        Supercritical fluids (SCF) are useful substances that are recently used in bioseparation process. Extraction of carthamin from safflower using supercritical carbon dioxide were examined at various conditions of temperature (35∼40℃), pressure (3000∼5000 psi) and CO₂ flow rate (900∼1200 m/hr). SCF was less effective than solvent methods in carthamin extraction from safflower. Concentration of carthamin was 50% more efficent at the use of alginate beads with 35 g cellulose per column loadings compared to the control (15g cellulose per column loadings).

        KLF5-trancripted miR-125b-5p is involved in enhancing the radio-sensitivity of breast cancer cells by targeting BRCA1

        Background A large number of clinical trials have proved that radiotherapy is an effective strategy for treating breast cancer (BC), but radioresistance is an urgent problem to be solved. Objective Herein, radio-tolerant BC cell lines named MDA-MB-231/R and MCF-7/R were successfully constructed, respectively, and it was shown that microRNA-125b-5p (miR-125b-5p) was lowly expressed in radio-tolerant cells, which was associated with poor prognosis of BC patients. Results Furthermore, miR-125b-5p over expression could hinder cell proliferation and strengthen radio-sensitivity. Mechanistically, Breast Cancer 1 (BRCA1) was identified as a downstream protein target of miR-125b-5p in BC cells, and Kruppellike factor 5 (KLF5) was identified as an upstream transcription factor involved in promoting miR-125b-5p expression. More importantly, over expression of KLF5 suppressed BC cell proliferation and increased its radiation sensitivity, which could be retained by miR-125b-5p downregulation. Conclusion In conclusion, these findings suggested that the KLF5/miR-125b-5p/BRCA1 axis may provide new information on radiation therapy for breast cancer.

        Integrated omics approaches to characterize a nuclear receptor corepressor-associated histone deacetylase in mouse skeletal muscle

        <P><B>Abstract</B></P> <P>Nuclear receptors regulate gene expression by differentially binding to coactivators or corepressors in a ligand-dependent manner, which further recruits a set of epigenome-modifying enzymes that remodel chromatin conformation. Histone acetylation is a major epigenomic change controlled by histone acetyltransferases (HATs) and histone deacetylases (HDACs). HDAC3 is the only HDAC that confers the enzymatic activity to the complexes nucleated by nuclear receptor corepressors NCoR and SMRT. To address the metabolic function of HDAC3, we have deleted it specifically in mouse skeletal muscles. We have performed the following omics profiling in skeletal muscles of these mice: (1) RNA-seq profiling of total RNA; (2) Global nuclear run-on (GRO-seq) analysis of nascent RNAs; (3) Chromatin immuno-precipitation (ChIP-seq) of HDAC3 at both early evening and early morning; (4) proteomics profiling with mass spectrometry; (5) snap-shot metabolomics profiling of water-soluble metabolites at the basal condition; (6) snap-shot metabolomics profiling of lipid species at the basal condition; (7) kinetic fluxomics analysis of glucose utilization using <SUP>13</SUP>C<SUB>6</SUB>-glucose <I>In vivo</I> during treadmill running exercise. These approaches have provided several novel insights into how nuclear receptors regulate circadian rhythm of skeletal muscle fuel metabolism, which has been published elsewhere. Here we present the original datasets and technical details during the execution, analysis, and interpretation of these omics studies.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A mouse model with skeletal muscle-specific KO of HDAC3. </LI> <LI> <I>in vivo</I> RNA-seq, GRO-seq, and ChIP-seq identified relevance to circadian clock. </LI> <LI> Total proteome profiling in muscle samples. </LI> <LI> Metabolomics profiling identified disruption of BCAAs metabolism. </LI> <LI> Fluxomics with <SUP>13</SUP>C-glucose <I>in vivo</I> during treadmill running. </LI> </UL> </P>

