The occurrences of HBsAg(Hepatitis B Surface Antigen) from sera of Korean dental patients, paid blood donors and normal control population were investigated by means of modified counterimmunoelectrophoresis. And it was pointed out, by the high occurrence of it from dental patients, that Korean dentist and dental patients are exposed to the possible danger to be infected by hepatitis B virus. 1. Of 542 tested from control population, 28 (5.16%) were positive in HBsAg and antiHBsAg was not detected. Among 267 male and 275 female population, HBsAg positives were 16 (5.99%) in male and 12 (4.37%), in female respectively. 2. Of 4,277 tested from paid blood donors from 1973 to 1975, 259 (6.05%) were positive in HBsAg, whereas antiHBsAg was not detected. 3. The investigation of HBsAg occurrence from dental patients resulted in that, of 95 tested, 9 (9.47%) were positive. but no antiHBsAg was detectable. Among 51 male and 44 female patients tested HBsAg-positives were 5 (9.8%) in male and 4 (9.47%), in female respectively.
A MELANOCYTE-SPECIFIC cDNA WHOSE EXPRESSION IS INDUCIBLE BY α-MSH AND IBMX

Normal human melanocyte의 λgtll cDNA library를 tyrosinase항체를 이용하여 검 색, 16 cDNA clones을 분리하였다. 이중 albino locus혹은 그인접부위에 존재하는 13clones는 tyrosinase gene으로 여겨지며 나머지 3clones는 상호간 상동성(homology)을 보이고 있으나 tyrosinase gene과 는 상이하였다. 이들 3clones의 대표적인 Pmel 17 - 1에 상응하는 mRNA(cRNA)는 사람과 mouse melanocyte에서만 특이하게 표현되며 크기는 약 2.4kb, 표현도는 melanin양과 비례 하였다. 여러종류의 melanoma cell을 αMSH 또는 IBMX로 자극시 Pmel 17 - 1의 cRNA표현은 증가되었으며 단일항체를 이용한 면역학적 실험으로 Pmel 17 - 1은 tyrosinase와 면역학적 동질성을 갖는 75KD glycoprotein을 합성함이 판명되었다. 또한 genomic DNA blot실험결과 Pmel 17 - 1 cDNA는 mouse albino locus 혹은 인접부위에 위치하지는 않으나 mouse와 사람에 single gene으로 존재하며 evolutionarily conserved gene임을 볼 수 있어 중요한 기능을 가지고 있을 것으로 사료된다.
항원수용체자극에 의한 Cytolytic T cell 특이전사체 표현유도

Cytolytic T cell(CTL)에서는 표현되나 다른 세포에서는 표현되지 않는 유전자를 검색하여 최근 저자는 3종의 CTL 특이 cDNA를 cloning하였다. CTL 특이 cDNA의 기능을 규명하기 위하여 CTL L3 cell을 항원수용체를 통하여 혹은 interleukin 2(IL-2)로 자극하여 활성화시킨 후 RNA blot analysis로 각 cDNA clone의 상응전사체 표현양상을 관찰, CTL활성화과정과의 연관성을 실험하였다. 이중 2종의 cDNA상응전사체표현은 항원수용체자극에 의해 현저히 증가된 반면 IL-2는 전혀 영향을 미치지 않았으며 이 같은 전사체표현증가는 cyclosporin A 처리로 완전히 억제되었다. 이상의 결과는 항원수용체자극으로 활성화되는 유전자가 IL-2에 의해 활성화되는 유전자와는 상이함을 보여주는 것이며 또한 2종의 cDNA clone이 IL-2에 의해 활성화되지 않으나 항원수용체를 통하여 중개되며 cyclosporin A에 예민하게 반응하는 CTL활성화과정의 특정경로에 관여하는 것으로 사료된다. Employing the approach to isolate the genes expressed preferentially in cytolytic T cell (CTL) but not in other types of cell, 3 CTL-specific cDNAs were recently cloned. To characterize these cDNA clones in relation to CTL activation, their expression pattern after T cell antigen receptor (TCR) or interleukin 2 (IL-2) stimulation were investigated by RNA blot analysis of cloned CTL L3 cells. Transcripts level of two cDNA clones were markedly elevated by TCR stimulation but not by IL-2. In addition, transcripts expression of both clones were abrogated by cyclosporin A treatment. These results indicated that gene activation mediated by TCR is distinct from that mediated by IL-2 and imply that those two unidentified cDNA clones are related to TCR-mediated, IL-2-independent but cyclosporin A-sensitive pathway for CTL activation.

대장균에서 인간의 면역 인터페론을 tac 프로모터를 이용하여 고도로 발현시키는 연구

The cDNA for the structural sequence of mature human mmune-interferon (IFN-γ) was introduced into E. coli high expression vector, pKK223-3,constructed by using tac promoter and rrnBT1T2 transcription terminator region and transformed to host, E. coli JM105, which has lacIq repressor gene on chromosomal DNA, E. coli MM294, and E. coli VMC9, which have laI repressor gene. The resultant ampicillin resistant transformants were analyzed by the immunoblotting and electron microscope. IFN-γ, which cross-reacted with mouse anti human IFN-γ serum, was produced as the soluble and the insoluble inclusion form. Their productions were efficiently regulated in E. coli JM105, but not in E. coli MM294, and E. coli YMC9 by using the IPTG.
