효소처리 및 가압추출 공정을 이용한 율피 추출물의항산화, 피부 미백 및 주름개선 효과

In this study, the anti-oxidant, whitening, and anti-wrinkle effects of Castanea crenata inner shell extracts processed by enzyme treatment and pressurized extraction were investigated. The Castanea crenata inner shell was first hydrolyzed using celluclast, viscozyme, or hemicellulase. Then, it was subjected to pressure extraction for different durations (30, 60, and 120 min). The yields of the Castanea crenata inner shell extracts processed by different enzyme treatments followed by pressurized extraction for different times are in the range of 12.42-29.80%. The total polyphenol, flavonoid, and tannin contents of the C30m (celluclast enzyme and autoclave extracts at 30 min) extract were 15.48, 10.82, and 15.82 g/100 g, respectively. The total sugar content of the H120m(hemicellulase enzyme and autoclave extracts at 120 min) extract is 61.07 g/100 g. The 1,1-diphenyl-2-pycrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities of the C30m extract at 1,000 µg/mL are 89.20, and 81.96%, respectively. The superoxide radical scavenging and ferric-reducing antioxidant power of the C30m extract at 1,000 µg/mL are 67.63% and 1,324.79 μM, respectively. Further, the tyrosinase and elastase inhibition activity of the C30m extract at 1,000 μg/mL are 61.32, and 61.06%, respectively. Our results indicate that the Castanea crenata inner shell extracts processed by enzyme treatment followed by pressurized extraction could have beneficial effects on facial skin and they should be considered for use in new functional cosmetics.
약용열매 혼합 열수 추출물의 이화학적 특성 및 항산화 활성

This study was conducted to examine the physicochemical properties and antioxidant activities of medicinal fruit extracts (Corni fructus, Schizandra chinensis, Rubus coreanus Miquel and Lycii folium) with different extraction mixing ratios (MS, an equal ratio of the medicinal fruit = 1.25:1.25:1.25:1.25; M1, 2:1:1:1; M2, 1:2:1:1; M3, 1:1:2:1 and M4, 1:1:1:2) from medicinal fruit. pH, sugar content and acidity of the extracts were 3.22~3.52, 3.20~4.20 °Brix and 3.60~5.85%, respectively. The extraction yield of M2 (42.33%) was higher than those of MS (36.03%), M1 (40.40%), M3 (32.53%) and M4 (35.90%). The total polyphenol and flavonoid contents of M3 were 14.54 g/100 g and 5.65 g/100 g, respectively. The DPPH and ABTS radical scavenging activities of M3 at 1,000 µg/mL were 86.09% and 90.49%, respectively. The ferric-reducing antioxidant power and the reducing power of M3 at 250~1,000 µg/mL were 0.36~0.86 µM and 0.21~0.96, respectively. The antioxidant activities of M3 were significantly higher than those of the other extracts. In conclusion, this study demonstrated that medicinal fruit extracts had potential as a functional material.
해방풍 부위별 용매추출물의 항산화 활성 및 nitric oxide 생성 억제

In this study, we compared the antioxidant and immuno-modulatory effects of water (HR, HL, HS), 70% ethanol (ER, EL, ES), and 70% methanol (MR, ML, MS) extracts of different parts (root, leaf, and seed) from Glehnia Radix. Yields were 17.40-30.17% for water extract, 11.47-28.67% for 70% ethanol extract, and 10.73-30.57% for 70% methanol extract, respectively. The total polyphenol and flavonoid contents of EL were 10.79 g/100 g and 2.01 g/100 g, respectively. The DPPH and ABTS radical scavenging activities of EL at 1,000 µg/mL were 84.70% and 57.64%, respectively. The superoxide radical scavenging and ferric-reducing antioxidant power of EL at 1,000 µg/mL were 84.05% and 975.28 μM, respectively. Moreover, 70% ethanol and 70% methanol extracts of root from Glehnia Radix significantly inhibited production of NO in LPS-stimulated macrophage RAW 264.7 cells without cytotoxicity. These results suggest that 70% ethanol and 70% methanol extracts of Glehnia Radix leaf may be a useful functional food material in the food industry.
추출방법에 따른 감태에서 분리한 조다당류의 이화학적 품질특성 및 항산화 활성

