting their translation. They have key roles in the tumorigenesis of several cancers, including non-small cell lung cancer (NSCLC). The aim of this study was to investigate the clinical significance of miR-638 in the evaluation of NSCLC patient prognosis in response to chemotherapy. First, we detected miR-638 expression levels in vitro in the culture supernatants of the NSCLC cell line SPC-A1 treated with cisplatin, as well as the apoptosis rates of SPC-A1. Second, serum miR-638 expression levels were detected in vivo by using nude mice xenograft models bearing SPC-A1 with and without cisplatin treatment. In the clinic, the serum miR-638 levels of 200 cases of NSCLC patients before and after chemotherapy were determined by quantitative real-time PCR, and the associations of clinicopathological features with miR-638 expression patterns after chemotherapy were analyzed. Our data helped in demonstrating that cisplatin induced apoptosis of the SPC-A1 cells in a dose- and time-dependent manner accompanied by increased miR-638 expression levels in the culture supernatants. In vivo data further revealed that cisplatin induced miR-638 upregulation in the serum derived from mice xenograft models, and in NSCLC patient sera, miR-638 expression patterns after chemotherapy significantly correlated with lymph node metastasis. Moreover, survival analyses revealed that patients who had increased miR-638 levels after chemotherapy showed significantly longer survival time than those who had decreased miR-638 levels. Our findings suggest that serum miR-638 levels are associated with the survival of NSCLC patients and may be considered a potential independent predictor for NSCLC prognosis.

        Integrative Modeling of Wireless RF Links for Train-to-Wayside Communication in Railway Tunnel

        In railway tunnel environment, the reliability of a high-data-rate and real-time train-to-wayside communication should be maintained especially when high-speed train moves along the track. In China and Europe, the communication frequency around 900 MHz is widely used for railway applications. At this carrier frequency band, both of the solutions based on continuously laid leaky coaxial cable (LCX) and discretely installed base-station antennas (BSAs), are applied in tunnel radio coverage. Many available works have concentrated on the radio-wave propagation in tunnels by different kinds of prediction models. Most of them solve this problem as natural propagation in a relatively large hollow waveguide, by neglecting the transmitting/receiving (Tx/Rx) components. However, within such confined areas like railway tunnels especially loaded with train, the complex communication environment becomes an important factor that would affect the quality of the signal transmission. This paper will apply a full-wave numerical method to this case, for considering the BSA or LCX, train antennas and their interacted environments, such as the locomotive body, overhead line for power supply, locomotive pantograph, steel rails, ballastless track, tunnel walls, etc.. Involving finite-difference time-domain (FDTD) method and uni-axial anisotropic perfectly matched layer (UPML) technique, the entire wireless RF downlinks of BSA and LCX to tunnel space to train antenna are precisely modeled (so-called integrative modeling technique, IMT). When exciting the BSA and LCX separately, the field distributions of some cross-sections in a rectangular tunnel are presented. It can be found that the influence of the locomotive body and other tunnel environments is very significant. The field coverage on the locomotive roof plane where the train antennas mounted, seems more homogenous when the side-laying position of the BSA or LCX is much higher. Also, much smoother field coverage solution is achieved by choosing LCX for its characteristic of more homogenous electromagnetic wave radiation.

        Digital Image-based Identification Method for the Determination of the Particle Size Distribution of Dam Granular Material

        The Particle Size Distribution (PSD) properties of dam granular material plays an important role in the construction process ofearth-rock dams, as it can affect the filling quality and structural safety. However, the conventional sieving method employed tocheck the PSD is labor-intensive, time-consuming and not highly accurate. In this study, a digital image-based identification methodis presented for the determination of the PSD of dam granular material, which mainly incorporates image acquisition technology, alarge database and a neural network. Digital Image Processing (DIP) technology is used to recognize the geometric size and gradingcurve of dam granular materials at a small scale, while statistical distribution models are used to determine the characteristicparameters of the grading curve and convert the graphical curve into mathematical variables. Furthermore, a large database and a BPneutral algorithm, which is improved using a genetic algorithm, are introduced as tools to reveal the implicit relationship between theDIP and sieving grading curves to correct the error of identification. A case study for the Changheba Hydropower Station is used toillustrate the implementation details of the presented method. The identification results demonstrate that the presented method canacquire and assess the gradation in spite of a degree of error, which can be decreased when more advanced DIP technologies areexplored, the amount of data in the database is increased, and a more optimized network algorithm is adopted.

