        Evaluation of mango saponin in broilers: effects on growth performance, carcass characteristics, meat quality and plasma biochemical indices

        Objective: The objective of the present study was to determine whether mango saponin (MS) could be used as a feed additive in broiler chicks by evaluating growth performance, carcass characteristics, meat quality, and plasma biochemical indices. Methods: A total of 216 1-d-old Arbor Acres male broiler chicks were randomly assigned into three dietary treatments supplemented with 0 (control), 0.14% (MS 0.14%), or 0.28% (MS 0.28%) MS. Each treatment had six replicates (cages) with 12 chicks each. The feeding trial lasted for six weeks. Results: Compared with the control, dietary supplemented with 0.14% or 0.28% MS increased average daily weight gain of chicks in the grower (22 to 42 d) and the whole (1 to 42 d) phases, and the final body weight of chicks on d 42 was higher in MS supplemented groups (p<0.05). Lower L45 min* (lightness) and L24 h* values, lower b24 h* (yellowness) value, and higher a45 min* (redness) and a24 h* values of the breast muscle were observed in chicks fed with 0.28% MS on d 42 (p<0.05). The total antioxidant capacity in plasma increased in MS 0.14% group on d 21 (p<0.001). Lower contents of plasma total cholesterol and triglyceride were observed in chicks fed with 0.28% MS on d 21 and d 42, whereas the group supplemented with 0.14% MS only decreased plasma triglyceride content on d 21 (p<0.05). The glucose content in plasma decreased in MS 0.28% group on d 42 (p<0.001). Conclusion: Overall, MS could be used as a feed additive in broiler chicks, and the supplemental level of 0.28% MS in diet could improve growth performance, meat quality, and plasma lipid metabolism in broiler chicks.

        True Digestibility of Phosphorus in Different Resources of Feed Ingredients in Growing Pigs

        To determine the true digestible phosphorus (TDP) requirement of growing pigs, two experiments were designed with the experimental diets containing five true digestible P levels (0.16%, 0.20%, 0.23%, 0.26% and 0.39%) and the ratio of total calcium to true digestible P (TDP) kept at 2:1. In Experiment 1, five barrows (Duroc${\times}$Landrace${\times}$Yorkshire) with an average initial body weight of 27.9 kg were used in a $5{\times}5$ Latin-square design to evaluate the effect of different dietary P levels on the digestibility and output of P and nitrogen. In Experiment 2, sixty healthy growing pigs (Duroc${\times}$Landrace${\times}$Yorkshire) with an average body weight (BW) of 21.4 kg were assigned randomly to one of the five dietary treatments (12 pigs/diet), and were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. The results indicated that the true digestibility of P increased (p<0.05) linearly with increasing dietary TDP level below 0.26%. The true P digestibility was highest (56.6%) when dietary TDP was 0.34%. Expressed as g/kg dry matter intake (DMI), fecal P output increased (p<0.05) linearly with increasing P input. On the basis of g/kg fecal dry matter (DM), fecal P output was lowest for Diet 4 and highest (p<0.05) for Diet 5. The apparent digestibility of crude protein (CP) did not differ (p>0.05) among the five diets, with the average nitrogen output of 12.14 g/d and nitrogen retention of 66% to 74% (p>0.05), which suggested that there was no interaction between dietary P and CP protein levels. During the 28-d experimental period of Experiment 2, the average daily gain (ADG) of pigs was affected by dietary TDP levels as described by Eq. (1): $y=-809,532x^4+788,079x^3-276,250x^2+42,114x-1,759$; ($R^2=0.99$; p<0.01; y = ADG, g/d; x = dietary TDP, %), F/G for pigs by Eq. (2): $y=3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2=0.99$; p<0.01; y = F/G; x = dietary TDP, %), and Total P concentrations in serum by Eq. (3): $y=-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ (R2 = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest F/G (1.07) and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet at a total Ca to TDP ratio of 2:1.

