        Association analysis of polymorphisms of G protein-coupled receptor 54 gene exons with reproductive traits in Jiaxing Black sows

        Objective: The aim of this study was to detect single nucleotide polymorphisms (SNP) of G protein-coupled receptor 54 (GPR54) gene and explore association of this candidate gene with reproductive traits in Jiaxing Black sows. Methods: Six pairs of primers of the gene were designed to amplify all exons thus sequences of which were detected by means of direct sequencing and then SNP loci were scanned. The effects of SNPs on total number of piglets born (TNB), number of piglets born alive (NBA), number of still born piglets (NSB), and litter weight at birth (LWB) of Jiaxing Black sows were analyzed. Results: Three SNP loci, including T3739C, C3878T and T6789C, were identified via comparison of sequencing and two genotypes (AB, BB) at each SNP site were observed. T3739C resulted in the change of amino acid (Leu→Pro) in corresponding protein, and C3878T resulted in synonymous mutation (Ile→Ile). Statistical results demonstrated that allele B was the preponderant allele at the three SNP loci and Genotype BB was the preponderant genotype. Meanwhile, Chi-Square test of these three SNPs indicated that all mutation sites fitted in Hardy-Weinberg equilibrium (p>0.05). For GPR54-T3739C locus, Jiaxing Black sows with genotype BB had 1.23 TNB and 1.28 NBA (p<0.01) that were more than those with genotype AB, respectively. Jiaxing Black sows that had the first two parities with genotype BB had additional 2.23 TNB, 2.27 NBA (p<0.01), and 1.94 LWB (p<0.05) compared to those with genotype AB, respectively. However, for other two loci, no significant difference was found between TNB, NBA, NSB, and LWB, and different genotypes of Jiaxing Black sows. Conclusion: In conclusion, the polymorphisms of GPR54-T3739C locus were significantly associated to TNB, NBA, and LWB and could be used as a potential genetic marker to improve reproductive function of Jiaxing black sows.

        Research on the Application of Blockchain Technology in the Comprehensive Health Industry

        Blockchain technology is widely used in finance, Internet and other fields, and has broad prospects. With the improvement of blockchain technology research, its application in the medical field has also made great progress. Many people believe that the healthcare field is the second largest blockchain application research field after the financial field. Whether the medical and health services are well developed is related to the country's economy and people's lives, and to the health of every citizen. It is also an important measure for the country to effectively deal with the problem of aging. China's policy has also released positive signals in the construction of the big health industry system. It encourages relevant institutions to give full play to the role of network technology and build an "Internet + medical and health" platform to make medical services more convenient. This paper mainly adopts the literature review method to explore the application of blockchain technology in the field of comprehensive health. It discusses the application ideas and systems of blockchain in the health care field by analyzing the core advantages of blockchain technology, the health industry development status, and combining with the current problems in the health care sector. It proposes three applications of blockchain technology in the health field: building a personal health database, an intelligent medical assistance platform, and an open medical resource mutual assistance and sharing center. This paper explores that blockchain technology can help hospitals, medical insurance, medicine, research institutions and other participating entities to share medical data safely and efficiently, improve the efficiency and quality of health care services, increase patients' sense of experience and autonomy, and realize the structure of the health industry. It aims promote the blockchain technology to better serve the health care department and individuals.

        Bibliometric analysis of publications on inclusive education from the Web of Science Core Collection published from 1992 to 2020

        Purpose This study analyzed the bibliometric characteristics of publications on inclusive education in the Social Science Citation Index and Science Citation Index Expanded in the Web of Science Core Collection from 1992 to 2020. Methods Terms related to “inclusive education” and “inclusion of education” were used as keywords to search for journal articles on July 3, 2020. Results There were 1,786 articles, representing 3,376 authors, in the 345 journals scanned. The United States, United Kingdom, and Australia were the three leading countries/regions in this field. In the top 12 countries, the top 15 institutions and the top 10 most-cited journals were identified by either the number of publications or the number of total citations. Core themes from the 30 most highly-cited articles were teachers’ attitudes, teachers’ self-efficacy, and the effects of inclusive education. Teachers included both pre-service and in-service teachers; students represented those with and without special educational needs. Conclusion The results indicate that the United States, United Kingdom, and Australia dominated inclusive education research, originating most of the highly-cited articles, having more prolific authors, and presenting the most-cited institutions. Furthermore, three emerging core themes from the 30 most highly-cited articles were teachers’ attitudes, teachers’ self-efficacy, and the effects of inclusive education. Frontline teachers are recommended to submit manuscripts about their teaching experiences to the most-cited journals, which have a large readership. To measure the effects of inclusive education, it is essential to formulate reliable, valid, and culture-free research instruments for future studies.

