Meiotic metaphase chromosomes of the testis of two storage insects, tobacco moth (Ephestia elutella H.) and cigarette beetle (Lasioderma serricorne F.) were examined to study their karyotypes. The number of haploid chromosomes of the tobacco moth was 31 and the karyotype was characterized by 29 metacentric and 2 submetacentric chromosomes. The metaphase chromosome length of tobacco moth ranged ca. 1.44-2.11${\mu}{\textrm}{m}$ and the average length was 1, 78${\mu}{\textrm}{m}$. The number of haploid chromosomes of the cigarette beetle at metaphase was 10 and the karyotype was characterized by 8 metacentric and 2 submetacentric chromosomes. The range of metaphase chromosome length of the cigarette beetle was ca. 1.79- 2.39${\mu}{\textrm}{m}$ and the average length 2.09${\mu}{\textrm}{m}$.
Additional Biological Characteristics of TMV Strains Isolated from Tobacco, Tomato and Pepper Plants

Some experiments were carried out to formulate artificial diets for mass-rearing of the Oriental tobacco budworm, Heliothis assulta G.. Larvae were reared on some different diets and the results obtained are summarized as follows. 1. Best growth of larvae was shown on corn-soybean diet complemented with milk, yeast, vitamines and anticontamination agents. 2. Most larvae on artificial diets molted 5 times, whereas the larvae on tobacco leaves were almost equal between the number of 5 or 6 molts. 3. There was no significant difference in developmental period of each instar among different diets in case of 5-molted larvae. But it was longer for the larvae on artificial diets than those on tobacco leaves then molted 6 times. 4. Each larval instar had wider head width, when reared on artificial diets than that on tobacco leaves, But the head width of last instar larvae was almost same, regardless of numbers of molting. 5. The mortality was low in the larvae on the artificial diets at the early instars, but higher at the prepual stage, when compared to that on the tobacco leaves. 6. Heavier pupae were obtained on artificial diets. 7. Higher pupal mortality was shown and longer days were required for emergence on artificial diets.

Streptomyces melanosporofaciens로 동정된 strain 88-GT-161 균주는, 그람양성세균 및 식물병원성 곰팡이에 각각 항균활성을 나타내는 phthalate 유도체 및 염기성 마크로라이드의 새로운 항생물질을 생산하는 것으로 밝혀졌다. 이들 활성물질들의 분리·정제과정에서 in vitro 및 in vivo(포트 시험) 항균활성을 조사하였으며, phthalate 유도체 항생물질은 IR, NMR, 질량분석 스펙트럼 조사를 통하여 bis (2-ethylhexyl) phthalate(dioctyl phthalate)로 동정하였다. Dioctyl phthalate가 Streptomyces sp.에 의하여 생산되며 생합성된 이 화합물이 항균활성을 가진다는 사실이 보고된 것은 이것이 처음이다. A phthalic acid derivative and basic macrolide antibiotics, with antimicrobial activity against Gram positive bacteria and phytopathogenic fungi, respectively, were found to be produced by a strain 88-GT-161 identified as being a variety of Streptomyces melanosporofaciens. This paper describes an isolation procedure of the active compounds produced by this strain, their in vitro and in vivo(pot test) antimicrobial activites, and structure determination of one of the compounds, bis (2-ethylhexyl) phthalate, a phthalic acid derivative antibiotic. This compounds, upon comparision with authentic bis (2-ethylhexyl) phthalate, dioctyl phthalate, revealed a difference in antimicrobial activity even though physico-chemical properties of these two compounds seemed indentical. This is the first report that dioctyl phthalate is biosynthetically produced by a Streptomyces sp. and shows antimicrobial activity.

새로운 phthalic acid 유도체 및 염기성 마크로라이드 항생물질을 생산하는 strain 88-GT-161은 numerical taxonomic data base에 의거 Strcptomyces melanosporofaciens로 동정되었으나, 139단위 특성 중 4가지 단위에 있어 S. violacconiger 혹은 S. violaccusniger로 대표되는 cluster No.32에 속하는 6균주의 공통된 특성과 분명히 다른 특성을 나타냈다. 따라서 본 논문에서는 strain 88-GT-161의 분류·동정학적 특성에 대하여 보고하고자 한다. Strain 88-GT-161 producing new phthalic acid derivative and basic macrolide antibiotics was identified as being S. melanosporofaciens based on numerical taxonomic data. However, 4 unit characters among 139 units were clearly different from the common properties of 6 strains belonging to cluster No. 32 represented by the name of S. violaceoniger or S. violaceusniger, leading us to designate as a variety of S. melanosporofaciens. This paper describes the taxonomic characteristics of the strain. Isolation and chemical structures, including biological activities of the active compounds produced by this strain will be presented elsewhere.
비전이성 외피단백질 cDNA 도입에 의한 감자 바이러스 Y 저항성 연초 품종 개발

A cDNA fragment, named PVY-CP, encoding coat protein (CP) of potato virus Y (PVY-VN) was obtained by RT-PCR. The PVY-CP cDNA was manipulated to construct an untranslatable form in which three stop codons are introduced near the start codon by site-directed mutagenesis. The untranslatable PVY-CP cDNA was inserted into plant expression vector and transferred to Nicotiana tabacum cv. NC 82 by Agrobacterium-mediated transformation method. The untranslatable PVY-CP cDNA transformed plants were regenerated and PVY resistant transformants were selected based on symptom development after mechanical inoculation. Genomic PCR and Southern blot analysis confirmed that one or more copies of the untranslatable PVY-CP cDNA was integrated into the genome of tobacco plants which are highly resistant to PVY.
