      • Arginine Methylation of CRTC2 Is Critical in the Transcriptional Control of Hepatic Glucose Metabolism

        <P><B>Getting a Sugar Rush from Arginine Methylation</B></P><P>Fasting causes the liver to produce and release glucose into the bloodstream, a process called gluconeogenesis. Fasting triggers the transcriptional activation of genes encoding gluconeogenic enzymes by CREB (cAMP response element–binding protein) and its coactivator CRTC2 (CREB-regulated transcription coactivator 2). Han <I>et al.</I> found that CRTC2 was targeted by the arginine methyltransferase PRMT6. In cells, arginine methylation of CRTC2 enhanced the association of CRTC2 with CREB and increased CREB activity. In mice, overexpression of PRMT6 increased the expression of genes encoding gluconeogenic enzymes and blood glucose concentrations, effects that required CRTC2. Obesity or insulin resistance leads to higher blood glucose concentrations, and acute deletion of PRMT6 restored blood glucose concentrations to normal in mice that were obese or insulin-resistant either through genetics or diet. Thus, hyperglycemia triggered by obesity or diet could be controlled by inhibiting the arginine methylation of CRTC2 by PRMT6.</P>

        체험적 환경조성을 통한 교육적 전시 콘텐츠로서의 생태박물관 인터랙티브 미디어 전시물 연구 -<Touch Touch Bugs>를 중심으로-

        본 논문은 전통적인 전시 연출 방식에서 벗어나지 못하는 국내 생태박물관들을 타겟으로 디지털 미디어 시대의 흐름에 맞춰 현대적 연출 방법을 제안한다. 이를 위해 생태박물관에 대한 이론적 고찰과 국내외 생태박물관의 현황을 분석하며 인터랙티브 미디어를 활용한 체험형 전시물을 제안하는데 목적을 둔다. 그리고 이러한 제안의 형식으로 프로토타입 전시물 작업<Touch Touch Bugs>를 제작한다. 이 작업은 생태박물관에서 전시되는 자연물 중 하나인 곤충이 생태 환경에서 성장 단계에 따른 정보를 자연 환경안에서 체험적으로 제공하기 위하여, 터치패드 기반의 게임과 월 디스플레이 두 가지 인터페이스로 구성된다. 이러한 과정은 일방적 전시물 형태를 벗어나 전시 관람자들로 하여금, 정보에 대한 이해와 학습을 함께 상호작용하는 과정을 유도하며 보다 흥미롭게 이끌고 생태적 환경과 생명체에 대한 유기적 이해를 돕게 하여 박물관에서의 자유로운 학습 효과를 도출해 낼 수 있다. This paper studies interactive media installations of ecological museums. Particularly this research focuses on the domestic ecological museums and how each museum develop its artefact contextually. The first two chapters show the theoretical background of this subject based on literature reviews for methodological approaches using interactive media and then analyze several good artefact examples from overseas museums. From this process, we found that modern digital and interactive media production can enhance its exhibition experience. Based on this theoretical studies, in the next chapter, we propose a prototype work of the ecological museum artefact, <Touch Touch Bugs>. This work provides the knowledge of bugs`` life and their ecological environment. Composed of two media interfaces, a game on touch-pad and a related background moving image on a wall-display, this work is to presenthow bugs live throughout their growing stages and in the optimized natural environments. Visitors may access to interactive experience through this artefact in the exhibition and this could help them understand their ecological environment and living organisms, thereby enhancing their understanding about the ecologies around our living world.

