        Immune-triggering effect of the foodborne parasite Kudoa septempunctata through the C-type lectin Mincle in HT29 cells

        Kudoa septempunctata is a myxozoan parasite that causes food poisoning in individuals consuming olive flounder. The present study aimed to investigate the currently insufficiently elucidated early molecular mechanisms of inflammatory responses in the intestine owing to parasite ingestion. After Kudoa spores were isolated from olive flounder, HT29 cells were exposed to spores identified to be alive using SYTO-9 and propidium iodide staining or to antigens of Kudoa spores (KsAg). IL-1, IL-8, TNF- and NFKB1 expression and NF-B activation were assessed using real-time PCR, cytokine array and western blotting. The immunofluorescence of FITC-conjugated lectins, results of ligand binding assays using Mincle-Fc and IgG-Fc, CLEC4E expressions in response to KsAg stimulation, and Mincle-dependent NF-B activation were assessed to clarify the early immunetriggering mechanism. Inflammatory cytokines (IL-1, GM-CSF and TNF-), chemokines (IL-8, CCL2, CCL5 and CXCL1) and NF-B activation (pNF-B/NF-B) in HT29 cells increased following stimulation by KsAg. The immunofluorescence results of spores and lectins (concanavalin A and wheat germ agglutinin) suggested the importance of Mincle in molecular recognition between Kudoa spores and intestinal cells. Practically, data for Mincle-Fc and KsAg binding affinity, CLEC4E mRNA expression, Mincle immunofluorescence staining and hMincledependent NF-B activation demonstrated the involvement of Mincle in the early immune-triggering mechanism. The present study newly elucidated that the molecular recognition and immune- triggering mechanism of K. septempunctata are associated with Mincle on human intestinal epithelial cells.

        Malignant esophagorespiratory fistula is a devastating and life-threatening complication of esophageal and bronchogenic carcinomas. As a non-surgical treatment, peroral stent placement into the esophagus or airway can close-off the fistula and prevent progression of the pneumonia. Although reopening of the fistula is not uncommon despite stent placement, interventional treatment is effective for sealing off reopened ERFs. Bronchopleural fistula is a well-recognized complication of pneumonectomy. There have been several reports to occlude the fistula with use of stents and much more experience is required.

        SWOT 분석을 통한 환경보건 연구의 동향과 전망 고찰

        Background: Research in environmental health (EH) is of crucial strategic importance for contemporary society. It is becoming even more critical in light of the increasingly rapid pace of environmental changes, opportunities, and threats. Objectives: This study aimed to identify trends and the prospective of environmental health research using SWOT analysis. Methods: The trends in environmental health research were reviewed in previous studies and reports. Reviewed manuscripts were searched for using the keywords of 'environmental health' and 'environmental hygiene' in the KCI (Korean Journal of Citation Index), KISS (Korean Academic Information), PubMed, and Google Scholar. Results: It is essential to center the EH research agenda around key priorities focusing on technological innovation, job creation, and the increasingly prominent role of the private sector. Given the rapidly evolving global sustainability agenda, greater clarity on the ever-increasing sources of complexity and growing expectations of the public might be needed. This requires the identification of criteria to identify EH research priorities with the ultimate goal of maximizing societal benefit. Public health relevance, such as extent and severity of health impact, level of exposure, and inequalities of effects, could be included. Conclusions: Considering the recent interest in and importance of environmental health, a comprehensive approach to environmental health research should be required through the application of the latest science and technology, citizen participation, and environmental health surveillance systems.

        자연경관에 도입되는 건축물의 경관민감도 측정에 관한 연구

        As high-rise buildings came in, the landscape of rural areas and natural landscapes often got damaged. Therefore, this study aims to prevent this, grasp the extent of the influence of the surrounding landscape, to grasp the range of height that can be permitted and present the direction of landscape management of agriculture and natural landscape. This study tried to grasp the range of height by using price sensitivity analysis method for two apartment and apartment complex which entered DangJin city and SeoSan city. First, in the case of a two apartment, the range of the height allowable section was from the 6th floor to the 11th floor in close range view, and it was a section from the 7th floor to the 12th floor in medium range view. In the case of the apartment complex, the range of the height allowable range was from the 10th floor to the 17th floor in close range view, the 9th floor to the 16th floor in medium range view. The stress index was found to be positive in a two apartment in close range view, and in the apartment complex case. therefore it was better to set it to a lower in the Range of Acceptable Height(RAH). Second, it showed no difference in the sensitivity of landscape to gender. Thirdly, the results of the landscape sensitivity analysis of major and non-major showed the difference in the medium range view picture of the apartment complex. Majors are lower than the point of minimum height(PMinH) than non-Majors. In the case of major, the stress index was 1.4. it turned out that it was better to make a decision closer to point of minimum height (PMinH). In the case of non-major, the stress index was -1.3. it was also able to accept decision close to the point of maximum height (PMaxH). Since the results of the above research gave changes only in the variable of the height of the landscape, we can not grasp the point of interaction with other variables, and future research is considered necessary.

