        Contributed Mini Review : The uniqueness of the plant mitochondrial potassium channel

        The ATP-inhibited Plant Mitochondrial K+ Channel (PmitoKATP) was discovered about fifteen years ago in Durum Wheat Mitochondria (DWM). PmitoKATP catalyses the electrophoretic K+ uniport through the inner mitochondrial membrane; moreover, the co-operation between PmitoKATP and K+/H+ antiporter allows such a great operation of a K+ cycle to collapse mitochondrial membrane potential (ΔΨ) and ΔpH, thus impairing protonmotive force (Δp). A possible physiological role of such ΔΨ control is the restriction of harmful reactive oxygen species (ROS) production under environmental/oxidative stress conditions. Interestingly, DWM lacking Δp were found to be nevertheless fully coupled and able to regularly accomplish ATP synthesis; this unexpected behaviour makes necessary to recast in some way the classical chemiosmotic model. In the whole, PmitoKATP may oppose to large scale ROS production by lowering ΔΨ under environmental/oxidative stress, but, when stress is moderate, this occurs without impairing ATP synthesis in a crucial moment for cell and mitochondrial bioenergetics.         The UN 2030 Education Agenda is a global platform directed towards creating opportunities for learners with special educational needs (LSENs) for their holistic development and eventual mainstreaming in the society. The schools being the catalysts of societal transformation are focal points for provision of instruction and intervention that emphasize inclusion, equity and gender equality among learners. Policies, programs, activities as well teachers’ trainings and orientation must be crafted to address the proper mainstreaming and placement of LSENs that will prepare them for self-independence and enhancement of their skills while ensuring that incidents of gender bias and bullying is eliminated, thus creating a wholesome learning environment. The objectives of this paper are mainly focused on the following areas; emphasis on the level of awareness of the goals of UN 2030 Education Agenda; assess the level of effectiveness of initiatives and activities for implementation; point out the severity of the problems that hinder implementation. A plan of action reflected as roadmap in order to attain the UN 2030 Education Agenda was conceptualized. Descriptive-analytical method of research was employed with survey-questinnaire as its data-gathering instrument. Respondents were teachers and stakeholders of the eleven (11) special education centers of DepEd Division of Pangasinan II. From the data gathered, it was concluded that respondents have very high awareness of the goals of UN 2030 education agenda, the iniatives and activities to be undertaken were perceived very much effective while the problems were moderately severe. The findings indicated that principals should keep track of the activities being implemented to ensure alignment with the set of goals, stakeholders must work collaboratively to intensify implementation of the activities and that applicable solutions must be thought of to minimize the problems encountered. Hence, principals in unison with the teachers and stakeholders must religiously implement the plan of action to ensure achievement of the goals on providing inclusive, equitable and gender equality education for learners with special educational needs of DepEd, Pangasinan Division II, Philippines.

        Evidence for the Involvement of Potassium Channel Inhibition in the Antidepressant-Like Effects of Hesperidin in the Tail Suspension Test in Mice

        The administration of hesperidin elicits an antidepressant-like effect in mice by a mechanism dependent on an interaction with the l-arginine-nitric oxide (NO)-cyclic guanosine monophosphate (cGMP) pathway, whose stimulation is associated with the activation of potassium (K+ ) channels. Thus, this study investigated the involvement of different types of K+ channels in the antidepressant-like effect of hesperidin in the mice tail suspension test (TST). The intracerebroventricular administration of tetraethylammonium (TEA, a nonspecific blocker of K+ channels), glibenclamide (an ATP-sensitive K+ channel blocker), charybdotoxin (a large- and intermediate-conductance calcium-activated K+ channel blocker) or apamin (a small-conductance calcium-activated K+ channel blocker) combined with a subeffective dose of hesperidin (0.01 mg/kg, intraperitoneally [i.p.]) was able to produce a synergistic antidepressant-like effect in the mice TST. Moreover, the antidepressant-like effect elicited by an effective dose of hesperidin (0.3 mg/kg, i.p.) in TST was abolished by the treatment of mice with pharmacological compounds K+ channel openers (cromakalim and minoxidil). Results showed that the antidepressantlike effect of hesperidin in TST may involve, at least in part, the modulation of neuronal excitability through inhibition of K+ channels and may act through a mechanism dependent on the inhibition of l-arginine-NO pathway.

        The age-adjusted Charlson comorbidity index as a predictor of survival in surgically treated vulvar cancer patients

        Objective: To evaluate the impact of age-adjusted Charlson comorbidity index (ACCI) in predicting disease-free survival (DFS), overall survival (OS), and cancer-specific survival (CSS) among surgically treated patients with vulvar carcinoma. The secondary aim is to evaluate its impact as a predictor of the pattern of recurrence. Methods: We retrospectively evaluated data of patients that underwent surgical treatment for vulvar cancer from 1998 to 2016. ACCI at the time of primary surgery was evaluated and patients were classified as low (ACCI 0–1), intermediate (ACCI 2–3), and high risk (>3). DFS, OS and CSS were analyzed using the Kaplan-Meir and the Cox proportional hazard models. Logistic regression model was used to assess predictors of distant and local recurrence. Results: Seventy-eight patients were included in the study. Twelve were classified as low, 36 as intermediate, and 30 as high risk according to their ACCI. Using multivariate analysis, ACCI class was an independent predictor of worse DFS (hazard ratio [HR]=3.04; 95% confidence interval [CI]=1.54–5.99; p<0.001), OS (HR=5.25; 95% CI=1.63–16.89; p=0.005) and CSS (HR=3.79; 95% CI=1.13–12.78; p=0.03). Positive nodal status (odds ratio=8.46; 95% CI=2.13–33.58; p=0.002) was the only parameter correlated with distant recurrence at logistic regression. Conclusion: ACCI could be a useful tool in predicting prognosis in surgically treated vulvar cancer patients. Prospective multicenter trials assessing the role of ACCI in vulvar cancer patients are warranted.