      • 사회적 비용의 관점에서 바라보는 계약과 우리 민법의 태도

        사적 자치의 원칙은 사법(私法)의 기본원리로서 이로부터 계약자유의 원칙, 계약의 구속력 등 계약법 이론을 구성하는 다양한 원칙들이 파생된다. 사적 자치의 원칙은 흔히 각자가 자신의 의지에 따라 생활관계를 전개하고 형성할 수 있어야 한다는 이념으로 표현된다. 그러나 사적 자치의 이면에는 누구도 자신의 의사에 기하지 않고서는 자유가 제약되어서는 안 된다는 또 다른 원리가 자리하고 있다. 지금까지의 계약법 이론은 계약을 평가함에 있어서 계약 내부의 당사자들만을 고려하는 관점을 취해 왔다. 그러나 계약은, 인간의 모든 행위가 그러하듯이 대부분 계약 당사자 외의 제3자에게도 영향을 미친다. 일반적인 경우 그러한 영향은 사회적으로 용인되는 수준이나, 그 비용이 지나치게 커진다면 규제할 필요가 생긴다. 그러나 계약자유의 원칙의 부작용을 최소화하기 위한 제한 법리조차도 당사자 간 불평등에 초점을 맞춘다. 이에 계약 외부에 존재하는 제3자는 계약법의 이론 체계 안으로 포함되기보다는 불법행위법의 영역에 방치되고 있다. 이 글은 제3자를 계약법 이론의 체계로 편입시키고자 하는 미국 학계의 이론들을 먼저 검토한다. 그리고 계약의 사회적 비용을 사법(司法)을 통해 규제하려는 관점이 우리 민법에서 어떤 위치를 차지하고 있는지를 학설과 판례를 통해 알아보고자 한다. 또한 최근 서울중앙지방법원에서 내려진 판결을 통해 사회적 비용 규제의 관점이 법원의 판단에서 어떤 방식으로 견지되어야 하는지 논증한다. 계약은 개인 간에 자유롭게 체결될 수 있어야 하고 일단 체결된 계약은 구속력을 가진다. 법관이 계약 외부의 손익을 고려하여 계약을 바라보는 것은 금기시되어 있다. 그러나 예상치 못한 위험이 언제든 발생할 수 있는 현대 사회에서는 계약을 당사자가 아닌 그 계약을 둘러싼 제3자, 공동체의 관점에서 바라보는 것도 필요하다는 점에서 논의의 의의가 있다. The principle of private autonomy is often expressed in terms of the idea that each person can develop and shape their life relationships according to their will. But behind that principle lies another principle: no one should be able to restrict the other’s freedom without his consent. However, contracts often have an impact on the third parties outside of the contracting parties. In most cases, these impacts are socially acceptable, but when the costs become excessive, it is necessary to regulate them. Even the doctrine of limitation has focused on the inequality between the parties, leaving the third parties outside the contract to the realm of tort law rather than incorporating them into the theoretical framework of contract law. This article first reviews the theories of American academics that seek to incorporate third parties into the theoretical framework of contract law. It then examines the place of the view of judicial regulation of the social costs of contracts in Korean civil law. How the perspective of social cost regulation should be upheld in the court’s judgment through a recent decision of the Seoul Central District Court is also discussed here. Generally, the judges are refrained from looking at contracts by considering gain and loss outside the contract. However, in the modern society, where unforeseen risks can arise at any time, it is necessary to look at contracts not only from the perspective of the parties, but also from the perspective of third parties and the community surrounding the contract.

        CASE REPORT : A Case of Posterior Mediastinal Plasmacytoma Confounded by Community-Acquired Pneumonia

        Plasmacytomas are extramedullary accumulations of plasma cells originating from soft tissue. Mediastinal plasmacytoma is a rare presentation. A 67-year-old man recovered after antibiotic treatment for community-acquired pneumonia. However, on convalescent chest radiography after 3 months, mass like lesion at the right lower lung field was newly detected. Follow-up chest computed tomography (CT) revealed an increase in the extent of the right posterior mediastinal mass that we had considered to be pneumonic consolidations on previous CT scans. Through percutaneous needle biopsy, we diagnosed IgG kappa type extramedullary plasmacytoma of the posterior mediastinum.

        Roles of Protein Arginine Methyltransferases in the Control of Glucose Metabolism

        <P>Glucose homeostasis is tightly controlled by the regulation of glucose production in the liver and glucose uptake into peripheral tissues, such as skeletal muscle and adipose tissue. Under prolonged fasting, hepatic gluconeogenesis is mainly responsible for glucose production in the liver, which is essential for tissues, organs, and cells, such as skeletal muscle, the brain, and red blood cells. Hepatic gluconeogenesis is controlled in part by the concerted actions of transcriptional regulators. Fasting signals are relayed by various intracellular enzymes, such as kinases, phosphatases, acetyltransferases, and deacetylases, which affect the transcriptional activity of transcription factors and transcriptional coactivators for gluconeogenic genes. Protein arginine methyltransferases (PRMTs) were recently added to the list of enzymes that are critical for regulating transcription in hepatic gluconeogenesis. In this review, we briefly discuss general aspects of PRMTs in the control of transcription. More specifically, we summarize the roles of four PRMTs: PRMT1, PRMT 4, PRMT 5, and PRMT 6, in the control of hepatic gluconeogenesis through specific regulation of FoxO1- and CREB-dependent transcriptional events.</P>

        Specialty on social interaction and cooperation between players of MMORPG has developed NPC antagonist's role of scapegoat. Further, NPC antagonists play a role of milestone to expanding user experiencing space meanwhile they act as catalysts which leads players to act. According to systems of MMORPG, there are players who play a role of powerful antagonist, which gives mental complications to protagonist players. It grant justice to player activity, enforce community which enrich player's life as a social member. Therefore, unlike antagonists in existed linear narratives which are targets to be eliminated, antagonists in MMORPG with unlinear and expanding narrative, plays important role in generating possibilities to create narratives.

