        ERK1/2 activation in quercetin-treated BEAS-2B cell plays a role in Nrf2-driven HO-1 expression

        Quercetin, a member of the flavonoids,has been heralded as chemopreventive agent, generally because of its free radical-scavenging and antiproliferative activities. In this process, NF-E2-related factor-2 (Nrf2) plays a role in protecting cells from oxidative damage; however, the exact mechanisms in which bronchial epithelial cells respond to quercetin by activating Nrf2 are not fully understood. Here, we report the role of extracellular signal-regulated kinase (ERK) activation in response to quercetin. Quercetin treatment decreased the viability of human bronchial epithelial BEAS-2B cells in a dose- and time-dependent manner. ERK1/2 was activated within 10 min of quercetin addition and then remained increased above control level throughout 48 h, whereas AKT phosphorylation was suppressed from the first 1 h of quercetin treatment. Inhibition of Erk1/2 phosphorylation attenuated quercetin-induced loss of cell viability. Nrf2levels in both nuclear and whole cell lysates increased after quercetin treatment and was accompanied by the increase of heme oxygenase-1 (HO-1) protein level,indicating that increased HO-1 expression is Nrf2-mediated. Activation of the Nrf2/HO-1 system after quercetin treatment was suppressed by the pretreatment with MEK inhibitor, PD98059. Also, silencing Nrf2 with siRNA decreased cell viability and augmented cytotoxic effect of anticancer drug LBH589. Overall, our results indicate that quercetin induces the up-regulation of Nrf2-mediated HO-1 expression, at least in part, via MEK/ERK1/2 signaling, as an adaptive mechanism which may provide advantages for cell proliferation and enhance survival despite endogenous and/or exogenous oxidative insults.

        ‘녹색성장’이 정책의 최대 화두로 떠오르고 있다. 비단, ‘녹색성장은 한 국가의 일시적 정책홍보성 수준이 아닌 전 세계가 합심하여 공동으로 대응해야만 비로소 그 빛을 발할 수 있는 광범위하고 지속적인 관심과 노력을 필요로 하는 역사적 프로잭트라 하겠다. 녹색성장이 거대담론 차원의 수사로 그치지 않기 위해서는 각계각층에서 실행 가능한 계획과 전략을 수립하여 단계적으로 추진하여야 할 것이다. 그러나 궁극적으로 녹색성장이 산업혁명이나 it혁명처럼 우리의 삶을 질적으로 변화시킨 녹색혁명이란 역사적 평가를 받기 위해서는, 무엇보다도 녹색인재 육성에 힘을 쏟아야 하겠다. 녹색인재는 하루아침에 양성될 수 없다. 이 글은 영유아기부터 가정, 유치원, 어린이집, 놀이터 등 일상생활 속에서 녹색생활을 자연스럽게 경험하고 체화하면서 녹색인재로 육성할 것을 제안하였다.

        한국적 이미지를 활용한 평창 동계올림픽의 유니폼 제작에 관한 연구

        The aim of this study is to create a uniform for the Olympic team of South Korea, the host of the Pyeongchang 2018 Winter Olympic Games, both in the form of a design as well as in life-sized pieces, which will utilize Korean images. In terms of the methodology, based on previous studies that utilized Korean images, as well as personal research on the Winter Olympics and its uniforms, a uniform for the Korean Winter Olympics team was created by utilizing shapes of Korean images in dangui, the colors and patterns of dancheong (traditional Korean paintwork), chokagbo(cloths of patchwork), and embroidery. 8 kinds of designs of Korean images for Pyeongchang 2018 Winter Olympics were developed, which reflected the symbolic, functional, and aesthetic characteristics of uniforms, and of these, 3 uniforms were manufactured at full scale. <Work 1> was manufactured in the form of a down jacket for women with the application shapes and colors of dangui. <Work 2> was manufactured in a padded jacket with the application of 7 colors of dancheong. In order to make the dancheong colors look clearer and more beautiful, the design used a near-black color on the body. <Work 3> was manufactured in the form of a down jumper for men through the application of the shapes and colors of chokagbo. Irregular faces and lines in which the sizes of faces gradually change were utilized, and in order to give a more refined sense, various demi-tints were used. For the symbolic characteristics of the uniform, the national flag of Korea and wappen were attached, and the emblem of Pyeongchang Winter Olympics was embroidered.. This is expected to enhance the value of the nation's brand and propagate the beauty of traditional Korean costume all over the world by creating uniform designs for Pyeongchang 2018 Winter Olympics that incorporate images of Korea, for the uniform that will be used as the official uniform for PyeongChang 2018 Winter Olympics.

