Leukocyte adherence inhibition test를 이용한 Mycobacterium 속균(屬菌) 감작(感作)기니픽의 세포면역반응(細胞免疫反應)의 특이성(特異性)

In order to measure in vitro cell mediated immunity in the guinea pigs sensitized with the killed bacilli of Mycobacterium bovis ($AN_5$), M avium (serotype 2), M tuberculosis and M intracellulare (serotype 8), leukocyte adherence inhibition (LAI) test was established using the antigens of purified protein derivatives (PPD) tuberculin. By using LAI test, specificity of cell-mediated immune responses of the guinea pigs inoculated with various Mycobacterium spp was investigated, and comparison between values of LAI and skin test was also made to evaluate the specificity of the newly designed test. The results obtained throughout the experiments were summarized as follows; 1. The optimal concentration of PPD antigens for LAI test was 1 to 2mg per ml of medium. 2. When the leukocytes of guinea pigs sensitized with both M bovis($AN_5$) and M avium (serotype 2) for 2 to 8 weeks were incubated with homologous or heterologous PPD antigens, mean values of LAI test were $61.2{\pm}11.2$ and $65.6{\pm}5.1%$ in homologous PPD antigens respectively, while $30.0{\pm}3.7$ and $32.8{\pm}5.7%$ in heteNlogous PPD antigens, showing the prominently high value of LAI in the homologous syst,em (p<0.01). 3. When the leukocytes of guinea pigs sensitized with both M tuberculosis and M intracellulare (serotype 8) for 2 to 8 weeks were incubated with homologous and heterologous PPD antigens, mean values of LAI test were $67.9{\pm}2.9$ and $66.9{\pm}5.0%$ in homologous PPD antigens, while $27.4{\pm}7.4$ and $24.4{\pm}7.1%$ in heterologous PPD antigens, showing the prominently high value of LAI in the homologous system (p<0.01). 4. Comparing with the specificity of LAI and skin tests on the basis of the value obtained from the homologous system, deviation of reaction was revealed to be 49.5 to 100.2 in LAI test, and -15.9 to 52.0 in skin test.

To establish a laboratory animal model for study on development of diagnostic methods for infectious bovine rhinotracheitis virus(IBRV), experimental infection of the virus to rabbits immunosuppressed with dexamethasone(DX) were carried out. Results obtained throughout the experiments were as follows. When lymphocyte activity was measured by lymphocyte transformation to phytohaemagglutinin in parallel with total and differential leucocyte counting, both groups treated with 2.0mg DX once and 1.0mg DX daily showed marked immunosuppression between 5 to 72 hrs. after administration. The degree of suppression of lymphocyte activities was more remarkable in the latter group. IBRV PQ7 strain at $10^{7.5}\;TCID_{50}/0.2ml$ was inoculated into conjunctival sacs of rabbits immunosuppressed with DX and non-treated. During 3 weeks observation, the immunosuppressed groups revealed mild conjunctivitis, viremia and virus recovery by 33.3 to 100%, whereas the DX nontreated group showed viremia and virus recovery with no clinical conjunctivitis by one of three rabbits(33.3%). In conclusion, it was indicated that experimental infection of IBRV PQ7 strain to rabbit was limited in prerequisite to immunologic modification by administration of immunosuppressive drugs.
설사증 이환견(罹患犬)으로 부터 분리한 canine parvovirus의 성상에 관한 연구

From 1988 to 1989, 8 strains of canine parvovirus-2(CPV-2) were isolated from the fecal specimens from the dogs that were clinically diagnosed as canine parvoviral enteritis in the veterinary hospitals located in the regions of Taejeon and Chungbuk province. The biological and physicochemical properties for the isolates were studied. Among 62 fecal samples collected from the dogs with enteric diseases, 24(38.7%) showed the haemagglutinating activity to porcine erythrocyte ranging from 16 to 16,384 of HA titers. In cytopathological studies with CRFK cells, intranuclear inclusion bodies were observed in all of eight specimens with the high HA titer over 1,000, of which three specimens showed cytoplasmic inclusions concurrently with the intranuclear inclusion bodies. It was found that the isolates revealed the highest haemagglutinating activity with porcine erythrocytes and the relatively lower haemagglutination titers with the erythrocytes from cat and rabbit. None of erythrocytes from the other animals reacted with the isolates. By the cross-haemagglutination inhibition test for the isolates with the reference viruses and sera, the isolates were evidently identified as the strains of CPV-2. In physicochemical property test, the isolates were stable in lipid solvent, pH and heat treatment at $56^{\circ}C$ for 30 min, and showed the virus particle size less than 25 nm, containing a DNA genome.

To investigate the pathogenicity and virological properties of TJ-89-2 strain of canine parvovirus(CPV -2) that was isolated from the diseased puppies in Korea, seven puppies-were inoculated intraorally with the isolate at the HA titer of 8,192. All of the puppies showed the signs of anorexia, vomiting, diarrhea and died at the 5th to 10th days post inoculation (pi). All of the fecal samples from the puppies revealed significantly high HA titers afterward the 5th days pi. Body temperature and the number of total leucocytes were slightly increased at the early stage of infection, but extremly decreased at the stage of collapse. HI titers of the sera began to increase at the 3rd to 4th days pi, reaching 512 to 1,024 at the 4th to 5th days pi.

In order to establish an in vitro method to measure cell-mediated immunity, that is leukocyte adherence assay, in guinea pig system, various factors affecting the adherence of guinea pig leukocyte on the glass hemocytometer surface were studied. Among anticoagulants heparin at concentration of 10 iu/㎖ showed the highest leukocyte adherence of 67.4±6.3%, compared with those of sodium citrate and ammonium oxalate. To remove erythrocyte in heparinized blood, 0.15 M NH₄C1 treatment was proved to be more efficient than distilled water. It was found that 10% of fetal calf serum supplemented in medium revealed the highest adherence capability of 73.4±12.1 % and 60 min. was the most adequate culture period in the cell concentration of 1.5×10 exp(5), 1.5×10 exp(6), and 1.5×10 exp(7) cells/㎖. The leukocyte attached to the glass surface were differentiated into polymorphs by 69.0±8.2%, lymphocytes by 27.6±4.6% and monocyte by 3.4±0.5%.
國內에서 分離한 Aujeszky's Disease Virus의 家兎에 대한 病原性

To evaluate pathogenicity in rabbits for Aujeszky's disease virus, NYJ-1 strain, isolated from pigs in Korea, the virus at 10^5.7 TCID_50/0.2㎖ was inoculated subcutaneously to rabbits, and virological and histopathological studies were carried out. All of the rabbits infected with 0.25㎖, 0.5㎖, l.0㎖ and 2.0㎖ of virus at 10^5.7TCID_50/0.2㎖ were died at 36 to 72 hours post-infection, manifesting the clinical signs of depression, frensy, scratching, biting and pruritus. The virus was recovered from the tissues of tonsil, spleen, brain, lung, liver, kidney and blood of the succumbed rabbits, showing the highest virus titers of 10^3.5 to 10^5.6TCID_50/0.2㎖ in tonsil and comparatively lower virus titers of 10^2.4 to 10^3.5TCID_50/0.2㎖ in brain. At autopsy no specific lesions were observed in parenchymal organs. By histopathological studies. congestion, hemorrhage and necrosis were recognized in the tissues of tonsil, liver, spleen, lung, kidney, adrenal gland and jejunum. Intranuclear inclusion body was observed in the epithelial cells of tonsil and jejunum.
