The antigenic potential of CFA-001, cefazolin, a cephalosporin derivative produced by an enzymatic semisynthesis, was determined in Hartley guinea pigs. A battery of tests employed consisted of active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA), and indirect hemagglutination test (IHA). The results were as follows: 1) In ASA, no signs attributable to anaphylaxis was observed in guinea pigs sensitized with CFA-001, whereas OVA-sensitized animals induced severe anaphylactic symptoms; 2) guinea pigs did not produce antibodies against CFA-001 when sensitized with or without Freund's complete adjuvant (FCA) in homologous PCA tests. Meanwhile, antibodies against ovalbumin (OVA) were clearly detected; 3) No CFA-001-specific hemagglutination was observed in the IHA using sera obtained from CFA-001-sensitized guinea pigs. These results suggest that CFA-001 has no antigenicity potential in guinea pigs.

Liver transplantation with preservation of the recipient vena cava (piggyback technique) has been performed as an alternative to the conventional method. Outflow disturbance or obstruction of the vena cava in the early period after liver transplantation is associated with high morbidity and mortality. We used side-to-side cavo-caval anastomosis (modified piggyback technique) in a deceased-donor liver transplantation (DDLT) for venous outflow reconstruction. On postoperative day 9, the patient developed abdominal discomfort, and abnormal liver function showing serum total bilirubin of 6.2 mg/dl and serum AST/ALT of 297/597 IU/L. Doppler ultrasound showed mono-phasic wave forms of the hepatic vein. Computed tomography showed focal narrowing of 9.5 mm±2 mm in diameter at the cavo-caval anastomosis site. Liver biopsy was showed that there was no evidence of acute allograft rejection. Direct venogram showed stenosis of the cavo-caval anastomosis with a pressure gradient of 12 mmHg. An interventional stent was inserted in the stenotic site of the inferior vena cava, and the pressure gradient decreased to 2 mmHg. He was discharged from hospital on postoperative day 23 without any other complications. Herein we report a case of deceased-donor liver transplantation using the modified piggyback technique, who received an inferior vena cava stent due to stricture of the reconstructed orifice of the vena cava. The water jetting system for a jack-up spudcan requires the suitable design considering the platform/spudcan particulars, environments, and soil conditions, either the surficial clay or surficial sand. The usage of water jetting depends critically on soil conditions. The water jetting is usually used for the smooth and fast extraction of the spudcan in the surficial clay condition. It is also required for inserting spudcan up to the required depth in the surficial sand condition, which is investigated in this paper. Especially, it should be very careful to use the water jetting during an installation of spudcan in the surficial sand condition, because there is a risk of overturning accident related to the punch-through. Therefore, in this study, the effect of water jetting flow rate and time on the change of soil properties and penetration resistance is analyzed to better understand their interactions and correlations when inserting the spudcan with water jetting in surficial sand condition. For the investigation, a wind turbine installation jack-up rig (WTIJ) is selected as the target platform and the multi layered soil (surficial sand overlaying clays) is considered as the soil condition. The environmental loading and soil-structure interaction (SSI) analysis are performed by using CHARM3D and ANSYS. This kind of investigation and simulation is needed to decide the proper water jetting flow rate and time of spudcan for the given design condition.

KR-33028 (N-[4-cyano-benzo[b]thiophene-2-carbonyl]guanidine) is a new cardioprotective agent for preventing ischemia-reperfusion injury. This study was performed to identify the metabolic pathway of KR-33028 in human liver microsomes and to compare its metabolism with that of cryopreserved human hepatocytes. Human liver microsomal incubation of KR-33028 in the presence of NADPH and UDPGA resulted in the formation of four metabolites, M1, M2, M3, and M4. M1 and M2 were identified as 5-hydroxy-KR-33028 and 7-hydroxy-KR-33028, respectively, on the basis of LC/MS/MS analysis with the synthesized authentic standard. M3 and M4 were suggested to be dihydroxy-KR-33028 and hydroxy-KR-33028-glucuronide, respectively. Metabolism of KR-33028 in cryopreserved human hepatocytes resulted in the formation of M1, M2, and M4. These data show a good correlation between major metabolites formed in human liver microsomes and cryopreserved human hepatocytes. In addition, KR33028 was found to inhibit moderately the metabolism of CYP1A2 substrates. Based on the results obtained metabolic pathway of KR-33028 is proposed.

