한국산 인삼의 Polyphenol 분획물의 항산화, Phospholipase $A_2$ 및 암세포증식 억제효과

한국산 인삼을 60% acetone으로 추출하여 Sepadex LH-20 gel column chromatography, MCI-CHP 20 gel column chromatography, ${\mu}-Bondapack\;C_{18}$ gel column chromatography을 이용하여 polyphenol 분획물을 3종류를 분리하여 항산화 효과, phospholipase $A_2$ 저해효과 및 암세포 증식억제 효과를 검토하였다. 항산화 효과에서 분획물 I은 150 ppm에서 $Cu^{2+},\;KO_2$ 그리고 $H_2O_2$에 대하여 각각 48.16%, 79.71%, 43.55%를 나타내었고 DPPH 수소공여능은 분획물 II가 200 ppm에서 35.17%의 라디칼소거능을 보였다. 분획물 III는 $Fe^{2+}$, OH가 존재 시 150 ppm에서 48.49%, 25%의 항산화효과를 나타내었다. Phospholipase $A_2$ 저해활성은 분획물 III이 $60\;{\mu}/ml$에서 48.9%의 활성을 나타냈다. HT 29 cell에 대한 인삼의 세포 독성은 분획물 II에서 0.25 mg/ml에서 73.29%의 가장 높은 억제능을 나타내었다. The polyphenol fractions of Korean ginseng were purified using Sephadex LH-20, MCI gel, Bondapak $C_{18}$ TLC, and HPLC from the 60% acetone soluble fraction. Fraction I showed 48.16%, 79.71% and 43.55% inhibition at 150 ppm against lipid oxidation in the presence of copper ion, superoxide and hydrogen peroxidation. Electron donating abilities of fraction II showed 35.17% inhibition at 200 ppm. Fraction III showed 48.49% and 25% inhibition at 150 ppm against lipid oxidation in the presence of ferrous ion and hydroxy radical ion. The phospholipase $A_2$ inhibitory effect of fraction III was 48.9% at the concentration of $60\;{\mu}g/ml$. The cytotoxic effects of fraction II was the highest (73.29% at 0.25 mg/ml) among the tested polyphenol fractions.

새송이버섯의 갓과 대를 열수, 50% 에탄올 및 100% 에탄올 등의 추출용매를 사용하여 건물 중량의 50배에 해당되는 부피(w/v)일 때, 추출물들의 생리활성을 탐색하였다. 전자 공여작용의 경우 갓의 50% 에탄올 추출물에서 88%의 높은 전자공여능을 나타내었으며, SOD 유사활성을 측정한 결과 갓의 열수 추출물이 62.57%로 다른 추출물에 비해 높은 활성을 보여주었다. Tyrosinase 저해활성의 경우 전자공여능 및 SOD 유사활성에서 대보다 갓의 활성이 높았던 것과는 반대로 대의 활성이 높았으며, 그중 100% 에탄올 추출물에서 58.57%로 비교물질로 사용된 0.1% L-ascorbic acid보다 31.29% 더 높은 활성을 가지는 것으로 조사되었다. Angiotensin converting enzyme 저해활성에서도 SOD 유사활성과 비슷하게 갓의 열수 추출물이 95.14%로 매우 높은 활성을 보였으며, 총 폴리페놀 함량의 경우 갓의 열수 및 50% 에탄올 추출물에서 각각 1427.25 ㎎%, 1426.82 ㎎%로 높은 함량을 나타내었다. 아질산염 소거작용을 측정한 결과 pH 1.2일 때 갓과 대의 소거능이 높게 나타났다. 이와같은 결과는 새송이버섯의 생리활성을 밝혀 기능성 소재로써 이 용도가 크게 증가할 것으로 판단된다. Physiological activities of pileus and stipe extracts from fresh Pleurotus eryngii were examined. Electron donating ability (EDA), superoxide dismutase (SOD)-like activity, tyrosinase inhibitory effect, angiotensin converting enzyme (ACE) inhibitory activity, total polyphenol contents and nitrite scavenging ability were examined for extracts of pileus and stipe of Pleurotus eryngii extracted with water, 50% and 100% ethanol. Volume of the solvent used was 50 time of sample on a weight/volume basis. EDA was 88% in 50% ethanol extract (pileus). Also, SOD-like activity was 62.57% in water extract (pileus). Tyrosinase inhibitory effects of all samples were higher than 0.1% L-ascorbic acid solution. ACE inhibitory activity of water extract of pileus was found to be the highest value of 95.14%. In addition, total polyphenol contents were the highest in water and 50% ethanol extracts of pileus (1427.25 ㎎%, 1426.82 ㎎%). Finally, nitrite scavenging ability of 50% ethanol extract of pileus at pH 1.2 showed approximately 95%. The results will be useful for understanding the physiological activities of Pleurotus eryngii extracts.
한국산 배로부터 분리한 Polyphenol 분획물이 지질대사에 미치는 영향