      • Utrecht Interstitial Applicator Shifts and DVH Parameter Changes in 3D CT-based HDR Brachytherapy of Cervical Cancer

        Background: For brachytherapy of cervical cancer, applicator shifts can not be avoided. The present investigation concerned Utrecht interstitial applicator shifts and their effects on organ movement and DVH parameters during 3D CT-based HDR brachytherapy of cervical cancer. Materials and Methods: After the applicator being implanted, CT imaging was achieved for oncologist contouring CTVhr, CTVir, and OAR, including bladder, rectum, sigmoid colon and small intestines. After the treatment, CT imaging was repeated to determine applicator shifts and OARs movements. Two CT images were matched by pelvic structures. In both imaging results, we defined the tandem by the tip and the base as the marker point, and evaluated applicator shift, including X, Y and Z. Based on the repeated CT imaging, oncologist contoured the target volume and OARs again. We combined the treatment plan with the repeated CT imaging and evaluated the change range for the doses of CTVhr D90, D2cc of OARs. Results: The average applicator shift was -0.16 mm to 0.10 mm for X, 1.49 mm to 2.14 mm for Y, and 1.9 mm to 2.3 mm for Z. The change of average physical doses and EQD2 values in Gy${\alpha}/{\beta}$ range for CTVhr D90 decreased by 2.55 % and 3.5 %, bladder D2cc decreased by 5.94 % and 8.77 %, rectum D2cc decreased by 2.94 % and 4 %, sigmoid colon D2cc decreased by 3.38 % and 3.72 %, and small intestines D2cc increased by 3.72 % and 10.94 %. Conclusions: Applicator shifts and DVH parameter changes induced the total dose inaccurately and could not be ignored. The doses of target volume and OARs varied inevitably.

        Cultivation of Nostoc flagelliforme on Solid Medium

        N/A In order to construct an artificial cultivation of Nostoc flagelliforme on solid medium, we attempted to assess the viability of approaches, which utilized either BG -11 agar or sand medium using both sterile and non-sterile algal segments. In the trial in which the BG-11 agar medium was inoculated with the non-sterile algal segments, the algae exhibited the rapid growth in the initial 4 days of cultivation. However, after 4 days of cultivation, the growth rate of the algae slowed, and the algal growth was completely stopped by 7 days of cultivation. When the BG-11 medium was inoculated with the sterile algal segments, the algae exhibited the rapid growth for a longer period of 8 days, reaching a length of 24.9 mm. The growth rate during this period was measured to be 24.5%. After the 8 days of cultivation, the algal growth rate began to slow and had almost stopped by the 13 days of cultivation. On the other hand, when the sterile algal segments were inoculated onto a sand plate, the algal segments decomposed, reaching total decomposition after 11 days of cultivation. By way of contrast, the desiccation treatment samples continued to grow for 14 days of cultivation. After 14 days of cultivation, the algae achieved a length of 26.1 mm, with a growth rate of 30.6%. Our results indicate that periodic desiccation may constitute an effective strategy for the prevention of algal decomposition.

        Synthesis and anticonvulsant activity evaluation of 8-alkoxy-5-(4H-1,2,4-triazol-4-yl)quinoline derivatives

        Two series of 8-alkoxy-5-(4H-1,2,4-triazol-4-yl)quinolines and 8-alkoxy-5-(2H-1,2,4-triazol-3-one-4-yl)quinolineswere synthesized.The anticonvulsant activity ofthese compounds was evaluated with maximal electroshockseizure test and rotarod test. Among the synthesized compounds,8-octoxy-5-(4H-1,2,4-triazol-4-yl)quinoline (4g)was the most active compound with ED50 of 8.80 mg/kg,TD50 of 176.03 mg/kg and protective index of 20.0. Itsneurotoxicity was lower than all other synthesized compoundsand also markedly lower than that of the referencedrug carbamazepine. In addition, the potency of compound 4gagainst seizures induced by pentylenetetrazole, 3-mercaptopropionicacid, and bicuculline suggested its broad spectrumactivity, and the mechanisms of action including inhibition ofvoltage-gated ion channels and modulation of GABAergicactivity might involve in its anticonvulsant activity.

      • Association of XRCC3 Thr241Met Polymorphisms and Gliomas Risk: Evidence from a Meta-analysis

        The relationship between the X-ray repair cross-complementing group 3 (XRCC3) Thr241Met polymorphism and gliomas remains inclusive or controversial. For better understanding of the effect of XRCC3 Thr241Met polymorphism on glioma risk, a meta-analysis was performed. All eligible studies were identified through a search of PubMed, Elsevier Science Direct, Excerpta Medica Database (Embase) and Chinese Biomedical Literature Database (CBM) before May 2013. The association between the XRCC3 Thr241Met polymorphism and gliomas risk was conducted by odds ratios (ORs) and 95% confidence intervals (95% CIs). A total of nine case-control studies including 3,533 cases and 4,696 controls were eventually collected. Overall, we found that XRCC3 Thr241Met polymorphism was significantly associated with the risk of gliomas (T vs. C: OR=1.10, 95%CI=1.01-1.20, P=0.034; TT vs. CC: OR=1.30, 95%CI=1.03-1.65, P=0.027; TT vs. TC/CC: OR=1.29, 95%CI=1.01-1.64, P=0.039). In the subgroup analysis based on ethnicity, the significant association was found in Asian under four models (T vs. C: OR=1.17, 95%CI=1.07-1.28, P=0.00; TT vs. CC: OR=1.79, 95%CI=1.36-2.36, P=0.00; TT vs. TC/CC: OR=1.75, 95%CI=1.32-2.32, P=0.00; TT/TC vs. CC: OR=1.11,95% CI=1.02-1.20). This meta-analysis suggested that the XRCC3 Thr241Met polymorphism is a risk factor for gliomas, especially for Asians. Considering the limited sample size and ethnicities included in the meta-analysis, further large scale and well-designed studies are needed to confirm our results.

