Hypoallergenic and Physicochemical Properties of the A2 β-Casein Fraction of Goat Milk

Goat milk has a protein composition similar to that of breast milk and contains abundant nutrients, but its use in functional foods is rather limited in comparison to milk from other sources. The aim of this study was to prepare a goat A2 β-casein fraction with improved digestibility and hypoallergenic properties. We investigated the optimal conditions for the separation of A2 β-casein fraction from goat milk by pH adjustment to pH 4.4 and treating the casein suspension with calcium chloride (0.05 M for 1 h at 25°C). Selective reduction of β- lactoglobulin and αs-casein was confirmed using sodium dodecyl sulphate-polyacrylamide gel electrophoresis and reverse-phase high-performance liquid chromatography. The hypoallergenic property of A2 β-casein fraction was examined by measuring the release of histamine and tumor necrosis factor alpha from HMC-1 human mast cells exposed to different proteins, including A2 β-casein fraction. There was no significant difference in levels of both indicators between A2 β-casein treatment and the control (no protein treatment). The A2 β-casein fraction is abundant in essential amino acids, especially, branched-chain amino acids (leucine, valine, and isoleucine). The physicochemical properties of A2 β-casein fraction, including protein solubility and viscosity, are similar to those of bovine whole casein which is widely used as a protein source in various foods. Therefore, the goat A2 β-casein fraction may be useful as a food material with good digestibility and hypoallergenic properties for infants, the elderly, and people with metabolic disorders.

The development of in vitro assays has been recommended to screening and test-ing the potential endocrine disruptors (EDs). These assay systems focus only on identifying thethe thyroid hormone (TH) disruptors. The aim of this study was to evaluate a test system to detectTH disruptors using rat pituitary tumor GH3 cells. The test system is based on the TH-dependentincrease in growth rate. As expected, L-3,5,3-triiodothyronine (T3) markedly induced a morphologicalchange in GH 3 cells from flattened fibroblastic types to rounded or spindle-shaped types. T3 stimu-lated GH3 cell growth in a dose-dependent manner with the maximum growth-stimulating effect9 M. In addition, T3 increased the release of growth hor-mone and prolactin into the medium of the GH3 cells culture. Using this assay system, the TH-dis-rupting activities of bisphenol A (BPA) and its related compounds were examined. BPA,dimethylbisphenol A (DMBPA), and TCI-EP significantly enhanced the growth of GH3 cells in therange of 1 × 10-5M to 1 × 10-6M concentrations. In conclusion, this in vitro assay system might bestandardization before it can be used as a broad-based screening tool.

Background: Adipose tissue is recently recognized as not only energy reservoir but also endocrine organ producing proin ammatory cytokines. Especially the impact o visceral adipose tissue(VAT) in critical illnesses has been proposed, but researches on the association between VAT and sepsis were scarce and quantitative measurement of VAT had not been used. Methods: In this retrospective cohort study, we enrolled patients with sepsis who examined abdominal computed tomography(A-CT) within 1 month of occurrence of sepsis, among the patients admitted to intensive care unit(ICU). Age, sex, anthropometric values, comorbidities and APACHE II score were reviewed. The areas of VAT and total adipose tissue(TAT) on the section of A-CT image of the umbilicus level were measured by calculating pixels presenting fat density. Results: Among 287 patients admitted to ICU due to sepsis, 178 patients were included for this study. Median age was 65 and 59.0% were men. In-hospital mortality rate was 57.9%. Women had higher TAT and subcutaneous adipose tissue, lower VAT/TAT ratio compared to men. The amount of VAT and VAT/TAT ratio were higher in in-hospital mortality group than in survivor group(90.41cm2 vs. 63.83cm2 and 45.88% vs. 32.79%, p=0.001 and <0.001, respectively). After adjusting age, sex, APACHE II score and comorbidities, multivariate logistic regression analysis revealed that the amount of VAT and VAT/TAT ratio were independent prognostic factors of sepsis with obvious dose-dependent relationship(VAT/TAT ratio quartile 3: OR 8.832, p<0.001 and quartile 4: OR 29.477, p<0.001, compared to quartile 1 respectively). Conclusion: The amount of VAT and VAT/TAT ratio quantitatively measured by A-CT were positively correlated with mortality in sepsis, and this association was dose-dependent. Visceral obesity should be considered as the poor prognostic factor of sepsis.