        Fertility-sparing surgery for women with stage I cervical cancer of 4 cm or larger: a systematic review

        Objective: To investigate current evidence on oncological, fertility and obstetric outcomes of patients with stage I cervical cancer of 4 cm or larger undergoing fertility-sparing surgery (FSS). Methods: Systematic review of studies including women affected by stage I cervical cancer ≥4 cm who underwent FSS. Main outcome measures: disease-free survival (DFS), overall survival (OS), pregnancy rate, live birth rate, premature delivery rate. Results: Fifteen studies met all eligibility criteria for this systematic review, involving48 patients affected by cervical cancer ≥4 cm who completed FSS. Three patients (6.3%) experienced a recurrence and one of them (2.1%) died of disease. The 5-year DFS rate was 92.4%. The 5-year OS rate was 97.6%. A significantly shorter 5-year DFS was reported for high-risk patients (G3, non-squamous histotype, diameter ≥5 cm) compared with low-risk (74.7% vs. 100%; log-rank test, p=0.024). Data about fertility outcomes were available for 12 patients. Five patients out of 12 (41.7%) attempted to conceive with an estimated pregnancy rate of 80%, a live birth rate of 83.3% and a premature delivery rate of 20%. Conclusion: Women with high tumor grade, aggressive histology and tumor size ≥5 cm have a higher risk of recurrence. Oncologic outcomes are encouraging among low-risk patients; however, the lack of high-quality studies makes it difficult to draw any firm conclusions. Prospective multicentric clinical trials with a proper selection of inclusion/exclusion criteria should be conducted in women with low-risk factors, strong desire to preserve their fertility and high likelihood to conceive.

        Micro-Raman investigation of vanadium-oxide coated tubular carbon nanofibers for gas-sensing applications

        Commercial tubular carbon nanofibers were uniformly coated with a 5nm thick vanadium oxide layer via a modified approach to atomic layer deposition. The composition and microstructure of the resulting hybrid material was analyzed by micro-Raman spectroscopy. The effect of the post-synthesis thermal treatment in air at temperatures in the range of 25-375<SUP>o</SUP>C was investigated in order to more deeply understand the behavior of the hybrid material in gas sensing devices. The obtained results demonstrate that the thermal treatment primarily affects the oxide coating-layer that is responsible for the sensing properties. The best sensor performance was obtained at the temperature at which the oxide layered-structure exhibits the highest structural order.

        El objetivo del presente trabajo es mostrar la actualidad del sector textil y vestuario (Maquila) en Guatemala y la estrategia identificada. Dada la presencia de muchas empresas coreanas en dicho país, hay que seguir una estrategia coherente y sistemática para enfrentarse con el reto que significa la competencia asiática. A lo largo de estas páginas, vamos a observar la situación de Maquila de Guatemala. Para mantener la estabilidad del trabajo en Maquila, el actual gobierno de Guatemala congeló el salario mínimo en Maquila, así que la competencia de la mano de obra se mantiene, sin embargo, el nuevo ambiente del mercado global ha dado como resultado la desaceleración como vemos en las gráficas. Las pequeñas y medianas empresas representan la fuente del mercado laboral en Guatemala, así que conviene observar la tendencia del mercado textil y nuevo sistema después de la eliminació del Acuerdos de Textil y Vestuario de la Organización Mundial de Comercio.

        An Improved Spectrophotometric Phospholipase A2 Assay Using 1-Palmitoyl-2-Linoleoyl-sn-Glycero-3- Phosphatidylcholine as Substrate and Lipoxygenase as Coupled Enzyme

        An improved spectrophotometric assay of phospholipase A2 (PLA2) activity based on the coupled PLA2/lipoxygenase (LOX) reactions using 1-palmitoyl-2-linoleoyl-sn-glycero-3-phosphatidylcholine (PCLIN) as substrate is reported. The PLA2-mediated release of free linoleate is continuously monitored by following the absorbance increase at 234 nm caused by its conversion into the conjugated diene hydroperoxide catalyzed by the coupled soybean LOX-1 reaction. The new protocol includes the use of Tween 20 (3 μL/μmol phospholipid) as surfactant and of ethanol (15 μL/mL reaction mixture), that ensure clearness of reaction mixture and linear increase of absorbance in the course of reaction. This method was tested on a purified secretory PLA2from honey bee venom (HBV-PLA2). The enzyme did not discriminate among PCLIN, phosphatidylcholine, and phosphatidylethanolamine,but showed the highest rate using 1,2-dilinoleoyl-sn-glycero-3-phosphatidylcholine (PCDILIN). Nevertheless,the use of PCDILIN is not recommended, as it may induce an overestimation of enzyme activity, because not only the free linoleate, but also the reaction product 1-linoleoyl-lysophosphatidylcholine,are known to be oxidized by LOX. HBV-PLA2showed maximal activity at pH 9.0, hyperbolic kinetics (Km,74.2±2.9 μM; Vmax, 827±7 μmol/min/mg protein) and competitive inhibition (Ki about 5 μM) by palmityl trifluoromethyl ketone, a classical PLA2 inhibitor. Interestingly, the HBV-PLA2/soybean LOX-1 coupled reactions also allow an accurate assay of PCLIN concentration. In the whole, these results demonstrate that this improved PLA2/LOX assay allows a very accurate, simple, and rapid measurement of enzyme activity and substrate concentration.
