Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.

Agrobacterium rhizogenes, a gram-negative soil bacterium, is one of the most widely studied among them. A, rhizogenes can transfer T-DNA, excised from Ri (root inducing)-plasmids from the bacterial to the plant cell. It is the causal agent of ‘hairy root’ diseases in plants, and has been used for the production of hairy root cultures from a multitude of species. Five different strains of Agrobacterium rhizogenes differed in their ability to induce Scutellaria baicalensis hairy roots and also showed varying effects on the growth in hairy root cultures. A. rhizogenes R 1000 is the most effective strain for the induction (57.3%) and growth (11.9 g ℓ?¹) in hairy root of Scutellaria baicalensis. Our results demonstrate that use of suitable strains of A. rhizogenes may allow study of the regulation of flavone biosynthesis in hairy root cultures of Scutellaria baicalensis.

Rice, one of the most important Cereal crop, is the staple food for more than half the world`s population. Future crop improvement needs the genetic variation from traditional varieties and related wild species to cope with the many biotic and abiotic stresses around the world. Established in 1977, the IRRI genebank has conserved more than 109,000 accessions including 5,000 accessions of wild. All introduced germplasms are multiplied after the necessary quarantine check by IRRI`s Seed Health Unit. Multiplication and regeneration are carried out only during the dry season between the end of October and March with the consideration of ecotype to produce seeds with the highest quality. A sufficient quantity of high qualified seeds are stored in base and active collections for long-term and mediumterm conservation, respectively. Germplasms are also conserved at the NCGRP(National Center for Gegenetic resources Preservation), USA and SGSV(Svalbard Global Seed Vault), Norway as a safety back-up. The decision to regenerate germplasm accessions depends equally upon the viability and quantity of seeds held in storages. Germplasms are available freely on request to bona fide researchers in both public and private sector institutions and farmers. An integrated information system is linked to all operations associated with germplsam conservation and management.

Farnesyl diphosphate synthase (FPS) catalyzes the biosynthesis of farnesyl diphosphate, a precursor for many important terpenoid products. A cDNA encoding FPS was first isolated from Allium sativum (AsFPS) using rapid amplification of cDNA ends (RACE) PCR. The sequence of AsFPS contains an open reading frame encoding a protein of 341 amino acids with a predicted molecular mass of 39.61 kDa. Alignment of AsFPS deduced amino acid revealed high identities with other plants ranging from 79% to 85% and showed 2-high conserved aspartate-rich motifs known to be important for FPS activity. Furthermore, AsFPS expression was stronger in the green organs such as bulbils, scapes, leaves, stems, but weaker in bulbs and roots than on-green organs of A. sativum.
