        바이오플락과 아쿠아포닉스를 이용한 도심형 양식시스템에서의 메기양식

        This study was conducted to determine whether the water in which nitrate accumulated during long-term fish culture in an aquaponics system without water exchange could be removed and reused as catfish-culturing water. The catfish (Silurus asotus) were cultured in the urban aquaculture system using BFT (Biofloc Technology) aquaculture and an aquaponics system (two rearing tanks, 3 tons each) without exchanging the rearing water. After 151 days (from March to August) of rearing, 2.8 g of fry had grown to an average weight of 171.3 g (total weight, 56.53 kg) and 235.5 g (total weight 71.1 kg), respectively. The overall survival rate was 65% in the urban aquaculture system. However, the survival rate was 77.7% before separation into the two tanks. The survival rates after the separation were 92.9% and 78.0%. In the early biofloc watermaking process, there was a high mortality rate. After water stabilization, the mortality rate decreased and some mortality occurred during the period when the total amount of suspended solids (TSS) increased. The results of monthly blood analysis of the catfish showed that the AST concentration was significantly higher in April. Blood ALT levels and triglycerides showed no difference in the rearing period and the glucose, cholesterol, and total protein levels were significantly higher in July. There was no difference in the other periods. The plants produced by the aquaponics system using catfish-rearing water were lettuce, basil, chard, and red chicory. These showed smooth growth and a total of 148.85 kg of plants were harvested in five months. It was possible to remove nitric acid from the aquaponics system and reuse it as catfish-rearing water. Maintaining proper plant quantity according to the capacity of the catfish showed that the combination of agricultural and aquatic products was possible. 도심형 양식시스템을 바이오플락 양식기술과 아쿠아포닉스 배양대로 구성하여 사육수를 교체하지 않고 메기를 (사육조 3.3톤 2개) 양성한 결과 151일의 사육 후 2.8 g의 종묘가 평균 무게 171.3 g (총중량 56.53 kg)과 235.5 g (총 중량 71.1 kg)로 성장하였다. 입식에서 수확까지의 누적 생존율은 65% 보였고, 성장 구간별로 입식에서 1차 성장 후 분조 이전까지 77.7%, 분조 이후 생존율은 수조에 따라 차이를 보여 각각 92.9%와 78.0%로 나타났다. 초기 바이오플락 사육수가 만들어지는 과정에서 일부 폐사가 발생하였고, 수질이 안정된 이후에는 폐사가 감소하였다. 메기의 혈액분석결과 사육초기 BFT 사육수가 안정화 이전인 4월에 간 손상 지표인 AST의 농도가 유의적으로 높은 값을 보였으며 ALT, triglyceride는 전 사육기간 내에 차이가 없었다. Glucose, cholesterol, total protein은 7월에 유의적으로 높은 값을 보이고 다른 기간에는 차이가 없었다. 메기 사육수를 이용한 아쿠아포닉스 가동 시 생산된 식물은 상추, 바질, 적근대, 적치커리 등이 원활한 성장을 보여 5개월간 총 148.85 kg의 식물을 수확하였다. 또한 아쿠아포닉스 시스템에서 식물재배에 따른 사육수 내의 질산 제거능력과, 질산이 제거된 사육수는 메기 사육수로 재사용이 가능한 것으로 확인되었다. 결론적으로, 본 연구에서는 도심형 양식시스템으로 물을 교환하지 않고 어류를 양식할 때 사육수에 축적된 질산을 제거하고 재사용이 가능한가를 아쿠아포닉스 기술을 결합하여 연구하였으며, 양식생물(메기) 수용량에 따른 적정 식물량을 유지하면 농수산 복합양식이 가능하다는 것을 보여주었다.

        소라, Batillus cornutus (Lightfoot, 1786) 혈구의 종류 및 미세구조적 특징

        Light and transmission electron microscopy of Batillus cornutus hemocytes revealed differences that the morphological distinctions between blast-like cell, granulocytes and hyalinocytes. Base on the morphological characteristics of the cells, we identified the eight types of hemocytes and present a categorization of the hyalinocytes into six sub-categories. The hemocytes of B. cornutus were observed basophilic cell under the light microscopy. Blast-like cells had a spherical profile with a central nucleus filling almost the whole cell. Granulocytes were characterized by presenting variable numbers of granules. This cell had spherical shape with diameter 7 μm and smooth endoplasmic reticula, granules, mitochondria, glycogen granules in the cytoplasm. Hyalinocytes were the most abundant cell type. Especially, hyalinocyte VI had iirregular an amoebal shape and observed autophagosome and heterophagosome in the cytoplasm. From these results, it is concluded that there are eight types of cells in the hemolymph of B. cornutus. Further studies are now needed to identify the role of these hemocytes in the enzymological and immunological response. 전라남도 완도군 인근연안에서 채집된 소라, Batillus cornutus의 혈구를 광학현미경과 전자현미경을 이용하여 혈구의 종류 및 미세구조적인 특징을 연구하였다. 광학현미경 상에서 소라의 혈구들은 호염기성 세포로 관찰되었다. 혈구의 형태학적인 특징으로 분류를 하였을 때 소라의 혈구는 모두 8종류로 나눌 수 있었다. 미성숙 혈구는 직경 약 4.5-5.5 μm로서 세포질 내에 지름 2-3 μm인 원형의 핵이 대부분을 차지하고 있었다. 과립세포는 직경 약 7 μm로서 세포질에는 전자밀도가 낮고 크기가 다양한 과립들과 미토콘드리아, 글리코겐 과립들이 관찰되었다. 초자세포는 핵의 크기 및 세포질에 존재하는 미세소관의 특징에 따라 6종류로 세분화 할 수 있었다. 초자세포 III은 라이소좀 및 글리코겐 과립의 형성이 미약하여식세포의 기능 보다는 영양분의 흡수 및 운반 기능을 수행하는것으로 판단되며, 특히 초자세포 VI은 형태가 불규칙한 아메바형으로 세포질에 글리코겐 과립의 분포가 높고, 이식포와 자식포가 분포하고 있었다. 따라서 이들 혈구들의 정확한 기능을파악하기 위해서는 효소학적 및 면역학적 연구를 추가적으로수행해야 할 것으로 판단된다.