        Research of 3D image processing of VR technology in medicine based on DNN

        According to a survey published in an authoritative journal in January 2020, the globalincidence rate of mental illness is 8.3% for men and 10.6% for women, which indicates thatmental illness has become a globally recognized obstacle. Therefore, specific psychotherapy including mental illness will become an important research topic. It is very effective forpatients with special mental diseases, such as mental illness, to reduce their mental reaction byexposure therapy; the system uses the virtual reality system of medical images processed by learningalgorithm to reproduce the effect of virtual reality exposure method of the high scene of transparent ladder. Compared with the old invasive exposure scene, the results show that theimprovement of both conditions has obvious effect, and the effect of human treatment underthe two conditions is not good. There are obvious differences, which show that virtual reality model will gradually replace the on-the-spot feeling. Finally, with more and more researchers have put forward a variety of other virtual reality image processing models, the research of image processing has gradually become more and more interested.

      • Gene Polymorphisms of OPRM1 A118G and ABCB1 C3435T May Influence Opioid Requirements in Chinese Patients with Cancer Pain

        Backgrounds: Polymorphisms of OPRM1 A118G and ABCB1 C3435T have been suggested to contribute to inter-individual variability regarding pain sensitivity, opioid usage, tolerance and dependence and incidence of adverse effects in patients with chronic pain. This study aimed to investigate the association of both two polymorphisms with opioid requirements in Chinese patients with cancer pain. Methods: The genotypes of rs1799971 (OPRM1) and rs1045642 (ABCB1) were determined by PCR-RFLP and direct sequencing methods respectively in 112 patients with cancer-related pain. Comparisons between the different genotype or allele groups were performed with t-tests or one-way ANOVA tests, as appropriate. The potential relationship of allele number with opioid response was performed with a trend Jonckheere-Terpstra test. Results: In the 112 subjects, the frequencies of variant 118 G and 3435T allele were 38.4% and 37.9%, respectively. Significant higher 24h-opioid doses were observed in patients with GG (P=0.0004) and AG + GG (P=0.005) genotypes than the AA carriers. The dominant mutant 118G allele tended to be associated with progressively increasing 24h-opioiddoses (P=0.001). Compared with CC/CT, patients with ABCB1 TT genotype received higher 24h- and weight-surface area-adjusted-24h- opioids doses (P=0.057 and 0.028, respectively). Conclusions: The OPRM1 A118G single nucleotide polymorphism (SNP) is a key contributor for the inter-individual variability in opioidrequirements in Chinese cancer pain patients. This may possibly extend to the ABCB1 C3435T SNP.

        Estimation of the load-deformation responses of flanged reinforced concrete shear walls

        As limited well-documented experimental data are available for assessing the attributes of different deformation components of flanged walls, few appropriate models have been established for predicting the inelastic responses of flanged walls, especially those of asymmetrical flanged walls. This study presents the experimental results for three large-scale T-shaped reinforced concrete walls and examines the variations in the flexural, shear, and sliding components of deformation with the total deformation over the entire loading process. Based on the observed deformation behavior, a simple model based on moment-curvature analysis is established to estimate flexural deformations, in which the changes in plastic hinge length are considered and the deformations due to strain penetration are modeled individually. Based on the similar gross shapes of the curvature and shear strain distributions over the wall height, a proportional relationship is established between shear displacement and flexural rotation. By integrating the deformations due to flexure, shear, and strain penetration, a new load-deformation analytical model is proposed for flexure-dominant flanged walls. The proposed model provides engineers with a simple, accurate modeling tool appropriate for routine design work that can be applied to flexural walls with arbitrary sections and is capable of determining displacements at any position over the wall height. By further simplifying the analytical model, a simple procedure for estimating the ultimate displacement capacity of flanged walls is proposed, which will be valuable for performance-based seismic designs and seismic capacity evaluations.

