      • cDNA Cloning and Expression of a Human Interferon-$\gamma$ in E. coli

        인간의 peripheralblood lymphocyte에서 분리된 poly (A)$^+mRNA$를 사용하여 ${\lambda}gt$ 10 cDNA library를 제조하였으며, 화학적으로 합성한 oligonucleotide를 이용하여, 이 cDNA library로부터 IFN-$\gamma$ 유전자를 갖는 DNA을 분리했다. 분리된 clone 가운데 하나인 pBK04는 IFN-$\gamma$의 전체 염기서열을 갖고 있었다. IFN-$\gamma$를 E. coli내에서 발현시키기 위해 lpp promotor를 함유하고 있 는 pINIIIA3 vector를 사용하여 IFN-$\gamma$를 발현시킬 수 있는 재조합 plasmid ($pKS{\gamma}3B$)를 제조하였다. $pKS{\gamma}3B$로써 형질 변환된 bacterial cell을 antiviral activity에 의해 역가 분석을 해본 결과, 높은 정도로 IFN-$\gamma$를 발현시켰으며 이 IFN-$\gamma$는 천연의 IFN-$\gamma$에 대하여 특이성이 있는 monoclonal antibody에 의해서도 인지되었다. High copy number를 갖는 expression vector를 제조 (pHK-3) 하였으며, 이 high copy number vector를 사용하므로써 IFN-$\gamma$의 생성이 증가되는 사실을 확인하였다. A ${\lambda}gt10$ cDNA library constructed with poly $A^+$ mRNA from human peripheral blood lymphocytes was screened by a synthetic oligonucleotide having IFN-$\gamma$ sequence. One of the clones (pBK04) which hybridized to the oligomer was confirmed to have the whole IFN-$\gamma$ coding sequence. The eDNA insert was used to express IFN-$\gamma$ in E. coli under the control of lpp promoter employing pINIIIA3 vector. Cells harboring the recombinant plasmid ($pKS{\gamma}3B$) expressed high level of IFN-$\gamma$ assayed by antiviral activity, The molecule was also recognized by monoclonal antibody specific to a natural IFN-$\gamma$. An expression vector with high copy number (pHK-3) was constructed and was successfully used for the improvement of the expression of IFN-$\gamma$.

        Recently, the use of digital technology in architectural technology is gradually increasing with the development of various industrial technologies. There are artificial intelligence and drones in the field of architecture, and among them, deep learning technology has been introduced to conduct research in areas such as precise inspection of buildings, and it is expressed in a highly reliable way. When a building is deteriorated, various defects such as cracks in the surface and subsidence of the structure may occur. Since these cracks can represent serious structural damage in the future, the detection of cracks was conducted using artificial intelligence that can detect and identify surface defects by detecting cracks and aging of buildings.

        흰쥐에서의 알파 - 인터페론의 약물체내 속도론적 연구

        Pharmacokinetic behavior of recombinant human interferon alpha (rIFN- α A) was studied in the rat. Serum concentrations of rIFN- α A after intravenous administration could be explained by two - compartment open model. Pharmacokinetic parameters of rIFN- α A administered intravenously at two different doses (9.0 × 10^6 and 3.6 × 10^7 IU/Kg) were calculated by fitting the observed data to the nonlinear least square program. There were no significant differences in pharmacokinetic parameters such as systemic clearance (CLs), apparent distribution volume at steady state (Vd_(ss)), biological half life (t_½) etc. between the two doses. There were also no significant differences between pharmacokinetic parameters of this rIFN- α A and those of Namalva IFN- α which were reported previously by us. rIFN- α A aministered intraveneously had a small distribution volume, which indicates its poor distribution to peripheral or poorly-perfused organs. Renal excretion which might be a sum of filtration, secretion, reabsorption or catabolism in the kidney didn`t show any saturation phenomena in the serum concentration range studied (1 × 10³ - 5 × 10⁴ IU/㎖). Renal clearance accounted less than 0.1% of systemic clearance of rIFN- α A.

        딥러닝 기반 콘크리트 균열 검출 기술에 관한 연구

        When buildings deteriorate, they may develop defects like surface cracks and structural subsidence. If left unaddressed, these issues can significantly weaken the structure, potentially leading to collapse accidents. Detecting cracks promptly is crucial to prevent such outcomes. With the advancements in artificial intelligence, researchers are exploring deep learning techniques to identify microscopic cracks, replacing traditional manual methods. As AI technology progresses, diverse AI models have emerged, enhancing the reliability of crack detection data for field inspections. This study focuses on leveraging the Yolo model, known for its superior performance and faster data acquisition compared to other AI models. By incorporating object detection methods used by CNN, the study aims to enhance the detection performance of the model by considering various variables across different AI models and detection techniques.