      • Protein arginine methyltransferase 1 regulates hepatic glucose production in a FoxO1‐dependent manner

        <P><B>Abstract</B></P><P>Postprandial insulin plays a critical role in suppressing hepatic glucose production to maintain euglycemia in mammals. Insulin‐dependent activation of protein kinase B (Akt) regulates this process, in part, by inhibiting FoxO1‐dependent hepatic gluconeogenesis by direct phosphorylation and subsequent cytoplasmic exclusion. Previously, it was demonstrated that protein arginine methyltransferase 1 (PRMT1)‐dependent arginine modification of FoxO1 interferes with Akt‐dependent phosphorylation, both in cancer cells and in the <I>Caenorhabditis elegans</I> model, suggesting that this additional modification of FoxO1 might be critical in its transcriptional activity. In this study, we attempted to directly test the effect of arginine methylation of FoxO1 on hepatic glucose metabolism. The ectopic expression of PRMT1 enhanced messenger RNA levels of FoxO1 target genes in gluconeogenesis, resulting in increased glucose production from primary hepatocytes. Phosphorylation of FoxO1 at serine 253 was reduced with PRMT1 expression, without affecting the serine 473 phosphorylation of Akt. Conversely, knockdown of PRMT1 promoted an inhibition of FoxO1 activity and hepatic gluconeogenesis by enhancing the phosphorylation of FoxO1. In addition, genetic haploinsufficiency of <I>Prmt1</I> reduced hepatic gluconeogenesis and blood‐glucose levels in mouse models, underscoring the importance of this factor in hepatic glucose metabolism <I>in vivo</I>. Finally, we were able to observe an amelioration of the hyperglycemic phenotype of <I>db/db</I> mice with PRMT1 knockdown, showing a potential importance of this protein as a therapeutic target for the treatment of diabetes. <I>Conclusion:</I> Our data strongly suggest that the PRMT1‐dependent regulation of FoxO1 is critical in hepatic glucose metabolism <I>in vivo</I>.         아민 옥사이드 양쪽성 계면활성제 첨가가 리포좀 특성에 미치는 영향에 관한 연구

        본 연구에서는 pH 변화에 따른 양쪽성 계면활성제 첨가가 리포좀 특성에 미치는 영향에 관하여 살펴보았다. 이를 위하여 아민 옥사이드(amine oxide) 양쪽성 계면활성제를 합성하고 1H NMR, 13C NMR 및 FT-IR 분석을 통하여 구조를 규명하였으며, 계면활성제의 임계 마이셀 농도(critical micelle concentration, CMC) 및 표면장력 등의 계면 물성(interfacial property)을 측정하였다. 또한 제타 전위(zeta potential) 측정을 통하여 양쪽성 계면활성제가 양이온 계면활성제(cationic surfactant)에서 음이온 계면활성제(anionic surfactant)로 작용이 전환되는 등전점(isoelectric point)을 결정하였다. 이 결과를 바탕으로 등전점을 중심으로 한 pH 변화 및 아민 옥사이드 계면활성제의 탄화수소 사슬 길이가 리포좀의 평균 입자 크기 변화 및 제타 전위 등과 같은 안정성에 미치는 영향에 살펴보았다. 또한 pH 및 계면활성제의 탄화수소 사슬 길이 변화에 따른 및 리포좀과 계면활성제의 결합 정도(binding degree) 및 리포좀 막의 가변형성(deformability) 등의 측정을 통하여 리포좀 막의 유동성(fluidity) 변화에 미치는 영향을 이해하고자 하였다. In this study, zwitterionic surfactants were added to liposome systems at different pH conditions to understand the effect of surfactants on liposome characteristics. For this purpose, amine oxide surfactants having different hydrocarbon chain lengths were synthesized and the structure of the resulting product was elucidated by using 1H NMR, 13C NMR, and FT-IR. In addition, the physical properties of newly synthesized surfactants such as critical micelle concentration (CMC), surface tension and isoelectric point were measured. The stability characteristics of liposome systems including average particle sizes and zeta potentials were measured by varying pH and hydrocarbon chain lengths of an amine oxide surfactant. Effects of the pH and hydrocarbon chain length of an amine oxide surfactant on fluidity of a liposome membrane were also examined by measuring the deformability and the binding degree between the surfactant and liposome.
