Effect of Cartilage Repair of Modified Gellan Gum Hydrogel by Cation Exchange Resin

Gellan gum is widely applied in tissue engineering owing to its biocompatibility and outstanding physicochemical properties and recently it became recognized polymer that spotlight in various fields. In this study, we have fabricated modified gellan gum hydrogel scaffold using Cation Exchange Resin that remove cation (Mg<sup>+2</sup> and Ca<sup>+2</sup>) to observe the effect of cartilage regeneration. Analysis can be used to provide insights into the modified gellan gum of gel materials removing cations. The physiochemical and biological characteristics of scaffold was evaluated by Compressive strength, FT-IR, MTT, Bio-SEM and RT-PCR. The measurement of tissue regeneration test was conducted with New Zealand white rabbits and Nude mouse to confirm modified gellan gum’s effect of cartilage repair in vivo.

For microstructural analysis of a friction stir welded (FSWed) joint of advanced high-strength steels, dual phase (DP) and complex phase (CP) steels, are studied. FSWed joints are successfully fabricated in the following four cases: (i) DP/DP; (ii) CP/CP; (iii) DP/CP, where the advancing side is DP and the retreating side is CP; (iv) CP/DP, where the advancing side is CP and the retreating side is DP. The stir zone (SZ) of (i) the DP/DP joint mainly consists of lath martensite, while the stir zone of (ii) the CP/CP joint consists not only of lath martensite but also of bainite. In the case of (iii) DP/CP and (iv) CP/DP, they exhibit a similar microstructure including acicular-shaped phases in the joints; however, cross-sections of the joints show differences in material mixing in each case. In (iv) the CP/DP joint, temperature towards the CP steel is sufficient to cause softening, thus leading to better mixing than that in (iii) DP/CP. The phases of the SZ in each of the four cases are formed by phase transformation during the FSWed process; however, the transformed phase volume fraction of CP steel is lower than that of DP steel, indicating that dynamic recrystallization occurs mainly in CP steel. The hardness values of the SZ are significantly higher than those of the base materials, especially, the SZ of (iii) the DP/CP joint has the highest value due to highest fraction of lath martensite.

The purpose of this study is to characterize Gallus gallus var Domesticus (Yeonsan ogolgye) demineralized bone powder. DBP have showed to promote the formation of red blood cells in the bone marrow and other medical effects. Also, the melanin extracted together with HAp from the GD has a positive effect in osteoblast differentiation. For the analysis of DBP, DNA quantification test, histochemical staining were conducted to Evaluate the Morphological characterization with optical microscopy. For the analysis of composed scaffold, SEM, compression test, FT-IR, XRD were measured. Collectively, in the case of DBP, The presence of melanin and Collagen type-I in DBP was confirmed. And in the case of scaffolds, composed scaffolds showed a suitable macro- and microporosity and high interconnectivity to support cells adhesion, proliferation.

Omeprazole (OM), typically used for patients with gastric problems, is a proton pump inhibitor, which reduces the amount of stomach acid production effectively by regulating the proton pump. However, the main side effect of OM is the calcium indigestion, which may cause the calcium deficiency in the long term. Seaweeds are abundant in calcium (approximately 560-1200 mg per 100 g) and nutrients such as vitamins (approximately 150-400 mg per 1 kg) and minerals. In particular, high level of magnesium in the seaweed can assist the absorption of calcium. Accordingly, seaweeds can be a good source of nutrients to support OM therapy. Herein, we produced a solid dispersion (SD) which is composed of OM and seaweed calcium (SC) and can be suitable for the oral treatment, via the ultra-multiple crystal method. We also optimized the film formulation so that the SD could be absorbed quickly and efficiently by the oral cavity. Due to many advantages of using the oral thin film (OTF), we chose to fabricate OTF with our optimized dispersion method. OTF allows for both the rapid and efficient dissolution within the oral cavity and the protection of the active ingredient (i.e., OM) from rapid degradation under acidic conditions. The prepared SD samples were characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD). Thermal analysis of SD was conducted by differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). Hydrophobicity of SD was evaluated by the contact angle. Furthermore, the dissolution test and the HPLC analysis of SD were performed.

