Piperine에 의한 위암세포 AGS 증식 억제와 Apoptosis 유도

본 연구는 후추과 식물의 주성분인 piperine이 위암세포 AGS의 항암 효과에 미치는 영향을 확인하기 위해 수행되었다. Piperine에 의한 위암세포 AGS의 생존율을 확인하기 위해 MTT assay를 수행한 결과 농도 의존적으로 암세포의 생존율이 감소하는 것을 확인하였다. 이러한 암세포의 생존율 억제가 apoptosis에 의한 효과인지를 확인하기 위해 DAPI staining을 실시한 결과 piperine을 처치한 군에서 apoptotic body와 염색질 응축이 증가하는 것을 확인하였다. MTT assay와 DAPI staining의 결과를 바탕으로 piperine이 위암세포 AGS에서 apoptosis와 관련한 단백질 발현 양상에 미치는 영향을 확인하기 위해 western blotting을 실시하였다. 그 결과 piperine에 의해 AGS 세포에서 apoptosis를 유도하는 p53, Bax의 발현은 농도 의존적으로 증가하였고, apoptosis를 억제하는 Bcl-2, XIAP의 발현은 농도의존적으로 감소하였다. Apoptosis의 궁극적 단계인 caspase-9와 손상된 DNA를 복구하는 PARP의 분절은 증가하였으며, apoptosis를 방해하는 인자인 Akt의 인산화는 농도의존적으로 감소하였다. Akt 억제제인 LY294002와 piperine을 병행 또는 단독으로 처치하여 western blotting을 진행한 결과 Bcl-2의 발현은 piperine을 단독으로 처치했을 때와 유사한 감소 경향을 보였지만, p-Akt의 발현은 대조군과 비교해 LY294002와 piperine을 단독으로 처치했을 때보다 병행했을 때 더욱 감소하였다. 이와 반대로 Bax와 cleaved-PARP의 발현은 강하게 증가하였다. 본 연구의 결과를 종합해볼 때 piperine이 위암세포 AGS에서 Akt 경로를 통해 apoptosis를 유도하는 것으로 여겨지며, 향후 위암예방제나 치료제로의 개발 가능성이 있을 것으로 생각한다. Piperine [(E,E)-5-(3,4-methtylenedioxyphenyl)-2,4-pentadienolypiperidide] is a principal of Piperaceae, including Piper nigrum L. and Piper longum Linn., which has been used as a spice and traditional medicine. In this study, we investigated whether or not piperine has anti-cancer effects on AGS human gastric cancer cells. The results demonstrated that piperine not only inhibited proliferation using MTT assay but also induced apoptotic bodies using DAPI assay in a dose-dependent manner in response to piperine. Expression levels of p53, Bax (pro-apoptotic), cleaved caspase-9, and cleaved-PARP increased, whereas expression levels of Bcl-2, XIAP (anti-apoptotic), and Akt decreased in a dose-dependent manner compared with the control by western blotting analysis. To identify the connection between phospo-Akt and Bcl-2 family in response to piperine, LY249002 (Akt inhibitor) was treated with piperine (150 μM). The results were shown that expression of phospo-Akt was reduced whereas expression of Bax and cleaved-PARP increased in a dose-dependent manner. These results indicate that piperine induced apoptosis in AGS cells and may serve as a chemopreventive or therapeutic agent for human gastric cancer.