        Improvement of lower hybrid current drive systems for high-power and long-pulse operation on EAST

        Aiming at high-power and long-pulse operation up to 1000 s, some improvements have been made for both 2.45 GHz and 4.6 GHz lower hybrid (LH) systems during the recent 5 years. At first, the guard limiters of the LH antennas with graphite tiles were upgraded to tungsten, the most promising material for plasma facing components in nuclear fusion devices. These new guard limiters can operate at a peak power density of 12.9 MW/m2 . Strong hot spots were usually observed on the old graphite limiters when 4.6 GHz system operated with power >2.0 MW [B. N. Wan et al., Nucl. Fusion 57 (2017) 102019], leading to a reduction of the maximum power capability. With the new limiters, 4.6 GHz LH system, the main current drive (CD) and electron heating tool for EAST, can be operated with power >2.5 MW routinely. Long-pulse operation up to 100 s with 4.6 GHz LH power of 2.4 MW was achieved in 2021 and the maximal temperature on the guard limiters measured by an infrared (IR) camera was about 540 C, much below the permissible value of tungsten material (~1200 C). A discharge with a duration of 1056 s was achieved and the 4.6 GHz LH energy injected into the plasma was up to 1.05 GJ. Secondly, the fully-activemultijunction (FAM) launcher of 2.45 GHz system was upgraded to a passive-active-multijunction (PAM), for which the density of optimum coupling was relatively low (below the cut-off value). Good coupling with reflection coefficient ~3% has been achieved with plasma-antenna distance up to 11 cm for the new PAM. Finally, in order to eliminate the effect of ion cyclotron range of frequencies (ICRF) wave on 4.6 GHz LH wave coupling, the location of the ICRF launcher was changed to a port that is located 157.5 toroidally from the 4.6 GHz LH system and is not magnetically connected

      • PANCHROMATIC OBSERVATIONS OF THE TEXTBOOK GRB 110205A: CONSTRAINING PHYSICAL MECHANISMS OF PROMPT EMISSION AND AFTERGLOW

        <P>We present a comprehensive analysis of a bright, long-duration (T-90 similar to 257 s) GRB 110205A at redshift z = 2.22. The optical prompt emission was detected by Swift/UVOT, ROTSE-IIIb, and BOOTES telescopes when the gamma-ray burst (GRB) was still radiating in the gamma-ray band, with optical light curve showing correlation with gamma-ray data. Nearly 200 s of observations were obtained simultaneously from optical, X-ray, to gamma-ray (1 eV to 5 MeV), which makes it one of the exceptional cases to study the broadband spectral energy distribution during the prompt emission phase. In particular, we clearly identify, for the first time, an interesting two-break energy spectrum, roughly consistent with the standard synchrotron emission model in the fast cooling regime. Shortly after prompt emission (similar to 1100 s), a bright (R = 14.0) optical emission hump with very steep rise (alpha similar to 5.5) was observed, which we interpret as the reverse shock (RS) emission. It is the first time that the rising phase of an RS component has been closely observed. The full optical and X-ray afterglow light curves can be interpreted within the standard reverse shock (RS) + forward shock (FS) model. In general, the high-quality prompt and afterglow data allow us to apply the standard fireball model to extract valuable information, including the radiation mechanism (synchrotron), radius of prompt emission (R-GRB similar to 3 x 10(13) cm), initial Lorentz factor of the outflow (Gamma(0) similar to 250), the composition of the ejecta (mildly magnetized), the collimation angle, and the total energy budget.</P>