        Vocational Education System Architecture based on Blockchain Technology

        With China's social and economic development, it has put forward various strategies for talent development, and all walks of life urgently need technical and skilled talents, which make vocational education more and more important. However, there are some urgent problems to be solved to develop China's vocational education, such as asymmetric information and imperfect certification and evaluation system. Although the state implements the "1+X" certificate system that is suitable for the training model of compound technical and skilled personnel in the new era, in the implementation process, there are insufficient anti-counterfeiting capabilities of academic certificates and vocational skills certificates, insufficient participation of multiple subjects, data security of credit banks and lower business efficiency. The blockchain technology not only contains the governance concept, but also has extremely high security and confidentiality performance. It can promote the linkage of education subjects and the integration of production and education, and can also be highly integrated and applied with intelligent technology. Therefore, it is very valuable in promoting and implementing of the new system. Through case analysis, comparative research, and summarization, this paper builds a vocational education system ground on blockchain technology with the help of blockchain-related features, smart contracts, decentralization, traceability, and non-tampering. This study integrates blockchain technology with certification evaluation system, resource circulation system, and learning credit system. With the application concept of vocational education resource database, decentralized certificate certification and evaluation system, online learning community based on "vocational learning currency", and intelligent learning materials and service trading platform are proposed. This study provides reference for vocational education system reform and sustainable development.

        Comparison of single-drop microextraction with microvolume pipette extraction directly coupled with capillary electrophoresis for extraction and separation of tricyclic antidepressant drugs

        This paper describes the use of single-drop microextraction and micro-volume pipette extraction coupled with capillary electrophoresis for the analysis of five antidepressant drugs (nortriptyline, imipramine, trimeprazine, promethazine, and iminodibenzyl). The five antidepressant drugs are well separated by using micellar electrokinetic capillary chromatography with SDS and organic modifiers. The best extraction was achieved within 10 min and effectively separated within 15 min by using ammonium acetate buffer at 28 kV. This method shows good reproducibility, with RSD values ranging from 4.3 to 8.7%. The LODs values were found to be 0.4 and 1.0 mg mL-1 for SDME-CE and 0.15 and 0.5 mg mL-1 for MVPE-CE. These methods are simple, rapid, sensitive and efficient for the extraction, separation and detection of tricyclic antidepressant drugs.

        Soft Ionization of Metallo-Mefenamic Using Electrospray Ionization Mass Spectrometry

        Detection of mefenamic acid (M, non-steroidal anti-inflammatory drug, NSAIDs) and its metallodrug was investigated using electrospray ionization mass spectrometry (ESI-MS) and fluorescence spectroscopy. ESI-MS data (500 μL, 1×10-3 M) revealed high detection sensitivity for the drug and metallodrug. ESI-MS spectra revealed peaks at 242, 580, and 777 Da corresponding to [M+H]+, [63Cu(M-H)2(H2O)2+H]+, and [56Fe(M-H)3+H]+, respectively. The metal:mefenamic ratios of ESIMS spectra are in complete agreement with the fluorescence spectroscopy results (1:2 for Cu(II) and 1:3 for Fe(III)). ESI is a soft ionization technique that can be used on labile metallo-mefenamic acids and is promising for the detection of these species in environmental samples and biological fluids.

        The effect of pharmacist-led medication therapy management in the multidisciplinary care of acute kidney injury survivors

        Background: The Acute Disease Quality Initiative advocates multidisciplinary care for the survivors of acute kidney injury (AKI). The bundled care strategy recognizes the role of pharmacists. However, their specific contributions in this context remain underexplored. Methods: This retrospective study examined the efficacy of pharmacist-led post-AKI pharmaceutical care in outpatient settings at a single center. Adults with recent AKI during hospitalization, maintaining an estimated glomerular filtration rate <45 mL/min/1.73 m2 postdischarge, were enrolled in a multidisciplinary team care program from March 2022 to January 2023, with a 6-month follow-up period. Pharmacist-delivered care adhered to international multidisciplinary consensus guidelines. Efficacy was evaluated by analyzing medication-related recommendations, medication adherence, nephrotoxic drug utilization, and renoprotective medication usage before and after the intervention. Results: A total of 40 patients were referred to the pharmacist-managed clinic. Of these, 33 patients (mean age, 63 ± 15 years; 60.6% male) attended the clinic. Nineteen patients completed follow-up visits. The pharmacist provided 14 medication-related recommendations to relevant physicians, with 10 of these recommendations (71.4%) being accepted. There was a significant decrease in the use of modifiable nephrotoxic drugs (p = 0.03). However, no significant improvements were noted in medication adherence or the utilization of renoprotective medications. Conclusion: Our study underscores the potential benefits of pharmacist-led post-AKI bundled care strategy in outpatient settings. We observed a significant reduction in the utilization of modifiable nephrotoxic drugs, indicating the effectiveness of pharmacist interventions in optimizing medication regimens to mitigate renal harm.