The G protein-activated inwardly rectifying $K^+$ channel (GIRK1) was coex-pressed in Xenopus oocytes along with the $5-HT_{1A}$ receptor, a 7-helix receptor known to be coupled to $K^+$ channels in many neural tissues. Thus, the activation of the $5-HT_{1A}$ receptor by its agonist leads to the opening of GIRK1. The GIRK1 current was measured using the two electrode voltage clamp technique with bath application of 5-HT in the presence of various external potassium concentrations $[K^+]_0$. GIRK1 showed a strong inward rectification since only hyperpolarizing voltages evoked inward currents. $K^{+}$ was the major ion carrier as evidenced by about 44㎷ voltage shift corresponding to a 10-fold external 〔$K^+$〕 change. 5-HT induced a concentration-dependent inward $K^+$ current ($EC_{50}{\equation omitted}10.7nM$) which was blocked by $Ba^{2+}$. Pertussis toxin (PTX) pre-treatment reduced the $K^+$ current by as much as about 70%, suggesting that PTX-sensitive G protein ($G_i or G_o$ type) are involved in the $5-HT_{1A}$ receptor-GIRK1 coupling in Xenopus oocytes.

The spudcan requires the suitable design considering the soil, platform, and environmental conditions. Its shape needs to be designed to secure sufficient reaction of soil so that it can prevent overturning accidents. Its shape also has to minimize the installation and extraction time. Even in the same soil condition, the reaction of soil may be different depending on the shape of spud can, mainly the slope of top and bottom plates. Therefore, in this study, the relation between the slope of plates and the reaction of soil with and without water jetting is analyzed to better understand their interactions and correlations. For the investigation, a wind turbine installation jack-up rig (WTIJ) is selected as the target platform and the Gulf of Mexico is considered as the target site. A multi layered (sand overlying two clays) soil profile is applied as the assumed soil condition and the soil-structure interaction (SSI) analysis is performed by using ANSYS to analyze the effect of the slope change of the bottom plate and water jetting on the reaction of soil. This kind of investigation and simulation is needed to develop optimal and smart spudcan with water-jetting control in the future.

We have investigated a Mo/Au gate scheme for use in AlGaN/GaN-on-Si HFETs. AlGaN/GaN-on-Si HFETs were fabricated with Ni/Au or Mo/Au gates and their electrical characteristics were compared after thermal stress tests. While insignificant difference was observed in DC characteristics, the Mo/Au gate device exhibited lower on-resistance with superior pulsed characteristics in comparison with the Ni/Au gate device.
Single dose pharmacokinetics of the novel transdermal donepezil patch in healthy volunteers

<P><B>Background</B></P><P>Donepezil is an acetylcholinesterase inhibitor indicated for Alzheimer’s disease. The aim of this randomized, single-blind, placebo-controlled, single-dose, dose-escalation study was to investigate the safety, tolerability, and pharmacokinetics of the donepezil patch in healthy male subjects.</P><P><B>Methods</B></P><P>Each healthy male subject received a single transdermal donepezil patch (72 hours patch-on periods) of 43.75 mg/12.5 cm<SUP>2</SUP>, 87.5 mg/25 cm<SUP>2</SUP>, or 175 mg/50 cm<SUP>2</SUP>. Serial blood samples were collected up to 312 hours after patch application. The plasma concentrations of donepezil were determined by using a validated liquid chromatography–tandem mass spectrometry method. Pharmacokinetic parameters were obtained by noncompartmental analysis. Tolerability of the patches and performance of the patches (adhesion, skin irritation, residual donepezil content in the patch) were assessed throughout the study.</P><P><B>Results</B></P><P>The study was completed by 36 healthy subjects. After patch application, the maximal plasma donepezil concentration (C<SUB>max</SUB>) and the area under the curve (AUC) increased in a dose-proportional manner. Median time to C<SUB>max</SUB> was ~74–76 hours (~2–4 hours after patch removal), and mean t<SUB>1/2β</SUB> was ~63.77–93.07 hours. The average donepezil residue in the patch after 72 hours was ~73.9%–86.7% of the loading dose. There were neither serious adverse events nor adverse events that lead to discontinuation. Skin adhesion of the patch was good in 97.2% of the subjects. All skin irritations after patch removal were mild and were resolved during the study period.</P><P><B>Conclusion</B></P><P>The donepezil patch appeared to be generally well tolerated and adhesive. Pharmacokinetic analysis of the donepezil patch demonstrated linear kinetics.</P>