최근에 방사선 조사에 대해 방호효과를 가지는 것으로 알려진 보중익기탕의 골수세포 분화 유도 효과에 대해 관찰하였다. 시험관에서 골수세포를 배양했을 때, 배양시간(5일, 10일)에 따라 대조군의 세포 수는 현저히 감소하였고, 보중익기탕의 total 분획을 첨가하였을 때도 비슷한 결과를 얻었다. 그러나 polysaccaride 분획을 첨가하였을 때는 감소하는 세포수가 일정한 수준에서 유지되며 더 이상 감소하지 않았다. 그리고 이들 세포가 어떤 종류의 세포인지를 유세포분석기로 분석한 결과, Pre-B세포의 특징적인 세포 표면 단백질인 CD19와 CD40을 동시에 발현한 세포인 것이 확인되었다. 또한, 이들 세포는 분화과정이 끝난 B세포가 분비하는 IgM 뿐만 아니라 IgG1, G2a, G3를 분비하였다. 이상의 결과 보중익기탕의 polysaccaride 분획에 골수세포가 B세포로 분화 증식하는 것을 유도하는 성분이 포함되어 있어 방사선 조사로 상해를 입은 조혈계에 대한 방호 효과를 나타내는 것으로 생각된다. This study was designed to evaluate the effect of Bu-Zhong-Yi-Qi-Tang extracts, a prescription of traditional oriental medicine, on development of the B cells. In the bone marrow cell cultures, progenitors viability, expressions of particular cell-surface proteins and production of immunoglobulins were investigated in the presence of Bu-Zhong-Yi-Qi-Tang extracts. The administration of Bu-Zhong-Yi-Qi-Tang polysaccharide fraction increased the viable cell numbers of the precursor B cells, and elevated expression levels of CD19/CD40 specific for pre-B cells after 10 days culture were demonstrated by flow cytometry analysis. The production of immunoglobulin M in the presence of polysaccharide fraction increased progressively in the culture supernatant, and preferentially induced class switching to IgG1, IgG2a and IgG3. These results indicated that Bu-Zhong-Yi-Qi-Tang strongly correlated with the development of precursor B cells in the bone marrow cell culture. Therefore the polysaccharide fraction of Bu-Zhong-Yi-Qi-Tang might be a useful radioprotector, especially since it is a relatively non-toxic natural product. Further studies are needed to better characterize the protective nature of Bu-Zhong-Yi-Qi-Tang extract.

Estimating the physical properties of the survey area and mapping the geotechnical basement play an important role in ocean engineering and construction field. In this study, we performed marine seismic reflection and refraction survey as an engineering application at shallow marine. We made use of the dual boomer-single channel streamer as a source-receiver in reflection seismic survey and air-gun source - the manufactured OBC(Ocean Bottom Cable)-type streamer in refraction survey. In the seismic reflection data, we could easily find the geological layers and basement. Moreover, seismic refraction data could present sediment thickness and velocity distribution.

Large scaled transportation complex(LSTC) are defined as certain boundary of areas centering large transit station houses with many floating pedestrians and diverse activated land uses. Major LSTC areas located in Seoul are faced with the problems of geographical walking disconnections and difficulty environment. In order to improve walkability for these LSTC areas, it is quite required moment to discuss on how to design and allocate walking facilities and infrastructures of LSTC, which have indispensible relationship with the transit oriented development (TOD) as well. Total number of LATC located in Seoul in this study are eleven. Among these, six are currently being operated and five planned in future. Six LATCs include Seoul, YongSan, Sindorim, Wangsimni, Cheongnyangni rail station LATCs, and one Express Bus Terminal LATC respectively. And five planned LATCs are Suseo-KTX, Samsung, Sadang, Jamsil, Susaek rail station LATCs respectively. The comparison analysis of Seoul rail station LATC and foreign rail station cases identifies that even though we do not have appropriate basic walking infrastructure dependable to alleviate the degree of walking inconvenience, foreign nations have adopted the concourse, that is a type of artificial ground, as major walking infrastructure for that purpose. This study aims at the development of strategies for efficient walking infrastructure construction and the evaluation of efficiency of walking facilities system. For these, this study includes four following research sub purposes. 1) development of walkability analysis model, 2) evaluation of alternative LATC scenarios based on toy pedestrian network, 3) evaluation of Seoul rail station LATC based on new alternative scenarios, 4) suggestion of implication and institutional realization methodologies. The simulation results implemented based on three alternative scenarios to an imaginary LATC area show that the best strategy for improving workability is to combine vertical and horizontal walking connection facilities with the main and sub concourses. Also in Seoul station LATC simulation case, it is also identified that the best alternative is to efficiently integrate vertical walking connection facilities such as escalator, elevator, stair etc, and horizontal facilities such as pedestrian exclusive deck, direct building bridge etc with the same concourse concept. However, country to the evaluation results, it is very difficult to create pedestrian friendly walking environment for LATCs under the current urban planning and development related law and institution. In order to overcome these problems, first of all, the walking infrastructure needs to be systematically reorganized as the key means of urban renewal. Within that system, new urban planning and development related laws and institutions differentiated from the existing are required to be introduced for the purpose of integrated design and construction of LSTC.
