Overexpression of KAI1 Protein in Diabetic Skin Tissues

Background: Patients with diabetes mellitus often have a difficult life, suffering from foot ulceration or amputation. Diabetes is characterized by chronic inflammation, and one of the features of inflammation is hypoxia. Recently, it has been reported that KAI1 is a hypoxia target gene. There is no published research on hypoxia-related KAI1 protein levels in human diabetic skin. Therefore, we have investigated the expression of KAI1 protein in diabetic skin tissue in vivo. Methods: The expression of KAI1 protein was evaluated by western blotting in 6 diabetic skin tissue samples and 6 normal skin samples. Immunohistochemical staining was carried out to identify KAI1 expression. Results: The western blotting revealed significantly increased expression of the KAI1 protein in diabetic skin tissues as compared to normal skin tissues. Immunohistochemical examination demonstrated that KAI1 was expressed in all diabetic skin tissues with moderate-to-strong positivity and weakly expressed in normal skin tissues. Conclusions: Our data suggest that a high expression of the KAI1 protein can be observed in diabetic skin tissue. To the best of our knowledge, this is the first report suggesting that KAI1 protein expression in diabetic skin tissues may be associated with chronic inflammatory states and hypoxia.
경구 Ibupenta와 2.5% Indomethacin, 0.25%Desoxymethasone 연고가 UVB로 인한 홍반에 미치는 효과에 대한 연구

The large amount of UVB irradiation of human skin cause acute erythema that is induced by prostaglandin. Ibupenta and indomethacin have antiinflammatory effect by inhibiting prostaglandin pathway. The purpose of this study was to investigate the effect of oral ibupenta and topical 2.5% indomethacin, 0.25% desoxymethasone ointment on erythema induced by UVB irradiation. In this study, healthy sixteen male medical students who did not have any experience of tanning on their back during the last year were included in this study. There 32 volunteers were devided into two groups, oral ibupenta was administrated to one group and not to the other. The minimal erythema dose(MED) of each volunteer was measured with colorimeter. According to the individual's MED, 1 MED, 2 MED,and 3 MED of UVB was irradiated to the volunteer's back, the erythemas before and after the applying the topical agents were evaluated with naked eyes and colorimeter. In group I applied by 2.5% indomethacin, 0.25% desoxymethasone without oral ibupenta, erythemas tested with 2.5% indomethacin were statistically significantly supressed according to data measured by colorimeter. In group Ⅱ applied by 2.5% indomethacin, 0.25% desoxymethasone with oral Ibupenta, erythemas tested with 2.5% indomethacin and 0.25% desoxymethasone were statistically significantly supressed according to data measured by colorimeter. The maximal supression of UVB induced erythema was observed in 2.5% indomethacin with oral ibupenta. 'These data indicate that oral ibupenta, as an inhibitor of prostaglandin, is more effective than topical indometahcin or topical desoxymethasone in inhibition of UVB induced erythema. There is a synergistic effect between oral ibupenta and topical desoxymethasone. This result indicate that 2.5% indomethacin with oral ibupenta is useful therapy in acute UVB induced erythema within 24 hours. To evaluate the erythema, colorimeter is supposed to be more sensitive than naked eye score.
악성흑색종에서 Melanin 세포 이동에 미치는 Endothelin-1 효과에 관한 연구

Endothelin-1 is a 21 amino acid vasoconstrictor peptide originally isolated from endothelial cells and keratinocytes. There are three isotype ET-1, ET-2 and ET-3. Only ET-1 involve human skin. Yada group indicated that human melanocyte possesed specific high affinity receptors for ET-1 and that ET-1 stimulated melanocyte proliferation and migratin. The purpose of this study was to determine weather endothelin-1 could enhance melanocyte migration or cell proliferation in malignatn melanoma. We performed Boyden Chamber Checkboard analysis by using control, 20nM, 40nM and 80nM ET-1 concentration in SK 28 cell line and SK30 cell line. The data were represented as percent of cell migration and statistical student's t-test was done. The following results were obtained from this study ; 1. In SK 28 cell line, the percentage of cell migration noted 100.04±10.12% at control, 248.86±6.91% at 20nM, 265.24±9.73% at 40nM and 285.15±8.34 at 80nM. Et-1 significantly (p<0.01) enhanced cell movement at the concentration of 20, 40, and 80 nM. 2. In SK 30 cell line, the percentage of cell migration noted 100.04±10.12% at control, 282.86±8.26% at 20nM, 290.86±7.05% at 40nM and 263.86±4.27 at 80nM. ET-1 significantly (p<0.01) enhanced cell movement at the concentration of 20,40 and 80 nM.