        Cellular and Molecular Mechanisms of Intestinal Fibrosis

        Intestinal fibrosis associated stricture is a common complication of inflammatory bowel disease usually requiring endoscopic or surgical intervention. Effective anti-fibrotic agents aiming to control or reverse intestinal fibrosis are still unavailable. Thus, clarifying the mechanism underpinning intestinal fibrosis is imperative. Fibrosis is characterized by an excessive accumulation of extracellular matrix (ECM) proteins at the injured sites. Multiple cellular types are implicated in fibrosis development. Among these cells, mesenchymal cells are major compartments that are activated and then enhance the production of ECM. Additionally, immune cells contribute to the persistent activation of mesenchymal cells and perpetuation of inflammation. Molecules are messengers of crosstalk between these cellular compartments. Although inflammation is necessary for fibrosis development, purely controlling intestinal inflammation cannot halt the development of fibrosis, suggesting that chronic inflammation is not the unique contributor to fibrogenesis. Several inflammation-independent mechanisms including gut microbiota, creeping fat, ECM interaction, and metabolic reprogramming are involved in the pathogenesis of fibrosis. In the past decades, substantial progress has been made in elucidating the cellular and molecular mechanisms of intestinal fibrosis. Here, we summarized new discoveries and advances of cellular components and major molecular mediators that are associated with intestinal fibrosis, aiming to provide a basis for exploring effective anti-fibrotic therapies in this field.

        Measurement of the radon and thoron exhalation rates from the water surface of Yixin lake

        The importance of determining the radon exhalation rate from water surface is emphasized by the increased use of radon and its daughter products as tracers in large-scale circulation studies of the atmosphere. There were many methods to measure radon exhalation from water surface. With the development of radon exhalation rate measurement methods and instruments on the surface of the soil, the rock and building materials, so the radon exhalation rate from water surface can be more accurately measured by applying these improved methods and instruments. In this paper, a cuboid accumulation chamber surrounded by foam boards and a RAD7 were used to measure the radon exhalation rate on the water surface at three different positions by Yixin lake. Each measurement was performed 2 h. The radon exhalation rate from the water surface was about 6 × 10<sup>-3</sup> Bq m<sup>-2</sup>s<sup>-1</sup>. The thoron exhalation rate from the water surface also can be estimated, it is about 0.16 Bq m<sup>-2</sup>s<sup>-1</sup>. These results hint that the radon transmission from the lake bottom soil to water and then into the atmosphere.

        Three homologous genes encoding functional D8-sphingolipid desaturase in Populus tomentosa

        Δ8-sphingolipid desaturase is characterized by itsability to catalyze desaturation at the C8 position of the longchainbase of sphingolipids in plants. No previous studieshave been conducted on genes encoding Δ8-sphingolipiddesaturases in the woody plant Populus tomentosa. In thisstudy, three genes that encode Δ8-sphingolipid desaturasewere isolated fromP. tomentosa. Among these genes, PtD8Aand PtD8B showed high sequence similarity; whereas PtD8Cexhibited large sequence divergence.RT-PCRresults showedthat PtD8A and PtD8B were expressed in all tissues detected,whereas PtD8C was not expressed in roots. Heterologousexpression in yeast revealed that PtD8A/B/C were functionalΔ8-sphingolipid desaturases, and can catalyze the C18-phytosphingeninedesaturation to produce 8(Z)- and 8(E)-C18-phytosphingenine.However, the conversion rate and ratios ofthe two products differed. Compared with control cells,transgenic yeasts expressing PtD8A/B/C exhibited enhancedaluminum tolerance. Our findings further elucidated thebiochemical functions and evolutionary history ofΔ8-sphingolipid desaturases in plants. Candidate genes forbreeding new poplar germplasm resources with enhancedtolerance ability to aluminium were also provided.