        <P>The Taiwanese-American Occultation Survey (TAOS) project has collected more than a billion photometric measurements since 2005 January. These sky survey data—covering timescales from a fraction of a second to a few hundred days—are a useful source to study stellar variability. A total of 167 star fields, mostly along the ecliptic plane, have been selected for photometric monitoring with the TAOS telescopes. This paper presents our initial analysis of a search for periodic variable stars from the time-series TAOS data on one particular TAOS field, No. 151 (R.A. = 17<SUP>h</SUP>30<SUP>m</SUP>6<img entity='fs' SRC='http://ej.iop.org/icons/Entities/fs.gif' ALT='fs' ALIGN='BASELINE' />7, decl. = 27°17'30'', J2000), which had been observed over 47 epochs in 2005. A total of 81 candidate variables are identified in the 3 deg<SUP>2</SUP> field, with magnitudes in the range 8 < R < 16. On the basis of the periodicity and shape of the light curves, 29 variables, 15 of which were previously unknown, are classified as RR Lyrae, Cepheid, δ Scuti, SX Phonencis, semi-regular, and eclipsing binaries.</P>

        <P>Rh0.5Pd0.5/CeO2 bimetallic nanoparticles were subjected to reducing and oxidizing atmospheres at different temperatures and characterized by means of the Ambient Pressure X-Ray Photoelectron Spectroscopy (AP-XPS). The surface atomic population is calculated for all cases. By using a high temperature pretreatment at 480 degrees C in H-2 atmosphere, it was possible to control the surface atomic population with Pd atoms frozen at the skin layer even in an oxidizing atmosphere, which is not observed without high temperature pre-treatment (Rh atoms at the skin layer). The change of the Rh/Pd surface segregation behavior after the high temperature pre-treatment is attributed to the geometrical factor of Strong Metal Support Interaction (SMSI) effect, i.e., the occurrence of the ceria capping layer around nanoparticles.       • Particulate and gas sampling of prescribed fires in South Georgia, USA

        Gaseous and particulate species from two prescribed fires were sampled in-situ, to better characterize prescribed burn emissions. Measurements included gaseous and fine particulate matter (PM<SUB>2.5</SUB>) species, particle number concentration, particulate organic carbon (POC) speciation, water-soluble organic carbon (WSOC) and water-soluble iron. Major PM<SUB>2.5</SUB> components included OC (~57%), EC (~10%), chloride (~1.6%), potassium (~0.7%) and nitrate (~0.9%). Major gaseous species include carbon dioxide, carbon monoxide, methane, ethane, methanol and ethylene. Particulate organic tracers of biomass burning, such as levoglucosan, dehydroabietic acid and retene, increased significantly during the burns. Water-soluble organic carbon (WSOC) also increased significantly during the fire and levels are highly correlated with total potassium (K) (R<SUP>2</SUP> = 0.93) and levoglucosan (R<SUP>2</SUP> = 0.98). The average WSOC/OC ratio was 0.51 +/- 0.03 and did not change significantly from background levels. Thus, the WSOC/OC ratio may not be a good indicator of secondary organic aerosol (SOA) in regions that are expected to be impacted by biomass burning. Results using a biomass burning source profile derived from this work further indicate that source apportionment is sensitive to levels of potassium in biomass burning source profiles. This underscores the importance of quantifying local biomass burning source profiles.

        A multiwavelength analysis of a collection of short-duration GRBs observed between 2012 and 2015

        <B>Abstract</B><P>We investigate the prompt emission and the afterglow properties of short-duration gamma-ray burst (sGRB) 130603B and another eight sGRB events during 2012-2015, observed by several multiwavelength facilities including the Gran Canarias Telescope 10.4 m telescope. Prompt emission high energy data of the events were obtained by INTEGRAL-SPI-ACS, Swift-BAT, and Fermi-GBM satellites. The prompt emission data by INTEGRAL in the energy range of 0.1-10 MeV for sGRB 130603B, sGRB 140606A, sGRB 140930B, sGRB 141212A, and sGRB 151228A do not show any signature of the extended emission or precursor activity and their spectral and temporal properties are similar to those seen in case of other short bursts. For sGRB 130603B, our new afterglow photometric data constrain the pre-jet-break temporal decay due to denser temporal coverage. For sGRB 130603B, the afterglow light curve, containing both our new and previously published photometric data is broadly consistent with the ISM afterglow model. Modeling of the host galaxies of sGRB 130603B and sGRB 141212A using the LePHARE software supports a scenario in which the environment of the burst is undergoing moderate star formation activity. From the inclusion of our late-time data for eight other sGRBs we are able to: place tight constraints on the non-detection of the afterglow, host galaxy, or any underlying ‘kilonova’ emission. Our late-time afterglow observations of the sGRB 170817A/GW170817 are also discussed and compared with the sub-set of sGRBs.</P>