        Recovery of Pseudomonas anguilliseptica from Diseased Striped Beakperch (Oplegnathus fasciatus) in Korea

        In recent years, several southern coastal fish farms in Korea have experienced 2-30% mortality in striped beakperch suffering from bacterial infections during the spring. In this study, we identified a bacterium isolated from diseased striped beakperch as Pseudomonas anguilliseptica via a biochemical test and 16S rDNA sequence analysis. To evaluate the susceptibility of striped beakperch to P. anguilliseptica, 4.39×107 or 4.39×105 CFU/fish of bacteria were injected intraperitoneally at 18±1°C into fish weighing 5.5 g. Cumulative mortalities reached 100% and 45% in the 4.39×107 and 4.39×105 CFU/fish infected groups, respectively. Experimentally infected fish showed cell-associated inflammation as well as bacteria in the kidneys and spleen. This study is the first report of striped beakperch mortality caused by P. anguilliseptica, which has pathogenicity in striped beakperch.

        국내 양식 흰다리새우에서 검출된 노랑머리바이러스(YHV-8)의 유전학적 특성

        This study was performed for screening yellow head virus(YHV) complex in 252 including 235 white leg shrimps (Litopenaeus vannamei) and 17 oriental shrimp (Fenneropenaeus chinensis) collected from 18 farms located in southwestern province of Korea. The virus complex was detected by nested reverse-transcriptase polymerase chain reaction (RT-PCR) assay. In the assay, amplicons were resulted in RNAs exracted from 38 shrimps (21 white leg shrimps and 17 oriental shrimps) obtained from 7 farms. In phylogenetic analysis using sequences of ORF1b gene, all 38 sequences obtained in this study formed an independent lineage with YHV-8 genotype firstly isolated in China, belonged to an YHV-8 clade.

      • Red Seabream Iridovirus (RSIV)의 가두리양식장내 감염동태

        Since 1998, Red seabream Iridovirus (RSIV) has been detected every year and has caused economic damages in net-cage cultured red seabream, Pagrus major and striped beakperch (Japanese parrotfish), Oplegnathus fasciatus (Temminck & Schlegel), in southern and western coastal areas of the Korean peninsula. This study was conducted to know epidemiological characteristics of the RSIV infection in net-cage farm. For the purpose, 116 red seabream and 89 striped beakperch were collected three fish farms in Namhae area during May to October in 2002. First diagnosis included field observations, gross and internal observations of fishes, Giemsa stain of the spleen and kidney stamp to identify enlarged cells which is a characteristic of the RSIV infection. In addition, virus culture by cell lines, histopathological studies and polymerase chain reaction (PCR) were conducted. In spring, RSIV was detected from the fish body but not in sea water. Although RSIV was exist in fish body no mortality was recorded. The mortality recorded from the August, and increased in September and October according with increasing of water temperature. During August and October, RSIV was detected not only dead fish but also sea water. Striped beakperch was more susceptible than red seabream against RSIV. RSIV could be transmitted via sea water and caused mortality in October to red seabream which is less susceptible to RSIV.

      • 연안해수중의 흰반점바이러스 동태에 관한 연구

        In this study, we were concentrating virus form natural seawater of near shrimp farms collected in the west and south coast and were detecting white spot syndrome virus (WSSV) from cultured shrimp using polymerase chain reaction (PCR). From Aug. 1999 to Aug. 2001, both of the cultured shrimp and concentrated seawater samples collected from Sinan, Kochang, Kangjin, and Sunchun could detect gene of WSSV. In Aug. 1999 samples, we could also detected WSSV in common brackish goby samples collected from shrimp farm of Kochang. All of the seawater samples and cultured shrimp collected from the west-south coast in 1999-2000 tested WSSV-positive, suggesting that WSSV is widely distributed in seawater of the west-south coast all time of the year.