Dapoxetine Hydrochloride (DH) have been developed for the treatment of premature ejaculation for men. DH promotes the ejaculatory delay as the selective serotonin inhibitor. However, it is necessary to delay time about 1-3 hours to absorb drug after oral administration. Therefore, rapid action is needed to reduce DH’s delay time. In this study, after making the Solid Dispersion (SD) with L-arginine (L-A) using rotary evaporation, we made Oral Thin Film (OTF) formulation for the immediately SD’s release. Characterizations of the samples were performed to analyze the morphology, crystallinity, the moecular structure by Scanning Electron Microscopy (SEM), Fourier Transform Infrared Spectroscopy (FTIR) and Powder X-Ray Diffractometer (PXRD). And, thermal porperty was also measured by Differential Scanning Calorimeter (DSC). Then, the dissolution test was used to analyze dissolution rate of the immediately released formulation via Batch (BAT). In this results, OTF is good formulation against the others and the BAT 3 is more dissoluble than other BATs. So, it is useful to cure the premature ejaculation.

최근 유전 및 노화로 인해 관절염(류마티스, 퇴행성 관절염 등)으로 고통받는 환자들이 증가하고 있으며, 이들 대부분의 환자들은 지속적인 통증으로 인해 건강 관련 삶의 질(HQRL)이 저하되고 있다. 통증 치료를 위해서는 비스테로이드성 항염증제(NSAIDs)가 널리 사용되고 있으며, 대표적인 치료제로 나프록센 나트륨(NAP)이 사용된다. NAP는 발열, 염증, 통증을 일으키는 프로스타글란딘(PG)의 생성에 관여하는 효소인 사이클로옥시게나제 1과 2(COX-1, COX-2)를 억제하는 장점이 있으며, 상대적으로 반감기가 12시간이다. 그러나 NAP는 하루에 2번 275-550 mg의 경구 투여에 의해 용해도가 낮고 흡수도가 높은 생물약제학적 분류체계 c la ss 2 약물이다. 나프록센 나트륨의 낮은 용해도를 개선하기 위해 수용성 고분자인 폴리비닐피롤리돈(PVP-K30), 폴리비닐알콜(PVA), 전분과 NAP를 습식과립법으로 과립을 제조하였다. 이후 NAP의 향상된 형태와 용해도를 확인하고자 주사전자현미경(SEM), 시차주사 열량분석법(DSC), X선 회절분석법(XRD), 푸리에 변환 적외선 분광학(FTIR) 및 고성능 액체 크로마토그래피(HPLC)를 통해 확인하였다. 이러한 결과를 바탕으로 습식 과립 형성을 통해 NAP의 용해도가 향상되었음을 보여주었고, 즉시 방출 효과인 속방정으로써 관절염에 대한 치료제로 응용 가능하다는 것을 보여주었다. Recently, many patients are suffered from arthritis including osteoarthritis, rheumatoid arthritis due to increment in genetic disease and aging. Most patients with arthritis have a relatively low health-related quality of life (HRQL) due to persistent pain. For the treatment of pain, non-steroidal anti-inflammatory drugs (NSAIDs) are widely used and among them, naproxen sodium (NAP) is typically applied. NAP has merits in inhibiting cyclooxygenases 1 and 2 (COX-1, COX-2), enzymes involved in the production of prostaglandin (PG) which cause fever, inflammation, and pain, and has a relatively long half-life of 12 h. However, NAP is a biopharmaceutics classification system (BCS) class 2 drugs with low solubility and high absorption by oral administration of 275~550 mg per time, twice a day. Herein, the low solubility of NAP was improved by wet granulation of the water-soluble polymer polyvinylpyrrolidone (PVP-K30), polyvinyl alcohol (PVA), starch, and NAP. The improved morphology and solubility of NAP was confirmed by scanning electron microscope (SEM), differential scanning calorimetry (DSC), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and high-performance liquid chromatography (HPLC). Overall results showed enhanced solubility of NAP through the wet granule formation and showed promising treatment for arthritis as immediate release medicinal effects.