<P><I>Lilium</I> bulbs have long been used as Chinese traditional medicines to alleviate the symptoms of various human inflammatory diseases. However, mechanisms of <I>Lilium</I> bulb-mediated anti-inflammatory activity and the bioactive components in <I>Lilium</I> bulbs remain unknown. In the present study, the anti-inflammatory activity of <I>Lilium</I> bulbs and the underlying mechanism of action were investigated in macrophages using <I>Lilium</I> bulb ethanol extracts (Lb-EE). In a dose-dependent manner, Lb-EE inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells and bone marrow-derived macrophages (BMDMs) without causing significant cytotoxicity. Lb-EE also down-regulated mRNA expression of inflammatory genes in LPS-stimulated RAW264.7 cells, which included inducuble nitric oxide synthase (iNOS), cyclooxygenase-2 (COX2), and tumor necrosis factor-<TEX>$ \alpha $</TEX> (TNF-<TEX>$ \alpha $</TEX>). Furthermore, Lb-EE markedly restored LPS-induced morphological changes in RAW264.7 cells to a normal morphology. HPLC analysis identified quercetin, luteolin, and kaempferol as bioactive components contained in Lb-EE. Mechanistic studies in LPS-stimulated RAW264.7 cells revealed that Lb-EE suppressed MyD88- and TRIF-induced NF-<TEX>$ \kappa $</TEX>B transcriptional activation and the nuclear translocation of NF-<TEX>$ \kappa $</TEX>B transcription factors. Moreover, Lb-EE inhibited IKK<TEX>$ \alpha $</TEX>/<TEX>$ \beta $</TEX>-induced activation of the NF-<TEX>$ \kappa $</TEX>B signaling pathway and IKK inhibition significantly reduced NO production in LPS-stimulated RAW264.7 cells. Taken together, these results suggest that Lb-EE plays an anti-inflammatory role by targeting IKK<TEX>$ \alpha $</TEX>/<TEX>$ \beta $</TEX>-mediated activation of the NF-<TEX>$ \kappa $</TEX>B signaling pathway during macrophage-mediated inflammatory responses.</P>

<P>The weakly coordinating ketone group directed C-H functionalizations of chromones, 1,4-naphthoquinones, and xanthones with various maleimides under rhodium(III) catalysis are described. These protocols efficiently provide a range of succinimide-containing chromones, naphthoquinones, and xanthones with excellent site selectivity and functional group compatibility. All synthetic compounds were screened for in vitro anticancer activity against human breast adenocarcinoma cell lines (MCF-7). In particular, compounds 7aa and 7ca with a naphthoquinone scaffold were found to be highly cytotoxic, with an activity competitive with anticancer agent doxorubicin.</P>
The Study on Androconia of Pieridae in Korea (I)

한국산 흰나비과 중 채집된 11종을 재료로 하여 각 종의 Androconia의 유무와 그 형태를 비교 검토하였다. Androconia는 Pieris rapae, P. napi, P. melete, P. canidia등 4종의 수컷에서만 관찰되었는데 반드시 앞, 뒷날개의 윗면에만 존재하고 있었다. 날개에서의 분포양상은 4종이 거의 유사하였으며 배열상태는 대부분 규칙적이었고 형태에 있어서는 4종이 약간의 차이점을 보여 주었다. Author has tried to find the presence of androconia at the eleven species within Pieridae in Korea. As a result, androconia was found at the four species among them Pieris rapae Linne, Pieris napi Linne, Pieris melete Menetries and Pieris canidia Sparrman. Andorconia was observed at the upper face of the fore and hind wings of male, not female. There is a similarity one another in the androconia distribution phase, and a slight difference in androconia size.

The objective of this study was to evaluate the effect of absorbable atelocollagen sponge on healing of hard tissue of buccal dehiscence defects in mongrel dogs. The mandibular second and fourth premolars of 12 dogs were extracted. Group 1 and Group 2 consisted of dogs without and with an inserted atelocollagen sponge after extraction with buccal plate preservation ; Group 3 and Group 4 consisted of dogs without and with an inserted atelocollagen sponge after removing two thirds of the buccal wall. At 10 weeks, Group 4 presented a significantly greater increase in the height of the alveolar bone crest than that of Group 3. Histologically, Group 2 and Group 4 showed better new bone formation ability than Groups 1, 3 at 6 weeks after extraction. The new bone formation ability of Group 3 was the poorest at 10 weeks. The results showed that the transplantation of absorbable atelocollagen sponge in the extraction socket can promote new bone formation and compensate marginal ridge reconstruction.