        Gastroprotective effect of cirsilineol against hydrochloric acid/ethanol-induced gastric ulcer in rats

        This study was designed to evaluate the gastroprotective activity of cirsilineol in hydrochloric acid (HCl)/ethanol-induced gastric ulcer model. Cirsilineol was administered at the doses of 20 and 40 mg/kg in HCl/ethanol-induced rats. The gastroprotective ability was verified by determining the ulcer score, total acidity, hemoglobin, inflammatory cytokines, lipid peroxides, and enzymatic antioxidants superoxide dismutase (SOD) and catalase (CAT) in gastric tissue and serum biochemical analysis. The results showed a favorable increase in the hemoglobin level, antioxidant enzymes (SOD and CAT), restored electrochemical balance (carbon dioxide & anion gap) while a noticeable decrease in ulcer index, total acidity, lipid peroxides, inflammatory cytokines (interleukin-1 beta [IL-1β], IL-6, and tumor necrosis factor alpha) in rats treated with the cirsilineol. The serum biochemical analysis on liver markers (alkaline phosphatases, alanine aminotransferase, and aspartate aminotransferase), kidney markers (urea, creatinine, albumin, globulin, total protein), and lipid profile (triglyceride, high-density lipoprotein, total cholesterol) were attenuated by cirsilineol treatment in rats. Histopathology showed enhanced gastric protection and preserved the integrity of gastric mucosa upon cirsilineol administration. These results ultimately suggest that cirsilineol has gastroprotective effects that prevent the development of gastric ulcer.

        Radio Frequency Ablation in the Rabbit Lung Using Wet Electrodes: Comparison of Monopolar and Dual Bipolar Electrode Mode

        <P><B>Objective</B></P><P>To compare the effect of radio frequency ablation (RFA) on the dimensions of radio frequency coagulation necrosis in a rabbit lung using a wet electrode in monopolar mode with that in dual electrode bipolar mode at different infusion rates (15 mm/hr versus 30 ml/hr) and saline concentrations (0.9% normal versus 5.8% hypertonic saline).</P><P><B>Materials and Methods</B></P><P>Fifty ablation zones (one ablation zone in each rabbit) were produced in 50 rabbits using one or two 16-guage wet electrodes with a 1-cm active tip. The RFA system used in the monopolar and dual electrode wet bipolar RFA consisted of a 375-kHz generator (Elektrotom HiTT 106, Berchtold, Medizinelektronik, Germany). The power used was 30 watts and the exposure time was 5 minutes. The rabbits were assigned to one of five groups. Group A (n = 10) was infused with 0.9% NaCl used at a rate of 30 ml/hr in a monopolar mode. Groups B (n = 10) and C (n = 10) were infused with 0.9% NaCl at a rate of 15 and 30 ml/hr, respectively in dual electrode bipolar mode; groups D (n = 10) and E (n = 10) were infused with 5.8% NaCl at a rate of 15 and 30 ml/hr, respectively in a dual electrode bipolar mode. The dimensions of the ablation zones in the gross specimens from the groups were compared using one-way analysis of variance by means of the Scheffe test (post-hoc testing).</P><P><B>Results</B></P><P>The mean largest diameter of the ablation zones was larger in dual electrode bipolar mode (30.9±4.4 mm) than in monopolar mode (22.5±3.5 mm). The mean smallest diameter of the ablation zones was larger in dual electrode bipolar mode (22.3±2.5 mm) than in monopolar mode (19.5±3.5 mm). There were significant differences in the largest and smallest dimension between the monopolar (group A) and dual electrode wet bipolar mode (groups B-E). In dual electrode bipolar mode, the mean largest diameter of the ablation zones was larger at an infusion rate of 15 ml/hr (34.2±4.0 mm) than at 30 ml/hr (27.6±0.1 mm), and the mean smallest diameter of the ablation zones was larger at an infusion rate of 15 ml/hr (27.2±7.5 mm) than at an infusion rate of 30 ml/hr (24±2.9 mm).</P><P><B>Conclusion</B></P><P>Using a wet electrode, dual electrode bipolar RFA can create a larger ablation zone more efficiently than monopolar RFA.</P>
