Vibrion vulnificus is an estuarine gram0negative human pathogen that affects people with chronic hepatitis, alcoholic cirrhosis, diabetes mellitus or other underlying diseases, V. vulnificus infection is mediated primarily by consumption of raw fish or by exposure of pre-existing wounds to seawater, causing permanent tissue damages or fatal septic shock. We have been developing a vaccine against V. vulnificus composed of whole cell lysate of a V. vulnificus O-antigen serotype 4 strain. Oral administration of the V. vulnificus vaccine elicited a high serum antibody response in rabbits. The induced antibodies were reactive not only to the homologous strain but also to heterologous O-antigen serotype strains, indicating cross-reactivities among serotypes. Western blot analysis revealed that the antibodies are mainly specific for outer membrane proteins (OMPs) and reacted equally well with OPMs purified from 9 O-antigen serotypes The rabbit antisera showed opsonophagocytic killing activity against heterologous strains as well as the homologous strain. Passively transferred rabbit antisera into mice were protective against a lethal V. vulnificus infection. These data demonstrate that oral administration of the V. vulnificus infections, suggesting that this vaccine preparation could be used to develop an oral vaccine against V. vulnificus. The thymus weight of the mouse was 54.1% in protein malnutritive diet group and 39.2% in group treated with saponin fraction of Panax ginseng in comparison to normal diet group. This decreasing effects of protein malnutritive diet and saponin fraction on the thymus weight practically disappeared after four weeks. The saponin fraction showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total peritoneal exudate cells, content of total serum protein and albumin content of the mouse, each 45, 8 and 10% respectively in comparison to that of normal diet group. And these values in protein malnutritive diet group were 61.2, 83.6 and 87.O% respectively in comparison to that of normal diet group, and recovered to the level of normal diet group by the supplement of the saponin fraction. The electrophoregram of the serum protein of the mouse fed with protein malnutritive diet was different from that of the mouse fed with normal diet, but this difference practically disappeared by the supplement of the saponin fraction.

Immunopotentiating activity of water extract of Lycii Fructus, Rubus Fructus, Torilis Fructus, Schizandrae Fructus and Cuscutae Semen were investigated using mouse fed with normal diet and protein deficient diet. Body weight, ratio of spleen or thymus to body weight, number of total peritoneal exudate cell, content of total serum protein and albumin content were decreased in the mouse fed with protein deficient diet as compared to the mouse fed with normal diet. Regardless of the nutritional condition, the supplement of the water extract of Lycii, Rubus, Torilis, Schizandrae Fructus or Cuscutae Semen increased the ratio of spleen to body weight. The increment of this ratio was significant by the supplement of water extracts of Lycii and Torilis Fructus in mouse fed with protein deficient diet. The supplement of water extracts of Torilis Fructus to the mouse fed with protein deficient diet increased the content of total serum protein and albumin content significantly. Similar results were obtained with Lycii, Rubus, Schizandrae Fructus and Cuscutae Semen. The electrophorogram shows the difference in the concentration of some serum protein constituents between the mouse fed with normal diet and protein deficient diet.

The changes of thymus weight, spleen weight, total number of peritoneal exudate cells, contents of total serum protein and albumin were measured and the change on the electrophoregram pattern of serum protein was observed in order to study on the effects of the saponin fraction of Panax ginseng as a tonic on the immune system of the mouse fed with protein deficient diet. The involution of the thymus of the mouse was induced by feeding protein deficient diet and the saponin fraction on the thymus weight was diminished after four weeks. The saponin fraction of Panax ginseng showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total number of peritoneal exudate cells and contents of total serum protein and albumin of the mouse, and overcame the effects caused by protein deficiency. The electrophoregram of serum protein of the mouse fed with protein deficient diet was different from that of the mouse fed with normal diet, but this difference seemed to be overcome by the supplement of the saponin fraction of Panax ginseng.

Mice were fed on normal diet(18% casein) or protein deficient diet(5% casein) with supplementation of the total saponin fraction of Panax ginseng for 2weeks, 4weeks and 8weeks, respecaively. The amounts of norepinephrine in the mouse brain were increased by the supplementation of the total saponin fraction of Panax ginseng in both normal and protein deficient diet fed groups. The amounts of dopamine in the mouse brain were only increased in the mouse fed with normal diet supplemented with the total saponin fraction of Panax ginseng for 8weeks. Norepinephrinergic neurons were more influenced than dopaminergic neurons by Panax ginseng and this influence was more amplified in the case of protein deficient diet.

We have been developing a vaccine (CJ-50002) against Vibrio vulnificus, composed of whole cell lysate of a V. vulnificus O-antigen serotype 4-strain. In order to evaluate the mutagenic potential of CJ-50002, 3 sets of mutagenicity tests were performed. In the reverse mutation test using Salmonella typhimurium TA1535, TA1537, TA98 and TA100, CJ-50002 did not increase the number of revertant at any concentration tested in this study (309.6, 154.8, 77.4, 38.7, 19.4 and 9.7 ㎍/plate). CJ-50002, at concentrations of 309.6, 154.8 and 77.4 ㎍/ml, did not increase the number of cells having structural or numerical chromosome aberration in cytogenic test using Chinese Hamster Lung cells. In mouse micronucleus test, no significant increase in the occurrence of micro nucleated polychromatic erythrocytes was observed in ICR male mice orally administered with CJ-50002 at the doses of 500, 250 and 125 mg/kg. These results indicate that CJ-50002 has no mutagenic potential in these in vitro and in vivo systems.

Effects of water extracts of Torilis Fructus, Rubus hructus, Schizandrae Fructus, Lycii Fructus and Cuscutae Semen on the immune system of ICR mice fed with normal diet and fed with protein deficient were investigated. In control group, body weight, the ratio of spleen per body weight, the ratio of thymus per body weight, peritoneal exudate cell number, total serum protein and albumin content were decreased in case of mice fed with protein deficient diet as compared to the mice fed with normal diet. Without regard to kinds of diet, water extracts of Torilis Fructus, Rubus the ratio of spleen per body weight and total serum protein and albumin content. Characteristic electrophoresis patterns of serum protein were obtained according to kinds of diet and sample.

CJ-50002 is an oral vaccine against V. vulnificus infection composed of whole cell lysate of V. vulnificus. The general pharmacological properties of CJ-50002 were evaluated in various animals and in vitro system. CJ-50002 at oral doses of 0.2, 2 and 20 mg/kg had no effect on general behavior in mice, chemoand electro-convulsions in mice, writhing syndrome induced by acetic acid in mice, body temperature in rats, charcoal meal propulsion in mice and urine and electrolytes excretion in rats. However, oral administration of CJ-50002 at dose of 20 mg/kg prolonged the hexobarbital-inuced sleeping inducing time in mice. In anesthetized dogs, CJ-50002 showed no effect on blood pressure, heart rate and ECG but decreased the respiratory rate and femoral blood flow at dose of 20 mg/kg. p.o. CJ-50002 had no effect on the contractile response of the isolated guinea pig ileum to various spasmogen at concentrations of 0.2, 2 and 20 ㎍/ml, respectively. Since these pharmacological effects of CJ-50002 were observed at dose much greater than those in clinical use (approximately 0.16 mg/kg, p.o.), it is likely that this vaccine may be relatively free of undesirable effects in clinical practice.