        조리방법을 달리한 쥐눈이콩의 항산화력 및 이소플라본 배당체·비배당체 함량 비교

        Purpose: In this study, small black beans (Rhynchosia nulubilis) that were produced in the Jungsun province of South Korea were selected for use in various cooking recipes because they are known to contain higher isoflavones and excellent antioxidant effects, as compared to any other domestic soybeans. Methods: Physicochemical and antioxidant characteristics of small black beans were analyzed with uncooked beans and four other cooking methods of pan broiling, boiling, steaming, and pressure cooking. Results: Contents of ${\beta}$-glucosides (daidzin, glycitin, and genistin) and aglycone (daidzein, glycitein, and genistein) in small black beans were significantly different depending on the cooking methods (p<0.001). The results of the experiment indicated that the amount of total polyphenol in pressure cooked beans was highest, showing a value of 7.16 mg/g (p<0.001). Most contents of isoflavones (${\beta}$-glucoside, aglycone) in uncooked beans appeared to increase after cooking. In particular, the amount of glycitein was highest in pan broiled beans ($9.63{\mu}g/g$). The total isoflavone content differed among the different cooking methods, ranging from highest to lowest in the following order : pan broiled beans ($759.49{\mu}g/g$), pressure cooked beans ($725.12{\mu}g/g$), boiled beans ($591.05{\mu}g/g$), steamed beans ($511.61{\mu}g/g$), and uncooked beans ($180.80{\mu}g/g$) (p<0.001). Conclusion: Especially, the amount of isoflavones increased significantly in pan broiled beans and pressure cooked beans, suggestive of optimized cooking methods for increasing nutritional and functional contents in cooked food.

        GPR 분석을 위한 자갈궤도 자갈의 감쇄특성을 이용한 이득함수 개발

        자갈궤도는 100여년 이상 전세계적으로 널리 사용되는 궤도구조로 저렴한 초기 건설비용, 유연한 유지보수와 진동 및 소음의 저감효과가 있지만, 열차운행에 따라서 지속적인 침하가 발생하는 단점이 있다. 이로 인해 지속적인 유지보수가 필요하며, 최근 열차의 속도, 용량, 중량의 증가로 유지보수비용이 증가하고 있는 실정이다. 파울링(fouling)은 노반 세립분이나 자갈입자가 파쇄되면서 발생한 세립분이 자갈 사이의 공극을 메우는 현상으로 배수가 나빠지고, 자갈궤도의 침하를 가속화시키는 등 자갈도상의 열화의 주원인으로 지목되고 있다. 본 논문에서는 GPR(Ground Penetrating Radar)을 이용하여, 도상의 상태를 평가 할 수 있는 분석방법을 제안하였다. 경험에 의존하는 기존의 분석기법을 대신하여, 고속선에 사용되는 자갈의 감쇠 특성을 반영한 이득함수(gain function)를 제안하였다. 실내 박스 실험과 실모형 실험을 통해 제안한 이득함수로 증폭한 GPR신호에 힐버트 변환을 추가로 적용하여 자갈층과 노반층에서 반사된 신호의 세기변화를 깊이에 따라 계산하였다. 이로부터 신호의 세기가 비교적 크게 변화하는 곳을 파울링층과 노반층의 경계로 구분할 수 있었다. 다만 현장적용을 위해서는 침목의 영향을 분리하고 전자기파 분산특성 등에 대한 추가 연구가 필요할 것으로 사료된다. Ballasted track has been used as track system for more than 100 years. Ballasted track has advantages of low construction cost, flexible maintenance, low noise and vibration, and so on. However, ballasted track leads to continuous settlement which causes maintenance. Recently, increase in speed, traffic volume, and weight of train requires more frequent maintenance. Fouling, well-known phenomenon of accumulation of fine materials due to intrusion of subgrade and breakage of ballast materials, expedites the settlement (i.e., irregular settlement) of track. Ground Penetrating Radar (GPR) can be one of non-destructive tools that can evaluate fouling level of ballast. In this paper, a gain function based on the attenuation characteristics of ballast material is suggested in conjunction with Hilbert transform. Lab box tests and full-scale tests indicate that the suggested method reasonably classifies clean, fouled layers, and subgrade. However, additional study to eliminate effect of sleeper and to include the scattering features of the electromagnetic wave in ballast voids should be required in order to enhance the accuracy.

      • Alloy를 이용한 CC3.1 기반의 EAL7등급 FSP 정형 명세 및 분석

        현재의 시스템 개발은 기능적인 사항들뿐만 아니라 보안적인 사항에 대해서도 상당한 이슈로 떠오르고 있다. 이에 국제표준으로 IT 보안평가결과를 상호 인정할 수 있는 수단인 CC(Common Criteria, 공통평가기준)를 개발하였으며 이 기준에 따라 시스템의 평가인증이 이루어지고 있다. CC3.1기반의 EAL7등급에서는 ADV클래스에서 정형적인 방법으로 기능을 명세한 FSP(Function SPecification)가 요구된다. 따라서 본 연구에서는 Relational Logic기반의 정형 명세언어인 Alloy를 이용하여 KCOS(K-Debit Card Operating System)의 FSP를 명세한 후 일관성을 검증하고 명세한 제약사항들의 유효함과 오퍼레이션들이 올바로 수행함을 보였다.
