포장 조건에서 CMVP0-CP 형질전환 고추 도입유전자의 지속성 조사

This study was conducted to evaluate the persistence of DNA in the transgenic chili pepper resistant to cucumber mosaic virus (CMV) in the field condition. We analyzed the persistence of genes in the leaf samples obtained from two field conditions, below and above soil. Transgenic and non-transgenic leaf tissues were buried in the soil at a depth of 10 cm or placed on the soil surface. Qualitative and quantitative PCR analysis showed that the amount of transferred genes from the transgenic peppers below and above soil was dropped to 28.3-42.7% one month after buried and it was rapidly reduced to 0.9-3.3% after two months. The transgenes were not detected three to four month after buried. In addition, DNA of the leaves placed below soil decomposed about 8% more than those on soil surface. The gene transfer from decomposing leaves of the transgenic pepper to soil was investigated by PCR analysis with the soil attached to the samples. The PCR result indicated that the gene transfer from the transgenic pepper to soil was not occurred.

유전자변형생물체(Genetically Modified Organisms, GMOs)는 식량, 환경, 에너지부족 등 인류가 직면하고 있는 각종 문제점을 해결하기 위한 대안으로 그 개발 및 이용이 가속화되고 있다. 특히 몇몇 국내 개발 GM 작물은 안전성평가 단계에 있어서 조만간 실용화될 것으로 전망되고 있다. GMO에 대한 일반인들의 인식조사 연구는 대부분 소비자를 대상으로 진행 되어왔으며 실제 GM 작물이 상업화되었을 때 이를 재배할 경작자에 대한 인식조사 연구는 부족한 실정이다. 본 연구에서는 농업인의 GMO에 대한 인식 파악 및 재배의도를 알아보기 위해 설문조사와 표적집단면접(FGI)를 병행하였다. 설문조사는 논산, 김천, 충주, 상주 지역의 농민 747명을 대상으로 실시하였으며, FGI는 농업분야에 종사하고 있는 남녀 농업인 5명을 대상으로 수행하였다. 조사 결과 GM 작물에 대한 인지 수준과 지식수준은 각각 85.5%, 65%로 일반인과 크게 다르지 않았다. GM 작물에 대한 정보 실태로써 ‘알고 있거나 들어 본적이 있는 GM작물 종류’에 대해 ‘수량증진’에 대한 응답이 가장 높았으며, ‘병저항성’, ‘해충저항성’ 순으로 조사되었다. GM 작물에 대한 인식은 ‘긍정적’과 ‘부정적’ 응답이 비슷한 분포를 나타냈으나, GMO에 대한 섭취 및 재배 의향은 전반적으로 부정적인 것으로 조사되었다. 특히 GMO의 재배보다는 섭취에 더 부정적인 것으로 나타났다. GM 작물 형질에 대한 선호도로는 ‘병 저항성’ 형질이 가장 높았으나 ‘맛’, ‘수량’, ‘건강개선’ 등의 형질에 대해서도 고른 응답을 보였다. The farmers` perception on genetically modified (GM) crops is one of the important factors for the development and commercialization of GM crops. Although many researches have usually studied on the consumers` perception on GMOs, the farmers` perception has not been studied much in Korea. In this study, a survey was conducted from Dec. 2009 to Jan. 2010 to investigate Korean farmers` perception and cultivating willingness of GM crops. The survey data were collected from 717 farmers who lived in four Korean cities (Nonsan, Kimcheon, Chungju, and Sangju) using standardized questionnaires. Results showed that 86% of the respondents were already exposed to the information related to GM crops. The reason of GM crops development that respondents had heard were ‘increased productivity’ (31%), ‘disease resistance’ (21%), ‘insect resistance’ (18%), and so on. The biggest information source was ‘TV/radio’ (50%), and the main information content was ‘concern about food safety’ (45%). More than half of respondents perceived that GM crops could be beneficial to people, however, 61% of them concerned about GM food safety at the same time. Thirty nine percent of farmers showed positive attitudes and were interested in cultivating GM crops, and the preferred GM traits was ‘disease resistance’ (20%), ‘good to health’ (18%), ‘increased productivity’ (16%), ‘taste & flavor improvement’ (14%).

The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world. Concerns about the potential risks of GM crops are also increasing. Safe management for the development and production of GM crops is required according to Living Modified Organism Act in Korea. Planning about the methods, duration, and frequency of environmental monitoring is also required for commercial use of GM crops. GM Zoysia japonica Steud. (event name: JG21) expressing resistance to glufosinate-ammonium has been generated previously. By using gamma ray treatment to JG21 we also developed male sterility and dwarf Z. japonica (event name: JG21-MS). The objective of this study was to establish the monitoring system for environment release of JG21-MS. In this study we extracted RNA from JG21 and JG21-MS and conducted RAPD (random amplified polymorphic DNA) method to distinguish JG21 and JG21-MS.

