Assessment of In Vitro Assay System for Thyroid HormoneDisruptors Using Rat Pituitary GH3 Cells

The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

This paper describes a web-based information system for plant disease forecast that was developed for crop growers in Gyeonggi-do, Korea. The system generates hourly or daily warnings at the spatial resolution of $240\;m{\times}240\;m$ based on weather data. The system consists of four components including weather data acquisition system, job process system, data storage system, and web service system. The spatial resolution of disease forecast is high enough to estimate daily or hourly infection risks of individual farms, so that farmers can use the forecast information practically in determining if and when fungicides are to be sprayed to control diseases. Currently, forecasting models for blast, sheath blight, and grain rot of rice, and scab and rust of pear are available for the system. As for the spatial interpolation of weather data, the interpolated temperature and relative humidity showed high accuracy as compared with the observed data at the same locations. However, the spatial interpolation of rainfall and leaf wetness events needs to be improved. For rice blast forecasting, 44.5% of infection warnings based on the observed weather data were correctly estimated when the disease forecast was made based on the interpolated weather data. The low accuracy in disease forecast based on the interpolated weather data was mainly due to the failure in estimating leaf wetness events.
KRAS targeting antibody synergizes anti-cancer activity of gemcitabine against pancreatic cancer

<P><B>Abstract</B></P> <P>Pancreatic cancer exhibits an oncogenic KRAS mutation rate of ∼90%. Despite research and drug development efforts focused on KRAS, no targeted therapy has been clinically approved for the treatment of pancreatic cancer with KRAS mutation. Also, the efficacy of gemcitabine is poor due to rapidly acquired resistance. We developed RT11-i antibody, which directly targets the intracellularly activated GTP-bound form of oncogenic RAS mutants. Here, we investigated the combined effects of RT11-i and gemcitabine <I>in vitro</I> and <I>in vivo</I>, and the mechanism involved. RT11-i significantly sensitized pancreatic cancer cells to gemcitabine. Also, the co-treatment synergistically inhibited angiogenesis, migration, and invasion, and showed synergistic anticancer activity by inhibiting the RAF/MEK/ERK or PI3K/AKT pathways. Furthermore, co-treatment inhibited endothelial barrier disruption in tumor vessels, which is a critical step in vascular leakiness of metastasis, and improved vessel structural stability. Importantly, co-treatment significantly suppressed tumor growth in an orthotopic tumor model. Taken together, our findings show that RT11-i synergistically increased the antitumor activity of gemcitabine by inhibiting RAS downstream signaling, which suggests RT11-i and gemcitabine be viewed a potential combination treatment option for pancreatic cancer patients with KRAS mutation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Pancreatic cancer exhibits an oncogenic KRAS mutation rate of ∼90%. Mixed connective tissue disease diagnosed by high serum level of anti-U1 RNP antibody

Mixed connective tissue disease (MCTD) is a disorder characterized by the overlap of features of systemic lupus erythematosus, scleroderma, and dermatomyositis. Laboratory findings of MCTD show distinctively high titer of anti-U1 RNP antibody, high titer of anti nuclear antibody (ANA), and elevation of creatine phosphokinase (CPK). And histopathologically, thinning and flattening of the epidermis, dermal mucin deposition, and predominantly lymphoplasmacytic infiltrate arranged along the dermal-epidermal junction, around hair follicles and eccrine glands can be seen. A 20-year-old female presented with erythematous diffuse variable sized patches and plaques on the face and erythematous to brownish discrete plaques and erosions on the both hands and feet. In laboratory test, anti-U1 RNP antibody was elevated more than 8.0 AI (ref.: < 1.0 AI), ANA was elevated more than 1:1280 and CPK was elevated by 244 U/L (ref.: 33-211 U/L). Histopathologic findings showed epidermal atrophy, pronounced dermal edema and perifollicular inflammatory cell infiltrate. On the basis of these clinical, laboratory, and histological findings, the diagnosis of MCTD was made. The patient was treated with methylprednisolone, hydroxychloroquine, and pentoxifylline, resulting in a significant improvement in skin lesions. Herein, we report a case of MCTD for whom high serum level of anti-U1 RNP antibody was used as an helpful diagnostic test.

<P>In the present study, we found that alpha-alumina hollow nanoshell structure can exhibit an ultrahigh fracture strength even though it contains a significant number of nanopores. By systematically performing in situ mechanical testing and finite element simulations, we could measure that the fracture strength of an alpha-alumina hollow nanoshell structure is about four times higher than that of the conventional bulk size alpha-alumina. The high fracture strength of the alpha-alumina hollow nanoshell structure can be explained in terms of conventional fracture mechanics, in that the position and size of the nanopores are the most critical factors determining the fracture strength, even at the nanoscales. More importantly, by deriving a fundamental understanding, we would be able to provide guidelines for the design of reliable ceramic nanostructures for advanced GaN light-emitting diodes (LEDs). To that end, we demonstrated how our ultrastrong alpha-alumina hollow nanoshell structures could be successfully incorporated into GaN LEDs, thereby greatly improving the luminous efficiency and output power of the LEDs by 2.2 times higher than that of conventional GaN LEDs.</P>

One objective of soybean breeders is to develop cultivars with elevated oleic acid content. Testing soybean seeds for oleic acid content is possible using gas chromatography (GC), however it is time-consuming and requires destructing the seeds. Using single seeds, we developed a near-infrared reflectance spectroscopy (NIR) calibration equation relating oleic acid measured with GC to oleic acid predicted by NIR. The slope of the regression line of oleic acid measured with GC on oleic acid predicted by NIR and the intercept was not different from zero. A set of 300 soybean seeds was used to calculate the NIR equation and an independent set of 100 soybean seeds was used for validation. This NIRS equation showed significant correlation between reference values and NIRS estimated values based on the SEP, r2, and the RSP of reference data to SEP. This research shows that NIR prediction of oleic acid using intact soybean seeds is accurate and rapid and should be especially useful for early generation screening.
Optimization of Drying Temperature and Time for Pork Jerky Using Response Surface Methodology

Response surface methodology (RSM) was applied to determine the optimum drying conditions for pork jerky. The physicochemical properties of pork jerky, such as final moisture content, water activity (Aw), pH, and shear force were investigated. In addition, sensory characteristics of pork jerky were evaluated and were used as a parameter for determining the optimum condition. Pork jerky samples were dried at different temperatures between 40 to 80℃ for the time ranged from 0 to 10 hr. The predicted values for moisture content, Aw, and shear force of dried pork samples were in good agreement with the experimental values with correlation coefficients (R²) of 0.95, 0.96, and 0.97, respectively. Both drying temperature and time significantly (p<0.01) affected moisture content, Aw, pH, and shear force and their interactions were also significant at p<0.01 except for Aw. RSM showed the optimum drying conditions for pork jerky, based on moisture content, shear force, and sensory evaluation to be 65-70℃ for 7-8 hr.
Metaplastic Spinal Meningioma in the Thoracic Spine: A Case Report and Review of the Literature

Spinal meningiomas account for 12% of all meningiomas and are usually located in the intradural extramedullary space. Among them, spinal metaplastic meningiomas are extremely rare. This is a case report on a 59-year-old man with thoracic spinal metaplastic meningioma. This male presented with progressively worsening right leg weakness, radiating pain and paresthesia. We present a rare case of pathologically confirmed metaplastic meningioma in the thoracic spine. Additionally, we review the pathological aspects and radiographic features of spinal metaplastic meningioma.