      • Comparison of Clinical Efficacy of Three Different Neoadjuvant Approaches (Chemotherapy Combined Vaginal Intracavitary Irradiation, Neoadjuvant Chemotherapy Alone or Radiotherapy) Combined with Surgery for Patients with Stage Ib2 and IIa2 Cervical Cancer

        A total of 285 patients with stage Ib2 and IIa2 cervical cancer were categorized into three groups, and received preoperative neoadjuvant chemotherapy combined with vaginal intracavitary irradiation, neoadjuvant chemotherapy alone or radiotherapy, respectively. The effective rate of 70.6 % in group 1 was much higher than 41.4% in group 2 (P=0.000) and 46.9 % in group 3 (P=0.000); The percentage of patients receiving postoperative adjuvant therapy was 44.1% in group 1, much lower than 67.8% in group 2 (P=0.001) and 64.6% in group 3 (P=0.004); The percentage of patients with no postoperative risk factor in group 1 was 52.0%, much higher than 32.2% in group 2 (P=0.006) and 35.4% in group 3 (P=0.019); The occurrence rate of surgery-related complications in groups 1, 2 and 3 were 29.4%, 28.7%, and 33.3%, respectively, with no statistical differences among the groups (P=0.981). Regarding preoperative neoadjuvant complications, none were obvious in group 3, while occurrence rates of myelosuppression in groups 1 and 2 were 89.1% and 86.6%, of nausea and vomitting were 78.4% and 78.2%, but without significant differences (all P>0.05). Among 166 patients who received postoperative adjuvant therapy in the three groups, the occurrence rates were: 65.4%, 64.3% and 61.1% respectively for myelosuppression; 42.3%, 38.1%, and 38.9% for nausea and vomiting; 9.6%, 9.5% and 9.7% for urocystitis; and 63.5%, 69.0% and 65.3% enteritis and rectitis. There were no statistically significant differences among them (all P>0.05). The five-year disease-free survival rates (DFS) in groups 1, 2, 3 were 78.3%, 75.1%, 80.9%, respectively; the five-year overall survival rates (OS) were 81.4%, 78.2%, and 81.1%, respectively. The five-year OS of 166 patients receiving postoperative in the three groups were 72.4%, 69.5%, and 71.8%, respectively, with no significant variation (all P>0.05). Although there were no differences among three groups in DFS and OS, preoperative neoadjuvant chemotherapy combined with intracavitary radiotherapy may increase the effective rate and the percentage of patients with no postoperative risk factors and decrease the percentage of patients receiving postoperative adjuvant therapy, thereby decreasing complications indirectly and increasing quality of life.

      • Use of an Artificial Neural Network to Construct a Model of Predicting Deep Fungal Infection in Lung Cancer Patients

        Background: The statistical methods to analyze and predict the related dangerous factors of deep fungal infection in lung cancer patients were several, such as logic regression analysis, meta-analysis, multivariate Cox proportional hazards model analysis, retrospective analysis, and so on, but the results are inconsistent. Materials and Methods: A total of 696 patients with lung cancer were enrolled. The factors were compared employing Student's t-test or the Mann-Whitney test or the Chi-square test and variables that were significantly related to the presence of deep fungal infection selected as candidates for input into the final artificial neural network analysis (ANN) model. The receiver operating characteristic (ROC) and area under curve (AUC) were used to evaluate the performance of the artificial neural network (ANN) model and logistic regression (LR) model. Results: The prevalence of deep fungal infection from lung cancer in this entire study population was 32.04%(223/696), deep fungal infections occur in sputum specimens 44.05%(200/454). The ratio of candida albicans was 86.99% (194/223) in the total fungi. It was demonstrated that older (${\geq}65$ years), use of antibiotics, low serum albumin concentrations (${\leq}37.18g/L$), radiotherapy, surgery, low hemoglobin hyperlipidemia (${\leq}93.67g/L$), long time of hospitalization (${\geq}14$days) were apt to deep fungal infection and the ANN model consisted of the seven factors. The AUC of ANN model($0.829{\pm}0.019$)was higher than that of LR model ($0.756{\pm}0.021$). Conclusions: The artificial neural network model with variables consisting of age, use of antibiotics, serum albumin concentrations, received radiotherapy, received surgery, hemoglobin, time of hospitalization should be useful for predicting the deep fungal infection in lung cancer.
