Antibacterial activities of magnolol (MGL) and honokiol (HKL) in combination with four representative antibiotics-amoxicillin (AMPC), oxytetracyclin (OTC), gentamicin (GM) and chloramphenicol (CAP)-were evaluated against four bacterial strains. When tested by disk-plate method, five out of eight combinations such as HKL-AMPC, HKL-CAP, MGL- AMPC, MGL-OTC, and MGL-CAP showed additive to synergistic interaction against gram- negative bacterium Salmonella typhimureum. Of these, MGL-AMPC combination turned out to be antagonistic against Sarcina lutea and Bacillus thurungiensis. Against these two grain-positive bacteria, only HKL-GM combination showed additivity to synergism. All the other combinations showed no interactions. Despite these results, however, no synergism was observed in checkerboard titration assay.

A protein-polysaccharide fraction of a Korean wild mushroomwas psathyrella velutina, PVP, was prepared and its antitumor immunomodulatory activity was investigated. When PVP was administered once daily for seven days from day 1 to day 7 into male ICR mice implanted with 1 ×105 cells of sarcoma 180 tumor cells into the peritoneum on day 4, it inhibited the growth of sarcoma 180 cells by 92.8%. In XTT assay; PVP also exherted in vitro anti-proliferation activity on U-937, a human monoblastoid cell line, as well as sarcoma 180 cells. PVP showed marked stimulatory activity on the immune system in that it induced the accumulation of PEC (the stimulation index, Sl=4.90 at 100 mgHg), stimulated the BALB/C mouse splenic Iymphocytes to form Iymphoblasts (Sl=5.75 at 100mcg/ml), and upregulated the expression of CD25 molecules. All these results strongly support that PVP exhorts its antitumor activity through stimulation of the immune system as well as antiproliferative activity on the tumor cells.
한국산 담자균류의 배양균사에 함유된 항암성분에 관한 연구

To develops new antitumor constituents from higher fungi, eight species of edible basidiomycetcs were shake-cultured and investigated for antitumor constituents using sarcoma 180 as a tumor cell and ICP mice as test animals. Among the tested species, Pluteus cervinus, Hydnum repandrm, Laccaria laccata, and Pleurotus pulmonarius were recognized to produce antitumor components and P. cervinus. was selected for further study. Four fractions were made from the mycelia and three from the culture filtrate of P. cervinus. Fach fraction was tested for its antitumor activity. The most active fraction was the water soluble fraction obtained from the hot water extract of the mycelia. The tumor inhibition ratio of this fraction were 73.9%, 86.3%, and 87.8%, when administered, i.p., at the dose level of 10㎎, 20㎎, and 50㎎/㎏/day for ten days. This antitumor fraction was subjected to gel filtration on Sephadex G-200 to be separated into two fractions. The higher molecular weight fraction which showed an anthrone positive sharp peak on Sephadex G-204 was fund to be active of the two and tenatively named cerviran A. When chemically assayed, cerviran A was found to be a protein-Bound polysaccharide. The polysaccharide moiety was composed of five sugars, and the protein moiety, 17 amino acids.
Phellinus linteus 균사 배양물로부터 분리한 단백다당체 Kp의 항암활성

A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation, Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 mg/kg. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation, Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 mg/kg. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 mcg/ml. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host''s immune system.