배로부터 polyphenol 화합물군을 분리 및 정제하여 5주간 고지방식이를 한 후, 혈장 및 간장에서의 지질대사에 미치는 영향을 분석한 결과 혈장에서는 배의 polyphenol 화합물군 PR-Ⅱ에서 총지질, 총콜레스테롤이 유의성 있게 감소하였다. 또한 간장에서는 총지질, 중성지질이 대조군에 비해서 유의성 있는 결과가 나타났다. 5주 동안 고지방 식이를 한 후 분변을 채취하여 총지질, 총콜레스테롤 및 중성지질을 측정한 결과 배에서 추출한 polyphenol 화합물 군이 고지방식 이만을 섭취시킨 대조군에 비해 다소 감소되는 경향을 보였다. 배의 polyphenol 화합물군을 섭취시킨 후 총단백질과 알부민 함량을 측정한 결과 대조군과 실험군에서 유의성 있는 차이는 나타나지 않았다. This study was conducted to investigate the effects of the polyphenol fraction isolated from pear on the reduction of fat accumulation in rats fed on high fat diet for 5 weeks, which was examined by analyzing the lipid composition in serum, liver and feces biochemically. It was shown that the levels of total lipid and total cholesterol in serum were remarkably reduced in polyphenol fraction supplemented group as compared with the control group. The levels of total lipid and triglyceride were also significantly lower in polyphenol group than control group. The contents of total lipid, total cholesterol and triglyceride in feces were tended to be slightly increased in polyphenol group compared to control group. The total protein and albumin of polyphenol groups were lower compared to those of control group, which were not significant.
양파로부터 분리한 생리활성분획이 생리활성 및 지질대사에 미치는 영향

양파를 80% ethanol로 추출하여 Sephadex LH-20 column chromatography로 fraction I, II 및 III의 생리활성물질을 분리하여 angiotensin converting enzyme(ACE), xanthine oxidase, tyrosinase의 저해작용 효과와 지질대사를 검토 하였다. Fraction I과 II의 ACE 저해작용은 50 ppm 농도에서 80%이상의 효소 저해효과를 나타내었고 fraction III에서는 500ppm 농도에서 90% 효소저해작용을 보였다. 100 ppm 농도에서 fraction III의 xantine oxidase 효소저해 효과는 82.5%로 나타났으며 tyrosinase의 저해효과는 42.6%이었다. 효소저해작용이 가장 강한 fraction III를 2개월 동안 고지방식이에 첨가하여 혈장 및 간장에서 지질대사에 미치는 영향을 분석한 결과 혈장에서는 총지질, 중성지질이 대조군에 비하여 유의성 있게 감소하였다. 간의 콜레스테롤량도 약 50% 정도 감소하는 결과를 얻었다. From the 80% ethanol extraction of onion flesh, fraction I, Ⅱ and Ⅲ were separated by Sephadex LH-20 column chromatography. Fraction Ⅱ and Ⅲ had a high inhibitory effect on ACE and xanthine oxidase. The ACE was about 80% at 50 ppm of fraction Ⅱ and Ⅲ. The xanthine oxidase was about 82.5% at 100 ppm of fraction Ⅲ. This research was studied to investigate the effects of the fraction Ⅲ isolated from onion on the reduction of fat accumulation in rats fed high fat diet for 2 months. The metabolism was examined by analyzing the fat in serum and liver. It was shown that cholesterol in serum was remarkably reduced in onion fraction Ⅲ as compared with those of control.

The biochemical components of Namcheondlggae, Miryangdlkkae 25, Boradlggae and Ipdlkkae 1 were measured. The samples were extracted with hot water, 60% acetone or 80% ethanol for screening physiological activity. The crude protein content (4.36%) was found in the Miryangdlkkae 25 and calcium content (497.5 mg%) was found in the Namcheondlggae among the tested 4 perilla leaves. Fructose was 30.86 mg% in the Namcheondlggae and free amino acids at all perilla leaves was detected seventeen. In Boradlggae, glutamic acid and alanin were 25.37 and 11.91 mg%. Totally nine non-volatile organic acids were also detected and the contents of malic acid and glutaric acid were 28.34 and 14.57 mg% in Boradlggae. The Miryangdlkkae 25 had the highest vitamin C amount which was 113.24 mg%. Angiotensin converting enzyme (ACE) inhibition activity of 80% ethanol extract of Boradlggae was 46.71%. Electron donating activity of 60% acetone extract from Namcheondlggae was the strongest inhibition activity as 98.19% when 200ppm level of the sample extracts were added.
한국산 배의 Polyphenol 화합물군이 쥐의 면역기능에 미치는 영향

This study was conducted to investigate immunofunctional activity of the polyphenol fractions isolated from Korean pear. In the experimental of Rosette forming cell, the results showed that all the polyphenol fractions enhance the cell count compared with the control group. Especially polyphenol fraction II and III showed highly significant effect on Rosette forming cell, and allergy inhibition. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.