      • Role of Dorsal Striatum Histone Deacetylase 5 in Incubation of Methamphetamine Craving

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Methamphetamine (meth) seeking progressively increases after withdrawal (incubation of meth craving). We previously demonstrated an association between histone deacetylase 5 (HDAC5) gene expression in the rat dorsal striatum and incubation of meth craving. Here we used viral constructs to study the causal role of dorsal striatum HDAC5 in this incubation.</P> <P><B>Methods</B></P> <P>In experiment 1 (overexpression), we injected an adeno-associated virus bilaterally into dorsal striatum to express either green fluorescent protein (control) or a mutant form of HDAC5, which strongly localized to the nucleus. After training rats to self-administer meth (10 days, 9 hours/day), we tested the rats for relapse to meth seeking on withdrawal days 2 and 30. In experiment 2 (knockdown), we injected an adeno-associated virus bilaterally into the dorsal striatum to express a short hairpin RNA either against luciferase (control) or against HDAC5. After training rats to self-administer meth, we tested the rats for relapse on withdrawal days 2 and 30. We also measured gene expression of other HDACs and potential HDAC5 downstream targets.</P> <P><B>Results</B></P> <P>We found that HDAC5 overexpression in dorsal striatum increased meth seeking on withdrawal day 30 but not day 2. In contrast, HDAC5 knockdown in the dorsal striatum decreased meth seeking on withdrawal day 30 but not on day 2; this manipulation also altered other HDACs (<I>Hdac1</I> and <I>Hdac4</I>) and potential HDAC5 targets (<I>Gnb4</I> and <I>Suv39h1</I>).</P> <P><B>Conclusions</B></P> <P>Results demonstrate a novel role of dorsal striatum HDAC5 in incubation of meth craving. These findings also set up future work to identify HDAC5 targets that mediate this incubation.</P>

        Association between Polymorphisms of MSTN and MYF5 Genes and Growth Traits in Three Chinese Cattle Breeds

        The objective of this study was to assess the association of polymorphisms in MSTN and MYF5 genes with growth traits in three Chinese cattle breeds. Only one homozygous animal with BB genotype at MSTN locus was observed in Jiaxian population which was at Hardy-Weinberg disequilibrium (p<0.05). The frequencies of allele A at MSTN locus and allele B at MYF5 locus in the three Chinese breeds were 0.9550/0.9730/0.9720 and 0.8275/0.7581/0.7523, respectively. Allele A at MSTN locus and allele B at MYF5 locus were dominant in these three populations. No statistically significant differences in growth traits were observed between the genotypes of the Jiaxian breed at MSTN and MYF5 loci and the Nanyang breed at MYF5 locus. However, there were statistically significant differences between the genotypes at MSTN locus of the Nanyang breed for WH, HG, HGI and HGBLR (p<0.05), and of the Qinchuan breed for BLI (p<0.05). The SNP in MYF5 had significant effects on WH and HHC of Qinchuan animals (p<0.05). These results suggest that MSTN and MYF5 are strong candidate genes that influence growth traits in cattle. Other SNPs of MSTN and MYF5 or other linked genes should also be studied, which could lead to the development of selection plans to improve the performance of Chinese cattle and also promote the breeding of genuine beef cattle in China.
