구기자나무 유식물체를 이용한 식물체의 재분화와 극성과의 관계

구기자나무로부터 캘러스의 형성과 shoot의 형성에 미치는 생장조절제의 영향을 연구한 결과 캘러스의 형성은 2,4-D를 포함하는 배지에서 효과적이었으나 shoot의 형성에는 억제효과를 보였다. Shoot의 형성은 0.1mg/L IAA + 0.1mg/L BA의 배지에서 형성률과 절편당 형성 개체수가 가장 높은 분화율을 보였다. 절편체의 부위에 극성에 다른 캘러스의 형성과 shoot의 재분화에 있어서는 자엽의 기부와 말단부를 20% 절제한 절편의 기부의 절단면에서는 shoot의 형성이, 하부의 절단면에서는 캘러스의 형성이 높았으며 시간이 경과함에 따라 하부에서도 shoot의 형성이 보였다. 재분화식물은 1/3 MS 배지에서 뿌리가 양호하게 발달하였으며 토양 순화과정에서도 90% 이상의 생존율을 보여 개화가 이루어졌다. This experiment was conducted out to investigate the effect of plant growth regulators on callus and shoot formation. The callus formation was effective on 1/2 MS medium containing 2,4-D, while shoot formation was suppressed. Shoot formation and differentiation were the highest in combination 0.1 mg/L of IAA and 0.1 mg/L of BA. The polarity of explants was investigated from cotyledon, which excised 20% of each basal and terminal parts. Formation of shoot was induced from excised ends of the basal part. In excised ends of the basal part, callus was induced vigorously and shoots were produced lately. Root induction was easily achieved in 1/3 MS medium from the adventitious shoot and more than 90% of regenerated plantlets acclimatized successfully and flowered normally.

L. longifilorum Georgia $\times$ L. elegans Kakutanohikari 잡종 배로부터 유식물체를 분화시키고, 이때 유도된 캘러스의 증식과 이 캘러스로부터 유식물체 분화과정을 조직학적으로 관찰하였다. 잡종 배는 배양한 60개 절편 중 48개 절편에서 86개체의 배주가 발아되었으며, 1 mg/L NAA가 캘러스로부터 식물체의 재분화에 가장 효과적이었으며, 재분화를 억제한 캘러스만을 증식시키기 위해서는 0.1 mg/L NAA + 1 mg/L BA가 가장 양호하였다. 또한 Pyridoxin-HCI을 첨가하지 않은 경우에는 유식물체의 재분화가 거의 보이지 않았으며 50 mg/L의 Pyridoxin-HCI을 첨가한 배지에서 유식물체의 재분화가 가장 효과적이었다. 배에서 발생된 캘러스의 원형성층 지역으로부터 체세포배가 형성되었으며, NAA가 전형성층 부분에서 배발생적 세포 또는 전배 발생의 증식을 자극하는 것으로 생각되었다. This study was performed to investigate regeneration of plants differentiated from hybrid embryos between L. longifilorum Georgia and L. elegans Kakutanohikari. In addition, proliferation of callus and process of differentiation were investigated by histological observation. The germination of hybrid embryos was observed in 86 individuals from 48 slice cultures. Plant regeneration was effective on a medium supplemented with 1 mg/L HPh, and only callus proliferation was the highest in combination of 0.1 mg/L HPh and 1 mg/L BA. Also, plant regeneration was the most effective on a medium supplemented with 50 mg/L pyridoxine. We concluded that somatic embryos were formed from procambium of callus and proliferation of embryonic or proembryonic cells were stimulated with NAA from procambial cells.

The stereocilia bundle has been mechanically modeled as three stereocilia linked with tip-links and side-links. The mechanical characteristics has been studied with respect to the variation of external force exerted on the longest stereocilia. Novel CNT array sensor which simulates the motion of stereocilia bundle has been modeled by considering the interaction between carbon nanotubes and fluid. Two patterns of CNT array such as in-line type and staggered type were examined to give maximum deflection. The optimal pitch between CNTs was given for the possible CNT array sensor through geometric optimization.

This paper presents the simulation results about the mechanical characteristics of the stereocilia bundle that is modeled as three stereocilia linked with tip-links and side-links. The effects of tip-links and side-links on bundle displacement and stiffness are given with respect to the variation of mechanical properties. The CNT array sensor that mimicks the motion of stereocilia bundle is modeled by considering the interaction between CNTs and fluid. The optimal pitch between CNTs was given for the possible CNT array sensor through geometrical optimization.

The mechanical model of the stereocilia bundle has been built using the finite element method. The stereocilia bundle was assumed to be isotropic and linearly elastic in mechanical properties. In the simple structure model, the deflection and the stiffness of the stereocilia bundle were calculated and examined with respect to the variation of the mechanical properties and the geometry. The maximum stiffness value was 6.894 mN/m and the deflections of the stereocilia were 3.55, 2.79, 2.09 ㎚ when the applied force was 10 pN and the Young's modulus of tip-link is 3 GPa. In the FSI model, the deflection of the stereocilia was investigated based upon the interaction between the stereocilia and the lymph fluid. The ratio of the stereocilia deflections was 3.8:2.1:1 which was less than that without considering the interaction between the stereocilia bundle and the lymph fluid.
홍경천 (Rhodiola sachalinensis A. Bor) 잎절편으로부터 식물체 재분화에 미치는 Cytokinin과 Putresine의 영향

홍경천(Rhodilola sachalinensis A. Bor)의 잎절편로부터 부정아를 유도하고자 BA와 kinetin을 처리한 결과, BA의 처리가 kinetin의 처리보다 부정아 유도와 신초의 생장에 양호한 것으로 나타났으며, 특히 BA 1 mg/L처리에서 부정아 유도빈도와 신초길이는 각각 71%와 3.0 mm로 가장 높게 나타났다. 또한 고빈도의 부정아를 유도하고자 다양한 농도의 putrescine를 처리한 결과, putrescine 100 mM 처리에서 부정아 유도빈도가 BA 1 mg/L 단용처리보다 약 20%가 증가되었고 신초길이 역시 약 10 mm가 증가된 것으로 나타났다. 이상의 결과에서 유도된 부정아는 생장조절물질이 포함되지 않은 1/2MS배지에서 발근과 신장이 정상적으로 이루어졌으며 약 7 cm 정도 생장한 유식물체는 peatmoss:sand가 1:1로 혼합된 상토에서 약 95%이상 정상적으로 순화되었다. The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

The miniaturization and integration are trend of modem blood analyses. Micro-Bio-Fluidics plays an important role in a micro blood analysis system. In this paper, analysis and design technology for blood analysis system is presented. Numerical simulations of a blood flow in micro separator and reservoir are conducted. As a result, we suggest on-chip micro separator, which performed plasma separation from whole blood in micro channels.
Flow Properties of Micro Column Packed with Perfusive Particles

In this work, perfusive particles are used to form a micro column in a microfluidic chip and flow properties of the micro column are investigated. The packing flow velocity and the column/particle size ratio are shown to be important parameters affecting the packing density of the micro column. Experimental results show that the effect of the column/particle size ratio on the flow resistance of the micro column is negligible. This contrasts with previous works on the effect of the column/particle size ratio on the total pressure drop across the column.
Power-Free Micro Blood Separator

In this study, micro blood separators capable of separating red blood cells and blood plasma using microstructure are fabricated and tested. Test results show the possibility of the separation method proposed in the present work. The micro blood separators do not require external energy for separation and the required whole blood for the separators is less than 1(μl). Therefore, the power-free separation method proposed in this work can be a good candidate for the blood separating component in miniaturized blood analysis systems.