        Knockdown of cysteine-rich 61 inhibits proliferation, migration, and invasiveness of prostate carcinoma PC-3 cells

        The overexpression of wild-type cysteine-rich 61 (Cyr61) is associated with the aggressiveness of cancer and poor prognosis in different human cancers. The aim of this study was to examine the expression of Cyr61 protein in prostate adenocarcinomas and to investigate the effects of inhibiting Cyr61 expression using small interfering RNA on the proliferation, migration, invasiveness, and sensitivity of prostate carcinoma PC-3 cells to TNF-related apoptosis-inducing ligand. Cell proliferation was measured by XTT assay. Colony formation assay, wound healing assay, and Matrigel invasion assay were also examined. Immunohistochemical staining of prostate tumor tissues showed that prostate carcinomas had significantly increased Cyr61 level compared with benign glands adjacent to carcinoma. siRNA-based knockdown of Cyr61 resulted in decreased cellular proliferation, clonogenicity, migration, and invasion. At the same time, Cyr61 silencing effectively reduced the levels of phosphorylated Akt and integrin-b3. Cyr61-specific siRNA combined with TRAIL increased the apoptosis of PC-3 cells and the cleavage of apoptosis hallmarkers such as PARP and caspase-3. Taken together, our data provide evidence that Cyr61 modulates integrinb 3 level as well as PI3-kinase/Akt activity through which Cyr61 may mediate tumorigenesis, and suggest the potential importance of Cyr61 targeting on enhancing the therapeutic efficacy of prostate cancer.

        사회적 일자리를 통한 은퇴 고령자 일경험의 의미: Modified Grounded Theory

        The purpose of this study is to explore deeply the meaning of the work experience of retired elderly people in the process of career maintenance through social employment. In order to achieve the purpose of this study, eight elderly people who have been engaged in schools to protect school violence were conducted in-depth interviews. The Modified Grounded Theory Approach proposed by Kinoshita was used. The collected qualitative materials were analyzed by utilizing the work analysis sheets, and three categories were derived as follow. Three categories are first, the necessity of continuing work, second, interaction with the work environment, third, consistent socialization and Life-span development. Based on these results, the meaning of the work experience of the elderly retired following. First, elderly people's job conducted role as a bridge connecting the past, present, and future. Second, they meant establishing the processes of self concept as a social member in old age. Third, sustainable development and socialization in old age were promoted as a mechanisms to adapt to changes in life. 본 연구의 목적은 은퇴 고령자의 사회적 일자리를 통한 직업유지과정에서 일경험의 의미를 심층적으로 탐색하는 것이다. 이를 위해 Strauss와 Corbin(1990)의 근거이론을 근본으로 기노시다(木下康仁)가 제안한 Modified Grounded Theory Approach의 분석방법을 활용하여 은퇴 고령자의 인적자본활용이 주로 이루어지는 학교 보안관과 배움터지킴이 업무에 종사하는 8명을 심층인터뷰하였다. 작업분석시트를 활용하여 동시다중적 분석을 실시하였으며, 일지속의 당위성, 환경과의 상호작용, 지속적 사회화와 발달로 범주화하였다. 분석결과 은퇴 고령자들은 일을 지속할 수밖에 없는 내·외부적 당위성을 가지며, 일을 통해 지속적이고 역동적으로 환경과 상호작용을 하며, 이를 통해 노년기에도 지속적 사회화와 발달과정을 거친다는 것을 알 수 있었다. 이러한 결과를 바탕으로 은퇴 고령자의 직업유지과정에서 일경험은 첫째, 과거, 현재, 미래 삶을 연결하는 가교적 역할을 한다. 둘째, 노년기 사회구성원으로서 자아개념을 확립하는 과정의 의미를 갖는다. 셋째, 변화한 삶에 적응하는 기제로 지속적인 사회화와 발달을 촉진한다.

        Glutary 7-Aminodeacetoxycephalosporanis Acid Acylase 생산균의 분리 및 동정

        Glutaryl 7-aminodeacetoxycephalosporanic acid(GL-7-ADCA)로부터 7-aminodeacetoxycephalosporanic acid(7-ADCA)를 생성하는 GL-7-ADCA acylase생산균을 자연계에서 폭넓게 검색하여 한균주를 선정하고 분리균의 형태 및 생리적 성질들을 조사하여 Alcaligenes sp.로 동정하였다. 분리균의 효소생산을 위한 배양조건을 검토한 결과 탄소원으로는 glucose, 질소원으로는 yeast extract, monosodium L-glutamate가 우수하였다. 효소의 작용최적 pH는 8.0이며, pH7.0~pH11.0 번위에서 비교적 안정하였다. 5l jar fermentor에서 배양경과를 조사한 결과 효소이 활성은 $37^{\circ}C$, 교반속도 300rpm, 통기량 1vvm 조건에서 20시간~30시간 배양시 가장 높았다. Microorganism producing glutaryl 7-aminodeacetoxycephalosporanic acid (GL-7-ADCA) acylase was screened from soil. The microorganism was identified as Alcaligenes sp. J-421 by its morphology and biochemical properties. Cultural conditions of Alcaligenes sp. J-421 were investigated for the production of GL-7-ADCA acylase. Optimum medium composition was 1% glucose, 1% beef extract, 0.5% yeast extract, 0.2% monosodium L-glutamate, 0.1% glutaric acid, 0.2% NaCl, 0.5% $K_2$ $HPO_4$, and 0.05% $CuSO _4{\cdot}5H_2O$. Optimum cultivation conditions for the production of the enzyme in 5 l jar fermentor were $37^{\circ}C$, tip speed 300 rpm, aeration 1 vvm. Optimum reaction pH of the enzyme was 8.0 and the enzyme was stable at pH7.0-11.0.