The possible gene transfer from transgenic chili pepper to soil bacteria in rhizosphere was evaluated. DNA was extracted from rhizosphere soil collected from growing transgenic chili pepper (Capsicum annuum L.) resistant to cucumber mosaic virus (CMV) and control line, and PCR analysis was performed with specific primer for transgene in the transgenic chili pepper. The results of the PCR targeting NPTII gene indicated that the transgene was present in the rhizosphere soil samples in all collecting times. To investigate the cause of the transgene detection in the samples, we isolated microorganisms from the soil and cultured them on bacterial and fungal medium containing kanamycin. About 430 thousands of bacterial colonies and 160 thousands of fungal colonies were investigated, however, colonies resistant to kanamycin were not detected, indicating gene transfer from transgenic chili pepper to culturable rhizosphere soil microorganisms was not occurred in the present study.

The aim of this study is to consider the effect hydrogen on dezincification behavior of Cu-Zn alloys. The investigations include microstructural observations with scanning electron microscope and chemical composition analysis with energy dispersive spectrometer. The dezincification layer was found to occur in high pressure hydrogen atmosphere, not in air atmosphere. In addition, the layers penetrated into the inner side along the grain boundaries in the case of hydrogen condition. The shape of the dezincification layers was porous because of Zn dissolution from the ${\alpha}$ or ${\beta}$ phase. In the case of stress corrosion cracks formed in the Cu-Zn microstructure, the dezincification phenomenon with porous voids was also accompanied by grain boundary cracking.

This study investigated the surface pit corrosion of SS420J2 stainless steel accompanied by intergranular crack. To reveal the causes of surface pits and cracks, OM, SEM, and TEM analyses of the microstructures of the utilized SS420J2 were performed, as was simulated heat treatment. The intergranular cracks were found to have been induced by a grain boundary carbide of $(Cr,Fe)_{23}C_6$, which was identified by SEM/EDS and TEM diffraction analyses. The mechanism of grain boundary sensitization occurred at the position of the carbide, followed by its occurrence at the Cr depleted zone. The grain boundary carbide of $(Cr,Fe)_{23}C_6$ type precipitated during air cooling condition after a $1038^{\circ}C$ solid solution treatment. The carbide precipitate formation also accelerated at the band structure formed by cold working. Therefore, using manufacturing processes of cooling and cold working, it is difficult to protect SS420J2 stainless steel against surface pit corrosion. Several counter plans to fight pit corrosion by sensitization were suggested, involving alloying and manufacturing processes.

The aim of this paper is to consider the effect of the manufacturing processes on corrosion and centerline cracking of ancient bronze spoons. The ancient bronze spoons in question were made by several steps of forging, in reheated condition with cast ingots. The manufacturing method is similar to that of the modern spoons. The investigations include observations from light and scanning electron microscopes of the microstructure in terms of the crack propagation. Cracks in the centerline are caused by solute segregation in the center-line region; this solute is solidified in the final stage of bronze spoon manufacture. Centerline cracking is also caused by ${\alpha}$ phase segregation, accompanied by forged overlapping along the longitudinal direction of the spoons. A vertical stripe with cracks along the centerline of the spoon's width is formed by folding in the wrought process. The overlapping area causes crack propagation with severe corrosion on the spoon surfaces over a period of a thousand years. The failure mechanisms of ancient bronze spoons may be similar to that of modern spoons, and the estimation of the failure mechanisms of ancient spoons can be appropriate to determine failure causes for such modern spoons.
ADINA & WINLIFE 활용한 압력용기 용접부 피로파괴 해석

The aim of this study is to consider effect of weld elastic modulus on simulations of stress concentration and fatigue life for pressure vessel. The investigations include analysis with ADINA and WINLIFE softwares for whole body model about using condition of the boiler vessel. Values of weld elastic modulus were divided by 5 steps in butt weld area of the boiler vessel body. The stress concentration of the butt weld more was increased in case of higher elastic modulus of weld area because of higher difference of material properties between matrix and weld. It was concluded that the fatigue lives were decreased along increasing stress concentration due to high elastic modulus of weld. The matrix microstructure was estimated as pearlitic structure of α·ferrite and pearlite. And the microstructures of welds along 5 steps of elastic modulus were estimated as bainitic fine pearlite and martensite as increasing elastic modulus.