This study was conducted to examine the effect of extraction methods on physicochemical characteristics and antioxidant activities of crude polysaccharides isolated from Ecklonia cava. The yield of extract ranged from 6.67 to 20.47%. The total polyphenol and total flavonoid contents of viscozyme extract were 16.56 g/100 g, 13.25 g/100 g, respectively. The total sugar content of viscozyme extract was 74.16 g/100 g, and the total protein content of neutrase extract was 20.95 g/100 g. The DPPH and ABTS radical scavenging activities of viscozyme extract at 1,000 μg/mL were 46.03% and 43.16%. The ferric-reducing antioxidant power of viscozyme extract at 1,000 μg/mL was 1.21 mM. The functionalities of extracts were investigated with L-132 cell lines. The crude polysaccharides not showed the toxicity on L-132 cell line in 100~1,000 μg/mL. The viscozyme extract (1,000 μg/mL) performed better than other extracts protective effects against oxidative stess in L-132 cells (86.47%) which was more potent than other extracts. There results suggest that the viscozyme extract may be a useful functional food material in the food industry.
맥문동, 오미자 및 인삼 혼합추출물의 이화학적 특성 및 생리활성

생맥산 원료인 맥문동, 오미자 및 인삼의 최적 혼합비율을 설정하고자 맥문동, 오미자 및 인삼 혼합추출물을 제조하여 이화학적 특성 및 생리활성을 조사하였다. 맥문동, 오미자 및 인삼 혼합추출물의 추출수율은 25.33-33.87%이었으며, 총 폴리페놀 및 총 플라보노이드 함량은 오미자의 혼합비율이 높은 추출물(ME1)에서 각각 1.01 g/100 g 및 0.07 g/100 g으로 가장 높은 값을 나타내었고, 총 당 함량은 맥문동의 혼합비율이 높은 추출물(MEC)에서 22.83 g/100 g으로 가장 높은 값을 나타내었다. DPPH radical 및 ABTS radical 소거활성은 ME1 추출물에서 각각 26.79% 및 21.09%로 가장 우수한 radical 소거활성을 나타내었으며, superoxide radical 소거활성 및 FRAP 활성 또한 ME1 추출물에서 각각 67.83% 및 $295.47{\mu}M$로 가장 우수한 항산화 활성을 나타내었다. 맥문동, 오미자 및 인삼 혼합추출물의 생리활성을 확인한 결과, $H_2O_2$에 의해 산화적 손상된 폐상 피세포인 L-132 세포의 생존율은 51.43%를 나타낸 반면 ME1 추출물($1,000{\mu}g/mL$)은 81.22%로 가장 높은 세포보호 효과를 나타내었다. LPS로 자극을 유도한 대식세포주인 RAW264.7 세포는 인삼의 혼합비율이 높은 ME2 추출물($1,000{\mu}g/mL$)은 $7.48{\mu}M$로 가장 적은 nitric oxide 생성량을 나타내어 nitric oxide 생성을 억제하였다. 따라서 본 연구에서는 동의보감 및 일반적으로 많이 이용하고 있는 MEC 추출물보다 ME1 및 ME2 추출물에서 항산화 활성 및 면역조절능이 우수함을 확인하여, 새로운 생맥산 혼합비율을 이용한 항산화 활성 및 면역조절용 기능성 식품 소재 개발이 가능할 것으로 사료된다. Meyer) with different extraction mixing ratios (MEC, 2:1:1; ME1, 1:2:1; ME2, 1:1:2; ME3, 1.34:1.33:1.33). The yield of extracts ranged from 25.33 to 33.87%. The total polyphenol and total flavonoid contents of ME1 extracts were 1.01 g/100 g, 0.07 g/100 g, respectively. The total sugar contents of MEC extract was 22.83 g/100 g, respectively. The DPPH and ABTS radical scavenging activities of ME1 extracts at $1,000{\mu}g/mL$ were 26.79% and 21.08%. The superoxide radical scavenging and ferric-reducing antioxidant power of ME1 extracts at $1,000{\mu}g/mL$ were 67.83% and $295.47{\mu}M$, respectively. The functionalities of extracts were investigated with L-132 and RAW264.7 cell lines. The extracts on different mixing ratios did not show the toxicity on L-132 and RAW264.7 cell line in $100-2,500{\mu}g/mL$. The ME1 extract of $1,000{\mu}g/mL$ performed better than other extracts protective effects against oxidative stess in L-132 cells (81.22%) and the ME2 extract at $1,000{\mu}g/mL$ decreased nitric oxide production by $7.48{\mu}M$ which was more potent than other extracts. There results suggest that the ME1 extracts may be a useful functional food material in the food industry.