        건설현장의 작업성 향상을 위한 기초적 연구 - 이동식 비계의 연구개발 동향 분석을 중심으로 -

        건설업에서 비계는 다른 공사와 더불어 공사장 안팎의 고소작업에 널리 사용된다. 이동식 비계는 조립과 해체가 쉽고 이동이 간편하지만, ‘전도’, ‘추락’ 등의 안전사고가 발생한다는 결점이 있다. 본 연구는 작업성을 향상시킨 이동식 비계의 개발을 위한 기초적 연구로써, 현재까지 보고된 비계 관련 연구 동향을 정리하고 작업성 관점에서 비계발판에 대한 분석을 실시하였다. 비계를 사용함에 있어 반복적인 작업을 줄이고 세부적인 설치를 간편화하게 된다면, 작업성을 증대시킬 수 있다. 숙련도와 관계없이 안전사고가 발생할 가능성이 있기 때문에 안전성을 중심으로 분석하였다. 그러나 작업성과 안전성을 향상시키기 위한 기술개발을 위해서는 높은 비용이 측정되며, 이를 보완하기 위한 비계 자재의 재사용 또는 재활용을 통한 비용감소가 필요할 것으로 판단된다. In the construction industry, scaffolding, along with other construction works, is widely used for high-rise work inside and outside the construction site. Mobile scaffolding is easy to assemble and dismantle and move, but it has the disadvantage of safety accidents such as “conducting” and “falling.” This study is a basic study for the development of mobile scaffolding that improved workability, and the research trends related to scaffolding reported so far were summarized and the analysis of scaffolding was conducted from the perspective of workability. If repetitive tasks are reduced and detailed installation is simplified in using scaffolding, workability can be increased. Safety was analyzed mainly because there is a possibility of safety accidents regardless of proficiency. However, high costs are measured to develop technologies to improve workability and safety, and it is judged that costs should be reduced through reuse or recycling of scaffolding materials to supplement them.

        사례분석을 활용한 시공단계 BIM 인력 투입 성과 분석

        BIM is effective to improve the labor productivity of construction participants . From this point of view, it is important to analyze the outcome related with BIM Labor which covers most of the BIM investment costs. This research focuses on BIM RFI which is one of the major task of the BIM labor and analyze the outcomes. In addition, this research was quantitatively analyzed by the standby time and related cost caused by BIM labor, which affect the results of the project participants. To this end, analytical standby queue model was utilized to analyze the labor focusing on micros TASK. 11 projects were selected to analyze the results of BIM labor and RFI that the project participants requested to the BIM labor was collected. Through this, it collected variables for analyzing results, and Finally, we pulled out 4 projects for analysis. In this study, the basic results analysis of RFI processing of the BIM labor, the probabilistic analysis of BIM labor service status, and the economic analysis of BIM labor optimal inputs were performed by using the research model presented. The results of this study can be utilized to formulate the optimal strategy for BIM labor inputs(e.g. number of employees, level, time point, etc.) of the construction phase. Moreover, it can contribute to ensuring the credibility of the BIM ROI results by presenting the cost of BIM services in BIM ROI analysis and the standby cost of project participants.

        The purpose of this study is to develop upcycling technology using waste plastic. Waste plastic has a great impact on the global environment. Recently, the use of plastic has increased due to the situation of Covid-19. As a result, ’upcycling’ technology is needed to recreate plastic as a high quality resource. In addition, the importance of maintenance of road facilities also raises and requires a repair solution to the pothole. It is going to conduct research on development of technology that solves repair materials for port holes while upcycling waste plastics. In this study, the possibility of using waste plastic as a porthole repair material was confirmed that molding was possible and curing was performed when high temperature melting was performed at 180℃.

        소규모 건축공사의 BIM 정보 활용을 위한 프로세스 제안 - 철골공사 중심으로 -

        The current study focused on the utilization of building information modeling (BIM) data in steel-frame structures, which help to reduce project durations because they employ prefabricated structural members that are assembled on-site. In addition, a business process model was proposed using BIM data collected during the preconstruction, structural steel fabrication, and on-site construction phases of an actual steel-frame project. The ultimate expectation is that BIM data support at each phase, as well as the increased understanding among project participants, will result in an increase in project management productivity. The results from the current study are summarized as follows: To implement a BIM capable of application to steel-frame projects and data utilization, existing theories were studied to develop the construction project steps, both generally into the preconstruction (A1), steel fabrication (A2), and on-site construction phases, (A3) and specifically into 19 BIM-applicable phases. Based on the derived BIM-applicable phases, the model elements of the BIM object were identified, and the shortcomings of existing steel-frame projects were ameliorated, resulting in an improved data flow model. Moreover, for the proposed BIM data flow to progress efficiently, the BIM specialist needs to be well-acquainted with the phase-specific three-dimensional (3D) model output, and the infrastructure to construct an error-free 3D model must be provided. Based on the actual construction example, the BIM data utilized steel-frame projects - via production reports, clash checks, two-dimensional (2D) drawings, four-dimensional (4D) simulations, and 3D scanning - to make cooperation and communication among participants easier.
